Hedgehog (Hh) signalling is a potent regulator of cell fate and function. While much is known about the events within a Hh-stimulated cell, far less is known about the regulation of Hh-ligand production. Drosophila Hyperplastic Discs (Hyd), a ubiquitin-protein ligase, represents one of the few non-transcription factors that independently regulates both hh mRNA expression and pathway activity. Using a murine embryonic stem cell system, we revealed that shRNAi of the mammalian homologue of hyd…

Ubiquitin-protein ligase Ubr5 cooperates with hedgehog signalling to promote skeletal tissue homeostasis
TLDR
This work shows that UBR5 interacts with the HH pathway to regulate skeletal homeostasis in and around joints of the legs and identifies targets that may be harnessed for biomedical engineering and clinical applications.
The N-recognin UBR4 of the N-end rule pathway is required for neurogenesis and homeostasis of cell surface proteins
TLDR
The results suggest that the N-recognin UBR4 plays a role in the homeostasis of cell surface proteins and, thus, cell adhesion and integrity.
UBR5 Is Coamplified with MYC in Breast Tumors and Encodes an Ubiquitin Ligase That Limits MYC-Dependent Apoptosis
TLDR
It is shown how ubiquitin ligase UBR5 functions as a molecular rheostat to prevent excess accumulation of MYC protein in cells and in protection from drug-induced apoptosis.
Targeting ubiquitin protein ligase E3 component N-recognin 5 in cancer cells induces a CD8+ T cell mediated immune response
TLDR
Experimental work in vitro and in vivo demonstrates that UBR5, functioning as an oncoprotein, plays a profound role in breast cancer growth and metastasis and has strong implications for the development of novel immunotherapy to treat highly aggressive breast cancers that resist conventional treatment.
The E3 ubiquitin ligase UBR5 regulates centriolar satellite stability and primary cilia formation via ubiquitylation of CSPP-L
TLDR
A key role is established for UBR5 in ciliogenesis that may have important implications in understanding cancer pathophysiology and cytoplasmic organization of CSPP1-comprising centriolar satellites in centrosomal periphery.
Ubiquitin Ligases Involved in the Regulation of Wnt, TGF-β, and Notch Signaling Pathways and Their Roles in Mouse Development and Homeostasis
TLDR
The current understanding of how the UPS regulates the Wnt, TGF-β, and Notch pathways is reviewed and the knowledge gained from related mouse models is summarized.
The ubiquitin ligase UBR5 suppresses proteostasis collapse in pluripotent stem cells from Huntington’s disease patients
TLDR
It is shown that proteasome activity determines HTT levels, preventing polyQ-expanded aggregation in iPSCs from HD patients (HD-iPSCs), and that UBR5 is a modulator of super-vigilant proteostasis of iPSCS.
The E3 ubiquitin ligase UBR5 regulates centriolar satellite stability and primary cilia
TLDR
A key role is established for UBR5 in ciliogenesis that may have important implications in understanding cancer pathophysiology, and it is shown that U BR5 ubiquitylates CSPP1, and that UBR 5 is required for cytoplasmic organization of CSPp1-comprising centriolar satellites in centrosomal periphery.

Regulation of hedgehog Ligand Expression by the N-End Rule Ubiquitin-Protein Ligase Hyperplastic Discs and the Drosophila GSK3β Homologue, Shaggy
TLDR
Drosophila-based findings highlight an apical role for Hyd and Sgg in initiating Hedgehog signalling, which could also be evolutionarily conserved in mammals.
The bHLH transcription factor dHAND controls Sonic hedgehog expression and establishment of the zone of polarizing activity during limb development.
TLDR
It is shown that the basic helix-loop-helix transcription factor dHAND is expressed posteriorly in the developing limb prior to Shh and subsequently occupies a broad domain that encompasses the Shh expression domain.
The ubiquitin ligase Hyperplastic discs negatively regulates hedgehog and decapentaplegic expression by independent mechanisms
TLDR
Photoreceptor differentiation in the Drosophila eye disc progresses from posterior to anterior in a wave driven by the Hedgehog and Decapentaplegic signals, with loss of hyperplastic discs causing the accumulation of full-length Cubitus interruptus protein, but not of Smoothened, in both the eye and wing discs.
Shaggy/GSK3 antagonizes Hedgehog signalling by regulating Cubitus interruptus
TLDR
It is proposed that Sgg/GSK3 acts in conjunction with PKA to cause hyperphosphorylation of Ci, which targets it for proteolytic processing, and that Hh opposes Ci proteolysis by promoting its dephosphorouslation.
An inversion involving the mouse Shh locus results in brachydactyly through dysregulation of Shh expression.
TLDR
The dysregulation of Shh expression causes the local upregulation of hedgehog target genes such as Gli1-3, patched, and Pthlh, as well as the downregulation of Ihh and Gdf5, which results in shortening of the digits through an arrest of chondrocyte differentiation and the disruption of joint development.
A Family of Mammalian E3 Ubiquitin Ligases That Contain the UBR Box Motif and Recognize N-Degrons
TLDR
The results establish the UBR box family as a unique class of E3 proteins that recognize N-degrons or structurally related determinants for ubiquitin-dependent proteolysis and perhaps other processes as well.
The mechanisms of Hedgehog signalling and its roles in development and disease
TLDR
Novel biological insights are revealed into the function of HH lipidation in the secretion and transport of this ligand and details of the signal transduction pathway, which involves Patched 1, Smoothened and GLI proteins, as well as, in vertebrates, primary cilia.
DYRK1B-dependent autocrine-to-paracrine shift of Hedgehog signaling by mutant RAS
TLDR
The molecular cross-talk between RAS and HH signaling is investigated and it is found that, although mutant RAS induces or enhances SHH expression and favors paracrine HH signaling, it antagonizes autocrine HH signal transduction.
