Cytochrome P450 2J2 oxidizes arachidonic acid to a series of epoxyeicosatrienoic acid (EET) isomers in human tissues. EETs regulate numerous homeostatic processes, including cytoprotective and proliferative responses against injurious stresses. There is little information currently available on the factors that regulate CYP2J2, but strategies to activate expression could use the beneficial effects of EETs in cells. The basic leucine zipper (bZIP) transcription factor c-Jun has been shown…

CYP2J2 – regulation, function and polymorphism
TLDR
Greater understanding of the mechanisms by which CYP2J2 is regulated could provide insights into important pathogenic processes in complex disease states and the development of agents that promote fatty acid epoxide production, or stable analogs that retain the activities of the epoxides, offer promising new avenues for utilizing the beneficial actions of these molecules.
Activity, Inhibition, and Induction of Cytochrome P450 2J2 in Adult Human Primary Cardiomyocytes
TLDR
This cell model can be a useful in vitro model to investigate the role of CYP2J2-mediated drug metabolism, arachidonic acid metabolism, and their association to drug induced cardiotoxicity.
Ophiopogonin D maintains Ca2+ homeostasis in rat cardiomyocytes in vitro by upregulating CYP2J3/EETs and suppressing ER stress
Aim:CYP2J3 in myocardium metabolizes arachidonic acid to 4 regioisomeric epoxyeicosatrienoic acids (EETs), which have diverse biological activities in rat heart. In this study we examined whether
Regulation of CYP2J2 and EET Levels in Cardiac Disease and Diabetes
TLDR
The mechanisms of regulation of CYP2J2 by inducers, inhibitors, and oxidative stress modeling certain disease states in various cell lines and tissues and their implication in the pathophysiology of diabetes and CVD will be discussed.
Activator Protein-1 Regulation of Murine Aldehyde Dehydrogenase 1a1
TLDR
The levels of activator protein-1 (AP-1) mRNA and protein, as well as the amount of phosphorylated c-Jun were significantly increased in mouse liver or Hepa1c1c7 cells treated with either BHA or acrolein, indicating that recruitment of c- Jun to the mouse Aldh1a1 gene promoter results in increased transcription.
15-Epoxyeicosatrienoic Acid Suppresses Cigarette Smoke Extract – Induced Apoptosis in Lung Epithelial Cells by Inhibiting Endoplasmic Reticulum Stress
TLDR
The data suggest that CYP2J2-derived EETs protect against CSE-induced lung injury possibly through attenuating ER stress.
Hyperhomocysteinemia results from and promotes hepatocellular carcinoma via CYP450 metabolism by CYP2J2 DNA methylation
TLDR
Hyperhomocysteinemia results from but can also promote hepatocarcingenesis via CYP450-EET metabolism by crosstalk of DNA demethylation of CYP2J2 and ERK1/2 signaling.
14,15-Epoxyeicosatrienoic Acid Suppresses Cigarette Smoke Extract-Induced Apoptosis in Lung Epithelial Cells by Inhibiting Endoplasmic Reticulum Stress
TLDR
The data suggest that CYP2J2-derived EETs protect against CSE-induced lung injury possibly through attenuating ER stress.
CYP2J2 Expression in Adult Ventricular Myocytes Protects Against Reactive Oxygen Species Toxicity
TLDR
This work is the first to determine the consequence of cellular stress, specifically high ROS levels, on CYP2J2 expression in human ventricular myocytes and discusses how this enzyme may play an important role in response to cardiac oxidative stress.

Role of activator protein-1 in the down-regulation of the human CYP2J2 gene in hypoxia.
TLDR
An important role is established for c-Jun in the control of CYP2J2 expression in liver cells after down-regulated at a pre-translational level in human hepatoma-derived HepG2 cells incubated in a hypoxic environment.
Characterization of a c-Jun-responsive module in the 5'-flank of the human CYP2J2 gene that regulates transactivation.
TLDR
It is demonstrated that the c-Jun-responsive module between -122 and -50 in the CYP2J2 proximal promoter contains an atypical AP-1 element at -105/-95 that has a major role in c- Jun transactivation and acts in conjunction with the -56/-63 element to regulate expression.
Butylated hydroxyanisole regulates ARE‐mediated gene expression via Nrf2 coupled with ERK and JNK signaling pathway in HepG2 cells
TLDR
Results strongly suggested that ERK and JNK signaling pathways played important and positive roles in BHA‐induced and Nrf2‐dependent regulation of ARE‐mediated gene expression, as well as the nuclear translocation of NRF2 in HepG2 cells.
Overexpression of cytochrome P450 CYP2J2 protects against hypoxia-reoxygenation injury in cultured bovine aortic endothelial cells.
TLDR
Treatment of BAECs with synthetic EETs and/or epoxide hydrolase inhibitors showed protective effects against HR injury, and the cytoprotective effects of CYP2J2 may be mediated, at least in part, by antioxidant effects.
Cytochrome P450 2C9-induced Endothelial Cell Proliferation Involves Induction of Mitogen-activated Protein (MAP) Kinase Phosphatase-1, Inhibition of the c-Jun N-terminal Kinase, and Up-regulation of Cyclin D1*
TLDR
The data demonstrate that the mitogenic effect of CYP 2C9 is due to the generation of EETs, which promote the MKP-1-mediated dephosphorylation and inactivation of JNK, effects ultimately culminating in the expression of cyclin D1 and endothelial cell proliferation.
Regulation of the rat CYP4A2 gene promoter by c-Jun and octamer binding protein-1.
Direct evidence that sulfhydryl groups of Keap1 are the sensors regulating induction of phase 2 enzymes that protect against carcinogens and oxidants
TLDR
This finding suggests that reaction of cysteine thiols is followed by rapid formation of protein disulfide linkages, which are the direct sensors of inducers of the phase 2 system.
Regulation of the antioxidant response element by protein kinase C-mediated phosphorylation of NF-E2-related factor 2.
TLDR
Findings indicate an important role of the PKC pathway in the ARE-mediated gene expression, and suggest that PKC-directed phosphorylation of Nrf2 may be a critical event for the nuclear translocation of this transcription factor in response to oxidative stress.
