STUDY QUESTION Which non-declared proteins (proteins not listed on the composition list of the product data sheet) are present in unconditioned commercial embryo culture media? SUMMARY ANSWER A total of 110 non-declared proteins were identified in unconditioned media and between 6 and 8 of these were quantifiable and therefore represent the majority of the total protein in the media samples. WHAT IS KNOWN ALREADY There are no data in the literature on what non-declared proteins are present…

Comprehensive analysis of soluble RNAs in human embryo culture media and blastocoel fluid
TLDR
The use of miRNAs as a reliable biomarker is questioned and the need for a critical methodological approach in miRNA studies is highlighted, as tiRNA fragments appear to be overexpressed in conditioned IVF media samples and could potentially be a novel biomarker worthy of investigation.
The composition of human preimplantation embryo culture media and their stability during storage and culture.
TLDR
The wide variation between culture media shows that the optimal composition is still unknown, and companies are urged to fully disclose the composition of their culture media.
Characterization and quantification of proteins secreted by single human embryos prior to implantation
TLDR
This study illustrates the potential of high‐sensitivity proteomics to measure clinically relevant biomarkers in minute samples and suggests that key aspects of embryo competence could be measured using a proteomic‐based strategy, with negligible risk of harm to the living embryo.
Bisphenol A in culture media and plastic consumables used for ART.
TLDR
Although it is demonstrated that some plastic materials used in ART contain BPA, under routine conditions none appear capable of leaching BPA at levels higher than those from maternal internal exposure.
GDF9 concentration in embryo culture medium is linked to human embryo quality and viability
TLDR
The GDF9 concentration in culture medium is linked to embryo quality and viability, and exhibited the potential to be a non-invasive biomarker for embryo selection.
Non‐Invasive Assessment of the Embryo Viability via the Analysis of the Culture Media
TLDR
This chapter sum‐ marizes the currently available metabolomic and proteomic approaches as the non‐inva‐ sive molecular assessment of embryo viability.
Ammonium accumulation in commercially available embryo culture media and protein supplements during storage at 2-8°C and during incubation at 37°C.
STUDY QUESTION Does ammonium accumulate in commercially available culture media and protein supplements used for in vitro development of human pre-implantation embryos during storage and incubation?
Differences in secretome in culture media when comparing blastocysts and arrested embryos using multiplex proximity assay
TLDR
The secretome and timing to development differ in embryos forming blastocyst and those that become arrested, and in high- versus low-quality blastocysts, and the levels of certain proteins also correlate to specific times to development.

Comparative protein-profile analysis of implanted versus non-implanted human blastocysts.
TLDR
The differences identified in the protein profile of the culture media in the presence of implanted versus non-implanted blastocysts can be considered a new non-invasive approach in the search for new tools to diagnose blastocyst viability.
The protein source in embryo culture media influences birthweight: a comparative study between G1 v5 and G1-PLUS v5.
TLDR
A significant difference was observed in gestational age- and gender-adjusted birthweight (Z scores) and the proportion of large-for-gestational age (LGA) babies between embryos cultured in G1 v5 and those cultured in LGA v5 media.
Secretome of the preimplantation human embryo by bottom-up label-free proteomics
TLDR
This approach seems to allow quantification and identification of unique proteins related to positive- and negative-implantation groups, which can be further validated as biomarkers for selection and transfer of a single embryo.
Non-invasive metabolomic profiling of Day 2 and 5 embryo culture medium: a prospective randomized trial.
TLDR
This study shows that adding NIR spectroscopy to embryo morphology does not improve the chance of a viable pregnancy when performing SET, and the NIR technology appears to need further development before it can be used as an objective marker of embryo viability.
Analysis of protein expression (secretome) by human and mouse preimplantation embryos.
Characterization of a diverse secretome generated by the mouse preimplantation embryo in vitro
TLDR
This study shows the feasibility of ESI mass spectrometry for identifying the proteins secreted by the preimplantation embryo in vitro and identifies a range of targets that now require detailed functional analysis to assess whether their release by the embryo is an important property of early embryo development.
Day 3 embryo selection by metabolomic profiling of culture medium with near-infrared spectroscopy as an adjunct to morphology: a randomized controlled trial.
TLDR
The live birth rate after embryo selection by NIR spectroscopy and morphology is not significantly different compared with the liveBirth rate after embryos were selected by morphology alone, and is probably selected for a better prognosis patient group.
Glucose consumption of single post-compaction human embryos is predictive of embryo sex and live birth outcome.
TLDR
The rapid screening of glucose metabolism by the human embryo on Day 4 and 5 may prove to be a useful metric in the development of algorithms for the selection of embryos for transfer in human IVF.
