Summary. Candida albicans was maintained in various culture media and incubated with different concentrations of the antifungal agent rilopirox. After fixation, dehydration and embedding in Spurr's medium, the cells were analysed at the ultrastructural level to investigate morphological aspects of the antifungal mode of action of this new hydroxypyridone compound. All untreated or sham‐treated control cells exhibited a normal ultrastructural appearance. The cells were surrounded by a…

TLDR
The morphological changes induced by the drug at two concentrations, the MIC and 10 times the MIC, during 24 and 48 h on filamentous opportunistic fungi indicated that the intensity of the ultrastructural breakdown depended on the dose and not on the time of exposure.
Ciclopirox Olamine Treatment Affects the Expression Pattern of Candida albicans Genes Encoding Virulence Factors, Iron Metabolism Proteins, and Drug Resistance Factors
TLDR
The view that the antifungal activity of ciclopirox olamine may at least be partially caused by iron limitation is supported, as a dramatic increase in sensitivity to oxidative stress was noted, which may indicate the reduced activities of iron-containing gene products responsible for detoxification.
In vitro investigations on the mode of action of the hydroxypyridone antimycotics rilopirox and piroctone on Candida albicans
TLDR
There are clear hints that reactive oxygen species (ROS) and available metabolic activity are important parts of the mode of action of the hydroxypyridone antimycotics rilopirox and piroctone.
In vitro activity of rilopirox against fluconazole‐susceptible and fluconazole‐resistant Candida isolates from patients with HIV infection
TLDR
Rilopirox was found to be able to inhibit growth of all clinical yeast isolates and may be used as an alternative not only in the treatment of vaginal candidosis, but also in oropharyngeal Candida infections, e.g. in AIDS patients.
Effects of Pycnoporellus fulgens (Fr.) Donk crude extract on Candida glabrata ultrastructure
TLDR
Transmission electron microscopy revealed that the extract acted on the cell envelope (cell wall and plasmalemma) and cell divisions were affected by thickening of the septum and a deficiency in the daughter cell wall texture.
The hydroxypyridones: a class of antimycotics of its own
TLDR
Interest mainly focuses on ciclopirox lacquer for onychomycosis and the relative role of hydroxypyridones will have to be finally established in comparative trials.
Candida albicans mitochondrial dynamics during a yeast to hyphal transition
TLDR
Comparison studies using cellular homogenates and crude mt fractions indicated that individual mt undergo changes in enzyme activity prior to mt division and growth, implying that mt processes are important in the yeast to hyphal transition.
MHP1, a Magnaporthe grisea hydrophobin gene, is required for fungal development and plant colonization
TLDR
Results indicate that MHP1 has essential roles in surface hydrophobicity and infection‐related fungal development, and is required for pathogenicity of M. grisea.
Six Novel Antimycotics
TLDR
Six new antimycotic agents which have potential applications for human cutaneous and mucosal diseases are reviewed, including rilopirox, lanoconazole, NND-502, butenafine, eberconazole and voriconazole.
Wirkungsweise von Ciclopiroxolamin auf Candida albicans
TLDR

Effects of in Vitro Treatment with Econazole on the Ultrastructure of Candida albicans
TLDR
Following the addition of 10 μg/ml of Econazole‐nitrate (2.2 × 10‐5 M), Candida albicans cells were incubated in vitro at 28d̀ C for 30 hours, resulting in clot‐like formations in ultrathin sections.

An electron microscopic comparison was made of the cytological responses of Candida albicans to benzalkonium chloride, amphotericin B, and filipin, all of which are thought to exert their primary pharmacological actions on the cytoplasmic membrane of susceptible microorganisms.

An Ultrastructural and Cytochemical Study of Candida aIbicans after in Vitro Treatment with Imidazoles
TLDR
The ultrastructural dianges in Candida albicans after exposure in vitro to different doses of clotrimazole and miconazole have been investigated and differed markedly between the two drugs with respect to the number of cells involved, the intensity of the lesions and their subcellular location.
[Studies on the mechanism of antifungal action of ciclopiroxolamine/Inhibition of transmembrane transport of amino acid, K+ and phosphate in Candida albicans cells (author's transl)].
TLDR
It is presumed from these results that Cic-mediated growth inhibition or death of fungal cells is primarily caused by intracellular depletion of some essential substrates and/or ions, and that such effects are brought about through blockage of their uptake from the medium.
Cytochemical and Biochemical Studies of Yeasts After In Vitro Exposure to Miconazole
TLDR
Yeast cells exposed to different doses of the antimycotic agent miconazole revealed important cytochemical changes in the topographic distribution of the phosphatases, and a hypothesis is advanced concerning the possible involvement of peroxidase and catalase in the mechanism of action of this drug.
Enzyme cytochemistry of Candida albicans.
TLDR
The problem of inadequate penetration of fixatives into yeast cells has been solved by sectioning solidified pellets of the cells in the presence of glutaraldehyde, a procedure that yields a fairly well preserved ultrastructure and sufficient enzyme activities.
Studies for the elucidation of the mode of action of the antimycotic hydroxypyridone compound, rilopirox.
TLDR
Studies on yeast mitochondria and submitochondrial particles show that rilopirox inhibits the respiratory chain and Complex I (NADH-ubiquinone oxidoreductase) contains iron-sulfur proteins and is the main system which is inhibited.
Rilopirox ‐ a New Hydroxypyridone Antifungal with Fungicidal Properties
TLDR
Rilopirox appears to meet the demand for an mtravaginal antifungal as a result of its very low water solubility of 50 ng/ ml and its immediate fungicidal action even under non‐proliferative conditions.
