This study investigated whether treating fetal fibroblast cells (donor cells) with epigenetic modification-inducing drugs could improve the development of porcine cloned embryos. Donor cells were treated with different DNA methylation inhibitors (5-aza-dC, zebularine or RG108; 5nM) or histone deacetylase inhibitors (TSA, NaBu or SCR; 50nM) for 1h, and then subjected to SCNT. All of the treated groups showed significantly higher blastocyst formation rates compared to the control group. We chose… Expand
Optimizing 5-aza-2'-deoxycytidine treatment to enhance the development of porcine cloned embryos by inhibiting apoptosis and improving DNA methylation reprogramming.
TLDR
The best 5-aza-dC treatment enhanced the development of cloned embryos by inhibiting apoptosis and improving DNA methylation reprogramming. Expand
Effect of histone deacetylase inhibitor romidepsin on the in vitro growth of foetal fibroblast cells and early development of porcine-cloned embryos
TLDR
Taken together, histone deacetylase inhibitor FK228 with optimal dose and exposure duration can enhance early developmental efficiency of porcine SCNT embryos and blastocyst quality. Expand
Optimizing treatment of DNA methyltransferases inhibitor RG108 on porcine fibroblasts for somatic cell nuclear transfer.
TLDR
The results showed that RG108 treatment resulted in time-dependent decrease of genome-wide DNA methylation on fetal fibroblasts, which only happened after 72h treatment in the authors' experiments, and no interaction effect between treatment time and concentration. Expand
Scriptaid improves the reprogramming of donor cells and enhances canine-porcine interspecies embryo development.
TLDR
It is demonstrated that scriptaid affected the reprogramming of canine donor and cloned embryos, as well as early embryo development in canine-porcine iSCNT, by regulating reprograming and apoptotic genes. Expand
Enhancement of epigenetic reprogramming status of porcine cloned embryos with zebularine, a DNA methyltransferase inhibitor
TLDR
It is found that Zb could modify DNA methylation levels in the early stages of porcine SCNT embryos and promote their developmental competence. Expand
Dynamic Methylation Changes of DNA and H3K4 by RG108 Improve Epigenetic Reprogramming of Somatic Cell Nuclear Transfer Embryos in Pigs
TLDR
Dynamic changes in DNA methylation caused by RG108 result in dynamic alterations in the patterns of H3K4me3, H3 k9Ac and histone H3 lysine 9 trimethylation, which leads to the activation of embryonic genome and epigenetic modification enzymes associated with H3k4 methylation, and contributes to reconstructing normal epigenetic modifications and improving the developmental efficiency of porcine SCNT embryos. 5-Azacytidine improves the meiotic maturation and subsequent in vitro development of pig oocytes.
TLDR
Data from the present study indicate that treatment of oocytes with 5-Aza-C may be an important approach to enhance the meiotic maturation and subsequent in vitro development of pig embryos. Expand
Effects of Trichostatin A on In Vitro Development and DNA Methylation Level of the Satellite I Region of Swamp Buffalo (Bubalus bubalis) Cloned Embryos
TLDR
Investigation of the effects of TSA on developmental potential of swamp buffalo cloned embryos showed that there was no significant difference in the rates of fusion, cleavage, and development to the 8-cell stage among treatment groups, and the DNA methylation levels in cloning embryos were approximately similar to those of donor cells. Expand
Trichostatin A-Mediated Epigenetic Transformation of Adult Bone Marrow-Derived Mesenchymal Stem Cells Biases the In Vitro Developmental Capability, Quality, and Pluripotency Extent of Porcine Cloned Embryos
TLDR
The improvements in morula/blastocyst yields and quality of cloned pig embryos seem to arise from enhanced abilities for promotion of correct epigenetic reprogramming of TSA-exposed ABM-MSC nuclei in a cytoplasm of reconstructed oocytes. Expand
Zebularine significantly improves the preimplantation development of ovine somatic cell nuclear transfer embryos.
TLDR
The results indicate that, in an ovine model, zebularine decreases overall DNA methylation levels in donor cumulus cells and reconstructed embryos, downregulates theDNA methylation profile in the promoter region of pluripotency genes in donor cells and ultimately elevates the expression of plurine genes in the reconstruct embryos, which can lead to improved development of SCNT embryos.
