Abstract: Barbituric acid (2,4,6‐pyrimidinetrione) can be transformed by a non‐enzymatic hydroxylation into alloxan (2,4,5,6‐pyrimidinetetrone). This transformation can be used as a reaction indicating the formation of hydroxyl radicals (·OH). This conversion was detected using HPLC. Formation of ·OH was demonstrated by electron spin resonance (ESR) spectroscopy combined with spin‐trapping techniques. It was shown that ·OH generated via the Fenton reaction abstracts first a hydrogen atom from… Expand
DCFH2 interactions with hydroxyl radicals and other oxidants – Influence of organic solvents
TLDR
The results of this study indicate that the potential of an oxidant does not always correlate with its efficiency to generate DCF-fluorescence. Expand
Melatonin: detoxification of oxygen and nitrogen-based toxic reactants.
TLDR
It is found that melatonin scavenges several oxygen-derived reactive agents including the hydroxyl radical (OH), hydrogen peroxide (H2O2), singlet oxygen (1O2) and hypochlorous acid (HOCl), and it is not the only means by which melatonin serves to protect molecules from oxygen and nitrogen-based reactive metabolites. Expand
Photo‐degradation of melatonin: influence of argon, hydrogen peroxide, and ethanol
TLDR
It is shown that N1‐acetyl‐N2‐formyl‐5‐methoxykynuramine (AFMK) appears to be the main, but obviously not the only product of photo‐degradation of melatonin, independently of whether the system contains oxygen or not. Expand
Effects of diphenyl diselenide on oxidative stress induced by sepsis in rats.
TLDR
It is shown that diphenyl diselenide (PhSe)(2) partially prevents the oxidative stress induced by sepsis, indicating the potential of this compound as a treatment for this pathology. Expand
Flavoprotein-Mediated Tellurite Reduction: Structural Basis and Applications to the Synthesis of Tellurium-Containing Nanostructures
TLDR
Bioinformatic data resting on some of the abovementioned enzymes enabled the identification of common structures involved in tellurite reduction including vicinal catalytic cysteine residues and the FAD/NAD(P)+-binding domain, which is characteristic of some flavoproteins. Expand
Melatonin, xanthurenic acid, resveratrol, EGCG, vitamin C and α‐lipoic acid differentially reduce oxidative DNA damage induced by Fenton reagents: a study of their individual and synergistic actions
TLDR
Melatonin was the most effective of these antioxidants tested, and reversed the pro‐oxidant effect of resveratrol and vitamin C, and exhibited synergism in combination with vitamin C and with LA (5 μM). Expand
Melatonin as an antioxidant: biochemical mechanisms and pathophysiological implications in humans.
TLDR
Melatonin's functions as an antioxidant include: a), direct free radical scavenging, b), stimulation of antioxidative enzymes, c), increasing the efficiency of mitochondrial oxidative phosphorylation and reducing electron leakage (thereby lowering free radical generation), and 3), augmenting the efficiencyof other antioxidants. Expand
Melatonin: an ancient molecule that makes oxygen metabolically tolerable
TLDR
It is hypothesized that the initial and primary function of melatonin in photosynthetic cyanobacteria, which appeared on Earth 3.5–3.2 billion years ago, was as an antioxidant and that the melatonin‐synthesizing actions of the engulfed bacteria were retained when these organelles became mitochondria and chloroplasts, respectively. Expand
Protective action of melatonin against oxidative DNA damage: chemical inactivation versus base-excision repair.
TLDR
Melatonin can have a protective effect against oxidative DNA damage by chemical inactivation of a DNA-damaging agent as well as by stimulating DNA repair, but key factors in BER, viz. glycosylases and AP-endonucleases, do not seem to be affected by melatonin. Expand

Scavenging effect of melatonin on hydroxyl radicals generated by alloxan
TLDR
It is demonstrated that hydroxyl radicals (OHzrad;) are formed in vitro by alloxan in the presence of glutathione (GSH) and chelated divalent iron and the potential of melatonin to prevent lipid peroxidation is considerably less pronounced. Expand

Cyclic 3-Hydroxymelatonin: A Melatonin Metabolite Generated as a Result of Hydroxyl Radical Scavenging
TLDR
When exogenous melatonin was administered to young rats, urinary 3-OHM levels increased significantly in the treated rats compared to those in controls, indicating that even in young animals there is insufficient endogenously produced melatonin to detoxify the basal levels of the toxic HO·. Expand

Ninhydrin was found also to be hydrophilic although this compound, in contrast to alloxan and its derivatives, was quite stable in aqueous solution, which may explain why ninhydrin does not share with allxan a selective toxic effect upon the pancreatic B-cell. Expand
