Tixocortol pivalate is a corticosteroid with topical anti‐inflammatory activity equal to that of hydrocortisone. It was evaluated in a group of 18 normal subjects to determine whether it exerted any systemic glucocorticoid activity after single oral or intrarectal doses and after short‐term dosing by the intranasal route. Effects of tixocortol pivalate were compared to those of oral dexamethasone and intrarectal betamethasone 21‐phosphate. By the three routes, tixocortol pivalate does not…

Tixocortol Pivalate* and Beclomethasone Dipropionate in Seasonal Allergic Rhinitis: A Double-Blind Study
TLDR
A tolerance study in man has shown that at doses of 250-2000 mg/p.o.o, TP did not induce any systemic glucorcorticoid effect, and this low systemic effect is due to an extensive first pass metabolism giving poorly active or ineffective metabolites and conjugated metabolites.
Local anti-inflammatory activities of tixocortol 21-pivalate, inhibition of prostaglandins and leukotrienes synthesis, in carrageenin-induced pleurisy. Reversion of effects by RU 486
TLDR
Local co-administration of RU 486 with either TP, T or Dex reversed the anti-inflammatory effects of all steroids, indicating in these conditions a local activity through glucosteroid receptor occupancy.

TLDR
Oral, slow release preparations of budesonide are efficacious in the treatment of active ileocecal Crohn's disease (CD), causing less suppression of endogenous plasma cortisol levels than does oral prednisolone, and giving rise to fewer, and less severe, side effects.

Fluticasone propionate in Crohn's disease.
TLDR
Fluticasone propionate is a promising therapeutic agent for Crohn's disease that offers the possibility of controlling inflammation without inducing systemic corticosteroid side effects and which merits evaluation in a double blind trial versus conventional corticosticosteroids.
Direct inhibition of cyclooxygenase pathway in platelets by tixocortol 21-pivalate: comparison with related structures, steroidal and non steroidal anti-inflammatory drugs
TLDR
Results indicate that tixocortol 21-pivalate in contrast to other glucosteroids is able to act directly on cyclooxygenase pathway and that some specific chemical environment of the 21 thiol side chain are required for this inhibition.
The sensitizing capacity of tixocortol pivalate
TLDR
The authors speculated on whether both the hydrocortisone and the tixocortol p1valate might have been the source of the observed allergic reactions, and determmed the sens1tizmg potency of lixoc Cortol p 1valate.
New steroidal anti-inflammatory drugs
TLDR
The slow increase in the number of chemically distinct steroids in the post-dexamethasone era is indicative of the difficulty in synthesizing effective steroids with new functional groups at non-conventional positions or modifying the 3-one-4-ene, 11-hydroxy and 17-ketol groups which have been regarded as essential for glucocorticoid activities.

Pharmacological study of a new anti-inflammatory steroid, tixocortol pivalate (JO 1016).
TLDR
It is possible the local or topical use of tixocortol pivalate in therapy may not cause the unwanted side effects of many of the corticosteroids in current use.

TLDR
Tixocortol pivalate would, by virtue of its low toxicity and absence of general immunosuppression, have potential advantages over both hydrocortisone acetate and beclomethasone dipropionate in the topical treatment of allergic conditions of mucous membranes.
A comparison of the effects of tixocortol pivalate (JO 1016), hydrocortisone acetate and beclomethasone dipropionate on collagen synthesis and degradation.
TLDR
Results suggest that in clinical use tixocortol pivalate would be much less likely than hydrocortisone acetate or beclomethasone dipropionate to cause degenerative changes in connective tissue.
A comparison of the effects of tixocortol pivalate (JO 1016), hydrocortisone acetate and beclomethasone dipropionate on the phagocytosis and lysis of microorganisms by alveolar macrophages.
TLDR
It is suggested that in clinical use correspondingly higher doses of tixocortol pivalate could be given without danger of affecting either phagocytic activity or the immune response.
Rate of adrenal cortisol production in response to maximal stimulation with ACTH.
TLDR
The rate of cortisol production during maximal stimulation with ACTH can be determined with radioisotopes, but a practical non-isotopic technique would be of 4 value, so patients were studied won separate occasions with cortisol and ACTH infusions.
