We show experiments proving the feasibility of scanning fluorescence microscopy by three-photon excitation. Three-photon excitation fluorescence axial images are shown of polystyrene beads stained with the fluorophore 2,5-bis(4-biphenyl)oxazole (BBO). Three-photon excitation is performed at an excitation wavelength of 900 nm and with pulses of 130 fs duration provided by a mode-locked titanium sapphire laser. Fluorescence is collected between 350 and 450 nm. The fluorescence image signal… Expand

For a given illumination wavelength the resolution for imaging a thick object in the case of three-photon fluorescence imaging can be improved by as much as 40-50% relative to that in two-phorescence microscopic imaging. Expand
Two-photon microscopy and spectroscopy based on a compact confocal scanning head.
The average rotational and translational diffusion coefficients measured with fluorescence polarization anisotropy and fluorescence correlation methods are in good agreement with the protein size, therefore validating the use of the microscope for two-photon spectroscopy on biomolecules. Expand

Molecular photobleaching kinetics of Rhodamine 6G by one- and two-photon induced confocal fluorescence microscopy.
It is shown that fluorescence excitation is in all cases limited by photolysis from higher-excited electronic states, and comparison with experimental data and an exact theoretical model show that only minor deviations between the different theoretical approaches can be observed for high-pulsed excitation irradiances. On the possibility of calcium imaging using Indo-1 with three-photon excitation.
It is shown that the calcium fluorophore Indo-1 can be excited by simultaneous absorption of three-photons at 885 nm, a wavelength readily available from Ti:sapphire lasers, and may provide higher spatial resolution than available using two-photon excitation. Expand
