Targeting mechanisms of neutrophil elastase (NE) and other luminal proteins stored in myeloperoxidase (MPO)-positive secretory lysosomes/primary granules of neutrophils are unknown. These granules contain an integral membrane protein, CD63, with an adaptor protein-3-dependent granule delivery system. Therefore, we hypothesized that CD63 cooperates in granule delivery of the precursor of NE (proNE). Supporting this hypothesis, an association was demonstrated between CD63 and proNE upon…

Neutrophil elastase and proteinase 3 trafficking routes in myelomonocytic cells.
Trafficking and function of the tetraspanin CD63.
Expression and role of a2 vacuolar‐ATPase (a2V) in trafficking of human neutrophil granules and exocytosis
TLDR
Findings suggest that the granule‐associated a2V isoform has a role in maintaining a pH gradient within the cell between the cytosol and granules in neutrophils and also in fusion between the surface and the granules before exocytosis.
TIMP-1 signaling via CD63 triggers granulopoiesis and neutrophilia in mice
TLDR
Elevated levels of TIMP-1 impact on neutrophil homeostasis via signaling through CD63 is revealed, which may provide a link to clinical observations, where TIMp-1 correlates with high severity and bad prognosis in inflammation-associated diseases.

TLDR
The results suggest that CD63 is a membrane component of LDCVs and the stability ofCD63 is dependent on the Muted protein, which provides a clue to the pathogenesis of LRO defects in HPS.
Intracellular and cell-surface neutrophil proteinase levels and distribution : changes following extravasation and microbial infection
TLDR
It is demonstrated that membrane-bound Pr3 (mPr3) is still detectable on the surface of neutrophils in the presence of purified inhibitors and autologous serum as a source of physiological inhibitors.
Granule Protein Processing and Regulated Secretion in Neutrophils
TLDR
This review will focus on advancement in three topics relevant to neutrophil secretion: what is known about basal level pro-inflammatory mediator synthesis, trafficking, and storage in secretory compartments, and the upregulation and de novo synthesis of pro- inflammatory mediators by neutrophils engaged at sites of infection.
T47D Cells Expressing Myeloperoxidase Are Able to Process, Traffic and Store the Mature Protein in Lysosomes: Studies in T47D Cells Reveal a Role for Cys319 in MPO Biosynthesis that Precedes Its Known Role in Inter-Molecular Disulfide Bond Formation
TLDR
T47D breast cancer cells stably transfected with an MPO expression plasmid are able to efficiently replicate all of the processing steps that lead to formation of the mature MPO heterotetramer, and the role of candidate receptors in the endocytic trafficking of MPO is investigated.
The tetraspanin CD63 regulates ESCRT-independent and -dependent endosomal sorting during melanogenesis.
Tetraspanins as Regulators of Protein Trafficking
TLDR
It has recently become apparent that in addition to their purely structural function as organizers of TERM, tetraspanins also regulate various aspects of trafficking and biosynthetic processing of associated receptors.

CD63 tetraspanin slows down cell migration and translocates to the endosomal-lysosomal-MIICs route after extracellular stimuli in human immature dendritic cells.
TLDR
CD9, CD63, CD81, and CD82 could play a role in modulating the interactions between immature DCs and their environment, slowing their migratory ability, but only CD63 would intervene in the internalization of complex antigens.
The tetraspanin CD63 enhances the internalization of the H,K-ATPase β-subunit
TLDR
Data indicate that CD63 serves as an adaptor protein that links its interaction partners to the endocytic machinery of the cell and suggest a previously uncharacterized protein-trafficking role for the tetraspanins.

Innate immunity defects in Hermansky-Pudlak type 2 syndrome.
TLDR
Observations suggest that type 2 Hermansky-Pudlak syndrome is characterized by defects of innate immunity, specifically the impairment of NK cytolitic activity.
Azurophilic granules of human neutrophilic leukocytes are deficient in lysosome-associated membrane proteins but retain the mannose 6-phosphate recognition marker.
TLDR
The azurophilic granules, which contain an abundance of lysosomal enzymes and Man 6-P GP, lack the LAMP glycoproteins and cannot be classified as lysOSomes, but rather may have the functional characteristics of a regulated secretory granule.
