The kinetics of a single i. v. dose of theophylline given either alone or with flumequine was studied in eight healthy volunteers. No statistically significant differences were observed in the pharmacokinetic parameters of theophylline (volume of distribution, elimination half-life, AUC, plasma clearance) following the two treatments.Pretreatment for 5 days with oral flumequine (400 mg, three times daily) had no significant effect on the disposition of a single i. v. dose of theophylline in…
Hepatic P450 monooxygenase response in rainbow trout (Oncorhynchus mykiss (Walbaum)) administered aquaculture antibiotics
TLDR
Results of O-dealkylation, isoform-selective inhibition, and immunoblotting showed that the effects of both oxolinic acid and flumequine were related to P4501A subfamily.
Deciding on Success Criteria for Predictability of Pharmacokinetic Parameters from In Vitro Studies: An Analysis Based on In Vivo Observations
TLDR
Analysis of published systemic clearance and volume of distribution at steady state (Vss) values taken from over 200 clinical studies shows that in some cases the twofold criteria method is an unreasonable expectation when the observed data are obtained from studies with small sample size.
A Re-evaluation and Validation of Ontogeny Functions for Cytochrome P450 1A2 and 3A4 Based on In Vivo Data
Background and ObjectivesCurrent cytochrome P450 (CYP) 1A2 and 3A4 ontogeny profiles, which are derived mainly from in vitro studies and incorporated in paediatric physiologically based
APPLICATIONS OF PHYSIOLOGICALLY BASED PHARMACOKINETIC MODELLING TO PREDICTION OF THE LIKELIHOOD OF METABOLIC DRUG INTERACTIONS IN PAEDIATRIC POPULATION AND STUDYING DISPARITIES IN PHARMACOKINETICS BETWEEN CHILDREN AND ADULTS
TLDR
The outcome suggests that the current ontogeny profiles result in under-prediction of CL values compared to clinical studies in infants and children and there is a need for better ontogenY models.

The influence of quinolone derivatives on theophylline clearance.
TLDR
There is growing evidence that the observed interaction is caused not by the parent drugs, but by the 4-oxo metabolite of enoxacin, pefloxac in and ciprofloxACin and nalidixic acid.
Comparative pharmacokinetics of theophylline following two fluoroquinolones co-administration
SummaryPretreatment for 3 days with oral ofloxacin or norfloxacin had no significant effect on the disposition of a single i.v. dose the theophylline in healthy volunteers.
Plasma and urine levels of flumequine and 7-hydroxyflumequine following single and multiple oral dosing.
Plasma and urine concentrations of flumequine and its microbiologically active metabolite, 7-hydroxyflumequine, were determined in healthy subjects following single oral doses of 400, 800, and 1200
Inhibition of theophylline metabolism by long‐term allopurinol administration
TLDR
Giving in a dose of 300 mg every 12 hr for 14 or 28 days, allopurinol inhibits theophylline metabolism, despite the failure of alloporinol in lower doses given for only 7 days to alter theophyLLine disposition.
Pharmacokinetic Interactions Between Theophylline and Other Medication (Part II)
SummaryPart I of this article, which appeared in the previous issue of the Journal, covered the effects or lack of effects on theophylline clearance of sympathomimetics, corticosteroids,
Enoxacin decreases the clearance of theophylline in man.
TLDR
In patients treated concurrently with theophylline and enoxacin, a new broad-spectrum antibacterial agent of the quinolone class, unexpectedly high plasma theophyLLine concentrations were measured and it was concluded that the rise of plasma theophical concentrations is caused by a reduced metabolic clearance of theophylla.

TLDR
Theophylline clearance (clearance/bioavailability) was found to be increased by isoprenaline (isoproterenol), terbutaline, some corticosteroids, phenytoin, phenobarbital, activated charcoal, felodipine moricizine, benzodiazepines and sulfinpyrazone - typically by about 25%, but sometimes by as much as 80% or more.
Alteration of theophylline clearance and half-life by cimetidine in normal volunteers.
TLDR
Although cimetidine-theophylline interaction needs confirmation in a patient population, it is advised caution when these agents are coadministered, and some adjustment in theophyllines dosage may be needed in treated patients who are also given cimeidine.
More on Cimetidine — Theophylline Interaction
TLDR
There is undoubtedly considerable interindividual variation in the magnitude of the cimetidine-theophylline interaction, as Ambrose and Harralson suggest.
Evaluation of a Rapid High‐Performance Liquid Chromatographic Method for Plasma Theophylline Analysis and Its Use in Therapeutic Drug Monitoring
TLDR
A high-performance liquid chromatographic (HPLC) assay for the-ophylline has been developed and evaluated and was superior with respect to precision and less expensive to perform.
