The CYP4A gene subfamily is composed of a number of genes that encode cytochromes P450 from various species, including human, which catalyze the hydroxylation of various saturated and unsaturated fatty acids, including arachidonic acid and prostaglandins. CYP4A7, a fatty acid metabolizing cytochrome P450 from rabbit kidney, was expressed in E. coli by adding the first 10 codons of CYP17alpha producing final yields of 20 nmol/L in order to perform detailed kinetic and spectral studies. CYP4A7… Expand
Catalytic turnover of pyrene by CYP3A4: evidence that cytochrome b5 directly induces positive cooperativity.
TLDR
It is demonstrated that b5 could directly induce positive cooperativity on CYP3A4 and that this biological factor needs to be carefully considered when included in in vitro P450 reactions. Expand
Diversity in Mechanisms of Substrate Oxidation by Cytochrome P450 2D6
TLDR
The P450 systems utilizing NADPH-P450 reductase, cumene hydroperoxide, and iodosylbenzene use similar but distinct chemical mechanisms, which are the basis for the variable product distributions. Expand
Expressed CYP4A4 metabolism of prostaglandin E(1) and arachidonic acid.
TLDR
The enzyme was purified, and its activity with substrates prostaglandin E(1) (PGE(1)) and AA was examined, and a conformational change occurred in CYP4A4 protein upon binding of cyt b(5) or apo b( 5). Expand
Expression and characterization of a functional canine variant of cytochrome b5 reductase.
TLDR
A direct comparison of the enzymes from two phylogenetic sources using identical expression systems that indicate that both enzymes have comparable spectroscopic, kinetic, thermodynamic, and structural properties is provided. Expand
P450 reductase and cytochrome b5 interactions with cytochrome P450: effects on house fly CYP6A1 catalysis.
TLDR
CYP6A1 is a P450 involved in insecticide resistance, this suggests that xenobiotic metabolism by constitutively overexpressed P450s may be linked to significant oxidative stress in the cell that may carry a fitness cost. Expand
Modulation of human cytochrome P450 1A1-mediated oxidation of benzo[a]pyrene by NADPH:cytochrome P450 oxidoreductase and cytochrome b5.
TLDR
The effect of cytochrome b5 on CYP1A1 conformation and the electron transfer to this enzyme may contribute to the cyto chrome b5-mediated stimulation of BaP oxidation. Expand
Cloning and expression of koala (Phascolarctos cinereus) liver cytochrome P450 reductase.
TLDR
Primary molecular data regarding koala CPR1 and CPR2 genes in this unique marsupial species is provided, which is consistent with that reported for rat CPR enzyme. Expand
Novel insights into oxidation of fatty acids and fatty alcohols by cytochrome P450 monooxygenase CYP4B1.
CYP4B1 is an enigmatic mammalian cytochrome P450 monooxygenase acting at the interface between xenobiotic and endobiotic metabolism. A prominent CYP4B1 substrate is the furan pro-toxin 4-ipomeanolExpand
Cytochrome b5 Activates the 17,20-Lyase Activity of Human Cytochrome P450 17A1 by Increasing the Coupling of NADPH Consumption to Androgen Production.
TLDR
It is concluded that b5 stimulation of the 17,20-lyase reaction primarily derives from more efficient use of NADPH for product formation rather than side products. Expand
Defining the in Vivo Role for Cytochrome b5 in Cytochrome P450 Function through the Conditional Hepatic Deletion of Microsomal Cytochrome b5*S⃞
TLDR
Data indicate that microsomal cytochrome b5 can play a major role in the in vivo metabolism of certain drugs and chemicals but in a P450- and substrate-dependent manner. Expand

Identification of the Binding Site on Cytochrome P450 2B4 for Cytochrome b 5 and Cytochrome P450 Reductase*
TLDR
These studies indicate that the binding sites for cy tochrome b 5 and cytochrome P450 reductase are, as predicted, located on the proximal surface of cytochromes P450 2B4 and are partially overlapping but not identical. Expand

TLDR
It was found that the activity increase of rabbit P450 1A2 coincides with the conformational change including raised α-helix content, and it is proposed that the interaction with the phospholipid molecules surrounding P450 2A2 in the endoplasmic reticulum is important for a functional conformation of P 450 1A1 in a monooxygenase system including NADPH-P450 reductase. TLDR
Results indicate that when b5 is reconstituted with the cytochrome P-450 isozyme 2 enzyme system under optimal conditions, substrate monooxygenation is enhanced, NADPH oxidation is unaffected, and hydrogen peroxide formation is decreased. Expand
Catalytic properties of rabbit kidney fatty acid ω-hydroxylase cytochrome P-450ka2 (CYP4A7)
Abstract We have examined in detail the substrate specificity of a rabbit kidney fatty acid ω-hydroxylase, designated cytochrome P-450ka2 (CYP4A7). The hydroxylation products were identified as ω-Expand
Structural analysis of the FMN binding domain of NADPH-cytochrome P-450 oxidoreductase by site-directed mutagenesis.
TLDR
Replacement of Tyr-140 with aspartate did not eliminate FMN binding, but reduced cytochrome c reductase activity about 5-fold, suggesting that FMN may be bound in a conformation which does not permit efficient electron transfer. Expand
Cytochrome P450 4A expression and arachidonic acid omega-hydroxylation in the kidney of the spontaneously hypertensive rat.
TLDR
CYP4A1 might play a significant role in contributing to the increased cortical/proximal production of 20-HETE seen in 7-week-old SHR, but given the high homology among members of the CYP4A gene family and the lack of specific tools to discern among these isoforms, additional studies have to be carried out to substantiate these findings. Lack of Electron Transfer from Cytochrome b5 in Stimulation of Catalytic Activities of Cytochrome P450 3A4
TLDR
It is found that apo-b5, devoid of heme, could substitute for b5 in stimulating two prototypic activities, testosterone 6β hydroxylation and nifedipine oxidation, and can facilitate some P450 3A4-catalyzed oxidations by complexing with P4503A4 and enhancing its reduction by NADPH-P450 reductase, without directly transferring electrons to P450. TLDR
Analysis of nucleotide substitution patterns in closely related members in four subfamilies, CYP2A, 2B, 2C, and 2D, consistently indicated that the SRSs have accumulated more nonsynonymous (amino acid-changing) substitutions than the rest of the sequence. Expand

The results indicate that the enzyme whose mRNA is most highly induced by clofibric acid (P4504A6) and the enzyme selectively elevated during pregnancy (P 4504A4) both exhibit relatively high rates for the omega-hydroxylation of arachidonic acid. Expand
Branched Phosphatidylcholines Stimulate Activity of Cytochrome P450SCC (CYP11A1) in Phospholipid Vesicles by Enhancing Cholesterol Binding, Membrane Incorporation, and Protein Exchange*
TLDR
The data suggest that different properties of P450SCC in membrane systems including cholesterol binding, membrane integration, and protein exchange are affected by branched PCs and probably by other phospholipids, too, and therefore must be considered in an explanation of the observed high stimulation of activity.
