We have tested the effect of methotrexate (MTX) on platelet activating factor (PAF)-induced neutrophil and eosinophil locomotion, neutrophil leukotriene B4 (LTB4) generation and mononuclear cell DNA synthesis. Neutrophils from patients treated with low dose methotrexate showed reduced PAF-induced chemotactic responses (727.8 +/- 72.2/10 HPF vs 481.9 +/- 87.3/10 HPF, p < 0.05). Both MTX and the specific PAF antagonist BN-52021 significantly inhibited PAF-induced eosinophil and neutrophil… Expand

Low-dose methotrexate treatment in severe glucocorticoid-dependent asthma: effect on mucosal inflammation and in vitro sensitivity to glucocorticoids of mitogen-induced T-cell proliferation.
TLDR
The absence of an inhibitory effect on inflammatory cells in blood and mucosa suggests that this effect is achieved by modulating cell function rather than cell number. Expand
Alternate treatments in asthma.
TLDR
The type of patient who may benefit from alternate, nonsteroidal agents that have been evaluated in prospective, randomized trials or have novel mechanisms of action is characterized and the current evidence behind their use is examined. Expand
Recent trends and directions in the rationalization of pharmacotherapy of bronchial asthma: probing for alternatives.
TLDR
Developments in the available pharmacologic panorama for both chronic and acute therapy and the development and approval of novel medications are summarized and their implications in treatment of BA are summarized. Expand
Methotrexate as a steroid sparing agent for asthma in adults.
TLDR
Methotrexate may have a small steroid sparing effect in adults with asthma who are dependent on oral corticosteroids, but the overall reduction in daily steroid use is probably not large enough to reduce steroid-induced adverse effects. Expand

An updated therapeutic overview of unconventional treatment options for refractory severe asthma is provided to provide an update on the limited published literature on the unconventional treatments of refractary severe asthma. Expand

Inhibition of leukotriene B4 synthesis in neutrophils from patients with rheumatoid arthritis by a single oral dose of methotrexate.
TLDR
The absence of a significant change in either 3H-labeled arachidonic acid release or platelet-activating factor generation indicates that the observed decrease in LTB4 synthesis was apparently not caused by diminished phospholipase A2 activity, and inhibition of 5-lipoxygenase activity and possible suppression of leukotriene A4 epoxide hydrolase activity. TLDR
PAF-acether appears to be a potent eOSinophilotactic agent which may play a role in inflammatory reactions characterized by eosinophil infiltration. Expand

The results indicate that MTX has rapid effects on IgM-RF synthesis, and this action might be associated with its therapeutic efficacy in RA. Expand
Activation of antibody-dependent cell-mediated cytotoxicity of human neutrophils and eosinophils by separate colony-stimulating factors.
TLDR
The results support the notion that human CSF have two sites of action: i) the progenitor cell, where they stimulate a relatively slow process of differentiation, and ii) the mature cell,Where they have a rapid action of increasing functional capacity. Expand
The in vitro Effects of methotrexate on the phagocytosis and intracellular killing of staphylococcus aureus by human neutrophils
TLDR
In these experiments, the in vitro incubation of circulating human neutrophils with methotrexate produced no significant alterations in neutrophilic phagocytosis or intracellular killing of S. aureus. Expand
The identification and partial characterization of a human mononuclear cell-derived neutrophil chemotactic factor apparently distinct from IL-1, IL-2, GM-CSF, TNF and IFN-gamma.
TLDR
Optimal chemotactic concentrations of partially purified MNC-derived NCF were of comparable potency to FMLP and LTB4 and had about 60% of the activity of optimal concentrations of C5a, C4a-des-Arg and platelet-activating factor (PAF). Expand
Bronchoalveolar eosinophilia during allergen-induced late asthmatic reactions.
TLDR
Bronchoalveolar lavage was performed in 19 asthmatic patients and in 5 control subjects to suggest that eosinophils and their mediators might be involved in the development of LAR after allergen inhalation. Expand

The data demonstrate that, in addition to causing release of endogenous substrate, the ionophore also activated the enzymatic system involved in the further transformations of arachidonic acid. Expand
Lack of benefit of methotrexate in severe, steroid-dependent asthma. A double-blind, placebo-controlled study.
TLDR
The study does not support the use of methotrexate in the treatment of severe asthma, and no significant benefit of metotrexate on asthma control could be shown. Expand
