2-BFI (2-(2-benzofuranyl)-2-imidazoline) is a prototypical I2-imidazoline receptor ligand. In vivo, however, 2-BFI (1-20 mg/kg) decreased the synthesis of dopa/dopamine (DA) in rat striatum through mechanisms not related to interaction with I2-imidazoline receptors or to inhibition of the enzyme monoamine oxidase. The aim of this study was to unravel the mechanism underlying this potent effect of 2-BFI in brain. In vitro 2-BFI showed very low affinity for D2-dopamine receptors (K(i)=47 microM… Expand
The effect of 1-(4,5-dihydro-1H-imidazol-2-yl) isoquinoline on monoamine release and turnover in the rat frontal cortex
TLDR
Assessment of the pharmacological actions of BU98008 in a functional in vivo model using the technique of in vivo brain microdialysis shows some functional similarities with known selective I(2)-BS ligands. Expand
The potential antidepressant-like effect of imidazoline I2 ligand 2-BFI in mice
TLDR
The results suggest that the antidepressant-like effect of 2-BFI might involve serotonergic, dopaminergic and imidazoline systems, and then the imidrazoline site could represent a new pharmacological target for the treatment of depression. Expand
Chronic treatment with selective I2-imidazoline receptor ligands decreases the content of pro-apoptotic markers in rat brain
TLDR
The disclosed anti-apoptotic mechanisms of selective I-imidazoline drugs may work in concert with other molecular mechanisms of neuroprotection (e.g. blockade of NMDA receptors) that are engaged by I2-ligands. Expand
Behavioral effects of the imidazoline I2 receptor ligand BU99006 in rats
TLDR
The data suggest that BU99006 is an imidazoline I2 receptor agonist with no evidence of I2 receptors antagonism in rats, and that it enhanced the discriminative stimulus and rate-suppressing effects of 2-BFI. Expand
Potential serotonergic and noradrenergic involvement in the discriminative stimulus effects of the selective imidazoline I2-site ligand 2-BFI
TLDR
The concept that the 2-BFI cue may contain both noradrenergic and serotonergic components is supported, as it is shown that the administration of I2-site ligands leads to an increase in extracellular levels of monoamines. Expand
Attenuation of acute and chronic effects of morphine by the imidazoline receptor ligand 2‐(2‐benzofuranyl)‐2‐imidazoline in rat locus coeruleus neurons
TLDR
It is demonstrated that 2‐(2‐benzofuranyl)‐2‐imidazoline (2‐BFI) interacts with the opioid system in the rat locus coeruleus and this interaction leads to an attenuation of both the hyperactivity of locus coercionuleus neurons during opiate withdrawal and the development of tolerance to morphine when 2‐B FI is chronically administered. Effects of imidazoline I2 receptor agonists on reserpine-induced hyperalgesia and depressive-like behavior in rats
TLDR
Examination of the antinociceptive and antidepressant-like effects of imidazoline I2 receptor (I2R) agonists in a reserpine-induced model of fibromyalgia in rats suggests that I2R agonists may be useful to treat Fibromyalgia-related pain and comorbid depression. Expand
Locomotor effects of imidazoline I2‐site‐specific ligands and monoamine oxidase inhibitors in rats with a unilateral 6‐hydroxydopamine lesion of the nigrostriatal pathway
TLDR
It is suggested that I2‐specific ligands have dual effects in the 6‐OHDA‐lesioned rat model of Parkinson's disease; a first effect associated with an increase in activity in the intact hemisphere, and a secondary action which, through the previously documented inhibition of MAO‐A and/orMAO‐B, increases the availability of dopamine produced by L‐DOPA. Expand
Novel imidazoline compounds as partial or full agonists of D2-like dopamine receptors inspired by I2-imidazoline binding sites ligand 2-BFI.
TLDR
The novel derivatives 3a-k inspired by 2-BFI scaffold were prepared to assess imidazoline molecules as D(2)-like dopamine receptor ligands and provided partial (3f) or nearly full (3e and 3h) agonists, all endowed with D( 2)-like potency comparable to that of dopamine. Expand
Clorgyline-induced switch from locomotion to mouthing in sensitization to the dopamine D2/D3 agonist quinpirole in rats: role of sigma and imidazoline I2 receptors
TLDR
The results are consistent with the suggestion that clorgyline and Ro 41-1049 affect the behavioral response to quinpirole via the MAOI-displaceable quinPirole binding (MQB) site, and the hypothesis that the MQB site selects what motor output becomes sensitized to repeated injections of quin pirole. Effects of imidazoline receptor ligands on monoamine synthesis in the rat brain in vivo
TLDR
Most imidazoline drugs modulated the synthesis of brain monoamines through interaction with α2-adrenoceptors or 5-HT1A receptors through contact with α1/α2 receptors, and the results do not provide functional evidence for the existence of presynaptic imidrazoline receptors regulating the synthesisof monoamines in the rat brain. Inhibition of monoamine oxidase A and B activities by imidazol(ine)/guanidine drugs, nature of the interaction and distinction from I2‐imidazoline receptors in rat liver
TLDR
The main aim of this study was to assess the inhibitory effects and nature of the inhibition of imidazol(ine)/guanidine drugs on rat liver MAO‐A andMAO‐B isoforms and to compare their inhibitory potencies with their affinities for the sites labelled by [3H]‐clonidine in the same tissue. Expand
Modulation of MAO Activity by Imidazoline and Guanidine Derivatives
TLDR
The strong inhibitory effects of I2 selective imidazoline ligands confirm the existence of I1‐BS as a regulatory site on MAO, and catecholamine content of livers obtained from animals treated with different IBS‐ligands was consequently increased. Expand

Ligand: a versatile computerized approach for characterization of ligand-binding systems.
TLDR
This approach provides two major advantages compared with other available methods: it uses an exact mathematical model of the ligand-binding system, thereby avoiding the possible biases introduced by several commonly used approximations and it uses a statistically valid, appropriately weighted least-squares curve-fitting algorithm with objective measurement of goodness of fit.
