Escherichia coli has the capacity to synthesise three distinct formate dehydrogenase isoenzymes and three hydrogenase isoenzymes. All six are multisubunit, membrane-associated proteins that are functional in the anaerobic metabolism of the organism. One of the formate dehydrogenase isoenzymes is also synthesised in aerobic cells. Two of the formate dehydrogenase enzymes and two hydrogenases have a respiratory function while the formate dehydrogenase and hydrogenase associated with the formate… Expand
Biosynthesis of the respiratory formate dehydrogenases from Escherichia coli: characterization of the FdhE protein
TLDR
It is shown that E. coli FdhE interacts with the catalytic subunits of the respiratory formate dehydrogenases, and site-directed mutagenesis shows that conserved cysteine motifs are essential for the physiological activity of the Fdh E protein and are also involved in iron ligation. Expand
The respiratory molybdo-selenoprotein formate dehydrogenases of Escherichia coli have hydrogen: benzyl viologen oxidoreductase activity
TLDR
It is demonstrated that the energy-conserving selenium- and molybdenum-dependent formate dehydrogenases Fdh-N and F dh-O of Escherichia coli exhibit a degree of promiscuity with respect to the electron donor they use and identify a new class of dihydrogen-oxidizing enzyme. Expand
Anaerobic Formate and Hydrogen Metabolism.
TLDR
Clear evidence now exists indicating that Escherichia coli can mature Hyd enzymes differentially, depending on metal ion availability and the prevailing metabolic state. Expand
Structural and Functional Features of Formate Hydrogen Lyase, an Enzyme of Mixed-Acid Fermentation from Escherichia coli
TLDR
Formate hydrogen lyase from Escherichia coli is a membrane-bound complex that oxidizes formic acid to carbon dioxide and molecular hydrogen, which depends on the external pH and the presence of formate. Expand
Differential effects of isc operon mutations on the biosynthesis and activity of key anaerobic metalloenzymes in Escherichia coli.
Escherichia coli has two machineries for the synthesis of FeS clusters, namely Isc (iron-sulfur cluster) and Suf (sulfur formation). The Isc machinery, encoded by the iscRSUA-hscBA-fdx-iscXoperon,Expand
Complex Multimeric [FeFe] Hydrogenases: Biochemistry, Physiology and New Opportunities for the Hydrogen Economy
TLDR
The discovery of [FeFe] hydrogenases coupled to electron bifurcating subunits solved the enigma of how the otherwise highly endergonic reduction of the electron carrier ferredoxin can be carried out and how H2 production from NADH is possible. Expand
Bacterial formate hydrogenlyase complex
TLDR
In this study, innovative molecular biology and electrochemical experiments reveal a hydrogenase enzyme with the unique ability to sustain H2 production even under high partial pressures of H2, and protein film electrochemistry experiments on Hyd-3 demonstrate that it has a unique ability among [NiFe] hydrogenases to catalyze production of H1 even at high partial pressure. TLDR
The versatile hydrogen metabolism of E. coli will be discussed and the roles and potential applications of the spectrum of different types of [NiFe]-hydrogenases available will be explored. Expand
The Aerobic and Anaerobic Respiratory Chain of Escherichia coli and Salmonella enterica: Enzymes and Energetics.
TLDR
The energetics of the individual redox reactions and the respiratory chains is described and related to the H+/2e- ratios. Expand
Dissecting the roles of Escherichia coli hydrogenases in biohydrogen production.
TLDR
The effect of a deletion in hycA on H2 production was found to be dependent upon environmental conditions, but H2 uptake was not significantly affected by this mutation, and E. coli to perform anaerobic mixed-acid fermentation is therefore an attractive approach for bio-hydrogen production from sugars. TLDR
A biochemical and immunological study has revealed a new formate dehydrogenase isoenzyme in Escherichia coli that cross reacts with antibodies raised to FDH-N and possesses a similar polypeptide composition to FDh-N, which seems likely to functions physiologically as a formate: quinone oxidoreductase. Escherichia coli formate-hydrogen lyase. Purification and properties of the selenium-dependent formate dehydrogenase component.
TLDR
The formate-hydrogen lyase complex of Escherichia coli decomposes formic acid to hydrogen and carbon dioxide under anaerobic conditions in the absence of exogenous electron acceptors and inhibited FDHH activity, which protected the enzyme from inactivation by oxygen. Expand
Molecular biology of hydrogen utilization in aerobic chemolithotrophs.
TLDR
Maturation of both types of hydrogenase is apparently complex, involving specific nickel incorporation and proteolytic processing steps, and in Alcaligenes eutrophus and Rhodobacter capsulatus, hydrogenase expression is regulated by transcriptional activators belonging to the response-regulator family. Expand
Cloning and nucleotide sequence of the structural genes encoding the formate dehydrogenase of Wolinella succinogenes
TLDR
The formate dehydrogenase of Wolinella succinogenes is a membraneous molybdo-enzyme which is involved in phosphorylative electron transport and the genes fdhA and C encode larger preproteins which differ from the corresponding mature proteins by N-terminal signal peptides. Expand

TLDR
Redox control of the synthesis of hydrogenase and formate dehydrogenase acts at the level of transcription; the size of the redox potential seems to be correlated with the amount of repression; β-galactosidase synthesis in the hyd:: lac and fdh::lac fusion strains is induced by formate. Expand
