The erythromycin-biosynthetic (ery) gene cluster of Aeromicrobium erythreum was cloned and characterized. The 55.4-kb cluster contains 25 ery genes. Homologues were found for each gene in the previously characterized ery gene cluster from Saccharopolyspora erythraea. In addition, four new predicted ery genes were identified. Two of the new predicted genes, coding for a phosphopantetheinyl transferase (eryP) and a type II thioesterase (eryTII), were internal to the ery cluster. The other two new…
A key developmental regulator controls the synthesis of the antibiotic erythromycin in Saccharopolyspora erythraea
TLDR
Results suggest that BldD concurrently regulates the synthesis of erythromycin and morphological differentiation, and the ery genes are the first direct targets of a Bld D ortholog to be identified that are positively regulated.
Knockout of the Erythromycin Biosynthetic Cluster Gene, eryBI, Blocks Isoflavone Glucoside Bioconversion during Erythromycin Fermentations in Aeromicrobium erythreum but Not in Saccharopolyspora erythraea
TLDR
This study showed that isoflavone metabolism could be blocked in A. erythreum by ery BI knockout but that eryBI knockout was not sufficient to block is oflav one metabolism in S. eries, suggesting that other β-glucosidases are present.
Engineered EryF hydroxylase improving heterologous polyketide erythronolide B production in Escherichia coli
TLDR
The designed mutant of SaEryF, I379V, achieved the yield of 131 mg l−1 EB, which was fourfold to that produced by wild‐type SaEyF, and the combined mutagenesis of multiple residues led to further boost the EB concentration by another 41%, which laid the foundation for efficient heterologous biosynthesis of erythromycin or other complex polyketides.
Genome Sequence of Aeromicrobium erythreum NRRL B-3381, an Erythromycin-Producing Bacterium of the Nocardioidaceae
TLDR
Aeromicrobium erythreum NRRL B-3381 has a 3,629,239-bp circular genome that has 72% G+C content and provides resources for metabolic and combinatorial engineering of polyketides.
Streptomyces autolyticus JX-47 Large-Insert Bacterial Artificial Chromosome Library Construction and Identification of Clones Covering Geldanamycin Biosynthesis Gene Cluster
TLDR
A bacterial artificial chromosome library with an average insert size of up to 150 kb was constructed from genomic DNA of Streptomyces autolyticus JX-47 and revealed that geldanamycin had the same gene arrangement and high gene homology in the polyketide synthase (PKS) region and its downstream with 84–100% DNA identity and 81-100% amino acid (AA) identity.
An erythromycin process improvement using the diethyl methylmalonate-responsive (Dmr) phenotype of the Saccharopolyspora erythraea mutB strain
TLDR
The Saccharopolyspora erythraea mutB knockout strain, FL2281, was found to carry a diethyl methylmalonate-responsive (Dmr) phenotype, which represents a new class of strain improvement phenotype and a theory to explain the biochemical mechanism for the Dmr phenotype is proposed.
The Use of ClusterMine360 for the Analysis of Polyketide and Nonribosomal Peptide Biosynthetic Pathways.
TLDR
The ClusterMine360 database, which stores the antiSMASH-based annotation of gene clusters in the NCBI database, linking the structure of the natural product to the biosynthetic gene cluster, is developed.
Metagenomics and Antibiotic Discovery from Uncultivated Bacteria
TLDR
Metagenomic cloning is a promising approach for mining the extensive chemical diversity of different microbial populations, in particular by removing the past constraint of cultivation and allowing equal access to the large proportion of the community comprising yet uncultivated members.

Analysis of eryBI, eryBIII and eryBVII from the erythromycin biosynthetic gene cluster in Saccharopolyspora erythraea
TLDR
Specific mutants were created in eryBI, and the resulting strains were found to synthesise erythromycin A, showing that this gene, despite its position in the biosynthetic gene cluster, is not essential for erystromycin biosynthesis.
Organization of a cluster of erythromycin genes in Saccharopolyspora erythraea.
TLDR
The new Ery phenotype, EryH, was marked by the accumulation of the intermediate 6-deoxyerythronolide B (DEB), suggesting a defect in the operation of the C-6 hydroxylase system, and a block in the synthesis or addition reactions for the first sugar group.
Analysis of seven genes from the eryAI –eryK region of the erythromycin biosynthetic gene cluster in Saccharopolyspora erythraea
TLDR
R roles are proposed for several of these ORFs in the biosynthesis of the deoxysugar mycarose and the deoxyaminosugar desosamine in the macrolide antibiotic erythromycin A by Saccharopolyspora erystraea.
Targeted gene inactivation for the elucidation of deoxysugar biosynthesis in the erythromycin producer Saccharopolyspora erythraea
TLDR
The nucleotide sequence of three such ORFs located immediately downstream of eryA, ORFs 7, 8 and 9 are reported, which considerably strengthen previous tentative proposals for the pathways for the biosynthesis of dTDP-D-desosamine and dTSP-L-mycarose in Sac.
Molecular characterization of a gene from Saccharopolyspora erythraea (Streptomyces erythraeus) which is involved in erythromycin biosynthesis
TLDR
The eryC1‐gene product, a protein of subunit Mr 39200, is therefore involved either as a structural or as a regulatory gene in the formation of the deoxyamino‐sugar desosamine or in its attachment to the macro‐lide ring.
Cloning and sequence analysis of genes involved in erythromycin biosynthesis in Saccharopolyspora erythraea: sequence similarities between EryG and a family of S-adenosylmethionine-dependent methyltransferases
TLDR
Comparison of the amino acid sequence of EryG with the sequence of other S-adenosylmethionine (SAM)-dependent methyltransferases has revealed that one of the sequence motifs previously suggested to be part of the SAM-binding site is present not only in EyG but also in many other recently sequenced SAM-dependent methyl transferases.
