Treatment of rat hepatocytes with 0.5 mM concentrations of uridine and cytidine results in increased cellular concentrations of UTP, UDP-sugars and CTP, whereas that of CMP-N-acetylneuraminate remained unchanged [Pels Rijcken, Overdijk, Van den Eijnden and Ferwerda (1993) Biochem. J. 293, 207-213]. The incorporation of radioactivity from 3H-labelled sugars into the cell-associated and secreted glycoconjugate fraction was influenced by these altered cellular concentrations of the nucleotides…

The results demonstrate that isotope labeling in combination with MALDI-TOF-MS can be used as an alternate tool to radioactive labeling of sugar substrates in metabolic studies and is of significance for glycoprotein production in mammalian cell culture.
Incorporation of ammonium into intracellular UDP-activated N-acetylhexosamines and into carbohydrate structures in glycoproteins.

The ammonium action on the synthesis of the intracellular UDP-N-acetylhexos- amines (UDPGNAc), which are precursors of glycosylation as well as on N-linked oligosaccharides of a recombinant human IL-2 mutant variant model glycoprotein expressed in BHK-21 cells, is investigated.
Role of Nucleotide Sugar Pools in the Inhibition of NCAM Polysialylation by Ammonia
- Biology, ChemistryBiotechnology progress
- 1998
In an attempt to increase NCAM polysialylation, N‐acetylmannosamine and cytidine were added to cultures in order to circumvent the feedback inhibition of CMP‐sialic acid synthesis, but this only slightly increased PolySia levels and failed to counter ammonia's inhibition of NCAMPolySia.
Liver-specific increase of UTP and UDP-sugar concentrations in rats induced by dietary vitamin B6-deficiency and its relation to complex N-glycan structures of liver membrane-proteins

It is demonstrated that, especially in liver, pyridoxal phosphate is closely involved in the control of uracil nucleotides during a defined period of development, and in vivo N-glycan biosynthesis in liver is regulated by a more complex or higher mechanism than substrate concentrations.

The Tissue Concentration of UDP-N-acetylglucosamine Modulates the Stimulatory Effect of Insulin on Skeletal Muscle Glucose Uptake*
- Biology, ChemistryThe Journal of Biological Chemistry
- 1997
It is suggested that the marked reduction in insulin action induced by GlcN and uridine is mediated by increased accumulation of muscle UDP-N-acetylhexosamines, perhaps via altered glycosylation of protein(s) in GLUT4-containing vesicles.

The incomplete intracellular sialylation of interferon-gamma (IFN-Gamma), produced by Chinese hamster ovary cell culture, was minimized by supplementing the culture medium with N-acetylmannosamine (ManNAc), a direct intrace cellular precursor for sialic acid synthesis.

Intracellular UDP−N‐Acetylhexosamine Pool Affects N‐Glycan Complexity: A Mechanism of Ammonium Action on Protein Glycosylation
- Biology, ChemistryBiotechnology progress
- 1998
It is shown that ammonium ions act on protein glycosylation by at least two independent mechanisms, one of which involves an increase in the UDPGNAc pool, which is responsible for the ammonium‐induced increase in complexity and decrease in sialylation state of the N‐linked oligosaccharide.
