It is likely that a proportion of people treated with the anti‐schistosomicidal drug praziquantel (PZQ) is also taking other drugs such as chloroquine (CHQ), a widely used anti‐malarial. The effect of CHQ on the pharmacokinetics and metabolism of PZQ in rats and in humans was therefore studied. CHQ decreased the bioavailability of PZQ and reduced its maximum serum concentrations to a significant extent in rats and in humans. The clearance was increased to a statistically significant extent in… Expand
Drug metabolism and pharmacokinetics of praziquantel: A review of variable drug exposure during schistosomiasis treatment in human hosts and experimental models
TLDR
The study confirmed previous findings of higher antischistosomal activity of (R)-PZQ enantiomer when administered alone compared to the racemate at the same dose as well as improved efficacy when the drug is administered with food. Expand
Effect of Dihydroartemisinin-Piperaquine on the Pharmacokinetics of Praziquantel for Treatment of Schistosoma mansoni Infection
TLDR
DHP co-administration significantly increases systemic PZq exposure, and this may contribute to increased effectiveness of PZQ + DHP combination therapy than PZZQ alone to treat schistosomiasis. Expand
The Contributions of Various Chloroquine Salts to the Biliary and Urinary Execretion of Hepatic Paracetamol Conjugation Metabolites in the Rat
TLDR
Chloroquine sulphate is therefore a better choice compared to other chloroquine salts in the treatment of malaria with paracetamol as an antipyretic and analgesic. Expand
Antiprotozoal and Anthelmintic Agents
TLDR
The potential for interaction between antiparasitics and antiretroviral drugs as there is extensive overlap in patient populations likely infected with both malaria and HIV, which may lead to increased probability of polypharmacy and drug interactions. Expand
Antiprotozoal and Anthelmintic Agents
TLDR
This chapter reviews the key interactions likely to occur or those that have been seen to occur when drugs are administered with food or with other agents in the management of malaria and helminthiases. Expand
Review on the clinical pharmacology of hydroxychloroquine sulfate for the treatment of COVID-19.
TLDR
It has been proved that HCQ, which has an established safety profile, is effective against SARS-CoV-2 with sufficient pre-clinical rationale and evidence. Expand
The potential inhibitory effect of antiparasitic drugs and natural products on P-glycoprotein mediated efflux.
TLDR
It is demonstrated that a substantial number of antiparasitic and natural compounds, in a range of concentrations, are capable of inhibiting Pgp mediated [3H]-taxol efflux in Caco-2 cells, without being substrates and this may have implications for drug interactions with Pgp. Expand
Pharmacokinetic interaction between ketoconazole and praziquantel in healthy volunteers
Background: Praziquantel, a broad‐spectrum anthelminthic, has been reported to undergo extensive first‐pass metabolism by cytochrome P450 (CYP) enzymes in vivo. Ketoconazole, a potent CYP3A4Expand
Metabolism and pharmacokinetics of pharmaceuticals in cats (Felix sylvestris catus) and implications for the risk assessment of feed additives and contaminants.
TLDR
This paper aims to assess the scientific basis and validity of the UF for inter-species differences in kinetics and the extra UF applied for cats through a comparison of kinetic parameters between rats and cats for 30 substrates of phase I and phase II metabolism. Expand
O efeito da pressão seletiva com praziquantel na diversidade genética da cepa LE de Schistosoma mansoni.
TLDR
Obtaining a resistant strain to PZQ is important for a better understanding of the parasite resistance mechanisms which would be also useful for further comparisons of sensitivity levels among field isolates. Expand

TLDR
The results suggest that the inhibitors tested may suppress the metabolism of praziquantel in humans and consequently increase the bioavailability and blood levels at doses common in human therapy. Expand
