Lentiviruses, the prototype of which is HIV-1, can initiate translation either by the classical cap-dependent mechanism or by internal recruitment of the ribosome through RNA domains called IRESs (internal ribosome entry sites). Depending on the virus considered, the mechanism of IRES-dependent translation differs widely. It can occur by direct binding of the 40S subunit to the mRNA, necessitating a subset or most of the canonical initiation factors and/or ITAF (IRES trans-acting factors…

Internal translation initiation from HIV-1 transcripts is conferred by a common RNA structure
TLDR
The data point to a model in which the common exon on HIV-1 transcripts acts as the ribosome landing pad, recruiting preinitiation complexes upstream of the initiation codon, followed by scanning to each transcript's initiator AUG.
Activity of the human immunodeficiency virus type 1 cell cycle-dependent internal ribosomal entry site is modulated by IRES trans-acting factors
TLDR
Data show that HIV-1 IRES-mediated translation initiation is modulated by cellular proteins, suggesting that the requirement for pre-formed stable secondary or tertiary structure within the HIV- 1 IRES may not be as strict as has been described for other viral IRESes.
HIV-1 Transcripts Use IRES-Initiation under Conditions Where Cap-Dependent Translation Is Restricted by Poliovirus 2A Protease
TLDR
During the first 24–48 hours of viral replication HIV-1 protein synthesis is strongly dependent on Cap-initiation, while at later time points IRES-driven translation initiation is sufficient to produce high amounts of viral particles.
A conserved structure within the HIV gag open reading frame that controls translation initiation directly recruits the 40S subunit and eIF3
TLDR
This work proposes a novel two step model whereby the initial event is the formation of a ternary eIF3/40S/IRES complex, and the level of information compaction within this viral genome reveals an additional level of evolutionary constraint on the coding sequence.
Two ribosome recruitment sites direct multiple translation events within HIV1 Gag open reading frame
In the late phase of the HIV virus cycle, the unspliced genomic RNA is exported to the cytoplasm for the necessary translation of the Gag and Gagpol polyproteins. Three distinct translation
Two ribosome recruitment sites direct multiple translation events within HIV1 Gag open reading frame
TLDR
Insight is provided into the Gag-IRES molecular mechanism and original hypotheses about the IRES physiological role and conservation among primate lentiviruses are proposed.
LOOP IIId of the HCV IRES is essential for the structural rearrangement of the 40S-HCV IRES complex
TLDR
This study suggests that the formation of a ‘kissing complex’ between the viral RNA and the 18S ribosomal RNA locks the 40S Ribosomal subunit in a conformation proficient for translation.
Functional and Structural Analysis of the Internal Ribosome Entry Site Present in the mRNA of Natural Variants of the HIV-1
TLDR
The secondary structure of the variant HIV-1 5′UTRs did not reveal significant variations when compared to the pNL4.3.3-5′UTR, and the increased IRES-activity observed for some of the HIV- 1 variants cannot be ascribed to a specific structural modification.

Lentiviral RNAs can use different mechanisms for translation initiation.
TLDR
The results show that HIV-1 is able to drive the synthesis of the Gag polyprotein both by a classical cap-dependent mechanism and an IRES, whereas HIV-2 and SIV appear to use exclusively anIRES mechanism.
A new type of IRES within gag coding region recruits three initiation complexes on HIV-2 genomic RNA
TLDR
The conservation of the IRES among fast evolving lentiviruses suggests an important physiological role and the structural and functional properties of gag coding sequence define a new type of IRES.
RNA structure-based ribosome recruitment: lessons from the Dicistroviridae intergenic region IRESes.
TLDR
This review describes and compares mechanistic and structural groups of viral IRES RNAs, and discusses in greater detail some recent advances in the understanding of the intergenic region IRESes of the Dicistroviridae, which use the most streamlined ribosome-recruitment mechanism yet discovered.
An Internal Ribosome Entry Segment Promotes Translation of the Simian Immunodeficiency Virus Genomic RNA*
TLDR
DNA transfection and viral transduction experiments in both NIH-3T3 and COS-7 cells confirmed that translation driven by the SIV leader is IRES-dependent and thus insensitive to the immunosuppressant rapamycin.
A search for structurally similar cellular internal ribosome entry sites
TLDR
It is proposed that cellular IRES are not defined by overall structure, as viral IRES, but are instead dependent upon short motifs and trans-acting factors for their function.
HIV-2 genomic RNA contains a novel type of IRES located downstream of its initiation codon
TLDR
It is reported that translation of the genomic RNA of human immunodeficiency virus type 2 takes place by attachment of the 48S ribosomal preinitiation complex to the coding region, with no need for an upstream 5′ untranslated RNA sequence.
The Human Immunodeficiency Virus Type 1gag Gene Encodes an Internal Ribosome Entry Site
TLDR
This report shows that mRNAs containing the human immunodeficiency virus type 1 (HIV-1) gag open reading frame (ORF) exhibit internal ribosome entry site (IRES) activity that can promote translational initiation of Pr55 gag, and suggests that this low-abundance Gag isoform may be important for wild-type replication of HIV-1 in cultured cells.
A Dormant Internal Ribosome Entry Site Controls Translation of Feline Immunodeficiency Virus
TLDR
Surprisingly, infection of feline cells with FIV, but not human immunodeficiency virus type 1, resulted in a great increase in FIV translation.
