Primary microcephaly genes (MCPH) are required for the embryonic expansion of the mammalian cerebral cortex. However, MCPH mutations may spare growth in other regions of the developing forebrain which reinforces context-dependent functions for distinct MCPH genes in neurodevelopment. Mutations in the MCPH2 gene, WD40-repeat protein 62 (WDR62), are causative of primary microcephaly and cortical malformations in humans. WDR62 is a spindle microtubule-associated phosphoprotein that is required for…

To the knowledge, this is the first case of 19q13 microdeletion syndrome to ever be reported in Romania and it is believed that this case presents additional features that have never been previously reported in this syndrome, namely, dilatation of the third ventricle and subependymal cyst, left iris coloboma, and tracheomalacia.

It is found that WDR62 mutant proteins (V66M and R439H) localize to the basal body but fail to recruit CPAP, a protein that is required for cilia formation, which underpins the maintenance of radial glia and implicates ciliary dysfunction as underlying the pathology of MCPH2 patients.

In human and mouse embryonic brain, it was found that WDR62 expression was restricted to neural precursors undergoing mitosis, lending support to the hypothesis that the exquisite control of the cleavage furrow orientation in mammalian neural precursor cell mitosis is critical both in causing MCPH when perturbed and, when modulated, generating the evolutionarily enlarged human brain.

The results suggest that Wdr62 interacts with Aurora A to control mitotic progression, and loss of these interactions leads to mitotic delay and cell death of NPCs, which could be a potential cause of human microcephaly.
