Trafficking of the Na+/H+ exchanger isoform 3 (NHE3) between sub-apical vesicles and apical membrane of epithelial cells is a suggested mechanism of regulation of NHE3 activity. When epitope-tagged NHE3 was stably expressed in NHE-deficient Chinese hamster ovary cells, a sizable fraction was found in recycling endosomes. This system was used to analyze the mechanism of endocytosis of NHE3. Immunofluorescence and radiolabeling experiments showed that inhibition of clathrin-mediated endocytosis…

Clathrin-mediated Endocytosis and Recycling of the Neuron-specific Na+/H+ Exchanger NHE5 Isoform
TLDR
Observations demonstrate that NHE5 is localized to the recycling endosomal pathway and is dynamically regulated by phosphatidylinositol 3′-kinase and by the state of F-actin assembly.
Carbachol-Mediated Endocytosis of NHE3 Involves a Clathrin-Independent Mechanism Requiring Lipid Rafts and Cdc42
TLDR
Basal NHE3 activity was increased in the presence of CME blockers (chlorpromazine; K+ depletion) supporting previous reports that constitutive NHE2 endocytosis is clathrin dependent, and CCH-mediated inhibition of N HE3 activity is not dependent on clathin and involves lipid rafts and requires Cdc42.
NHE3 serves as a molecular tool for cAMP-mediated regulation of receptor-mediated endocytosis.
TLDR
NHE3 is important for cAMP sensitivity of clathrin-dependent RME, using megalin/cubilin-mediated albumin uptake in opossum kidney cells and transfection into deficient cells restores RME and the effects of cAMP.
Cyclic AMP-mediated endocytosis of intestinal epithelial NHE3 requires binding to synaptotagmin 1.
TLDR
It is concluded that intestinal epithelial Syt 1 plays an important role in cAMP-stimulated endocytosis of apical NHE3 throughcAMP-dependent phosphorylation of S605 that is required for NHE2 and Syt1 association and internalization.
Regulation of apical NHE3 trafficking by ouabain-induced activation of the basolateral Na+-K+-ATPase receptor complex.
TLDR
It is proposed that ouabain can simultaneously regulate basolateral Na(+)-K(+-ATPase and apical NHE3, leading to inhibition of transepithelial Na(+) transport, which may be relevant to proximal tubular Na(+, handling during conditions associated with increases in circulating endogenous cardiotonic steroids.
Role of the Cytoskeleton in Mediating cAMP-dependent Protein Kinase Inhibition of the Epithelial Na+/H+ Exchanger NHE3*
TLDR
It is shown that inhibition of NHE3 activity by forskolin, an activator of adenylate cyclase, occurs without changes in surface expression of the exchanger but is associated with altered cytoskeletal structure, suggesting that reorganization of the cytoskeleton by PKA is involved in mediating inhibition ofNHE3.
The recycling regulation of sodium-hydrogen exchanger isoform 3(NHE3) in epithelial cells
TLDR
This review describes the molecular steps and protein interactions involved in the recycling movement of NHE3 from the apex of epithelial cells, into vesicles, where it is reprocessed and reassembled, and returned to its original location on the plasma membrane,Where it exerts its physiological function.
Chloride Channel (Clc)-5 Is Necessary for Exocytic Trafficking of Na+/H+ Exchanger 3 (NHE3)*
TLDR
The results show that ClC-5 plays an essential role in exocytosis of NHE3, and that the rates of basal and dexamethasone-stimulated exocyTosis ofNHE3 were attenuated in Clc-5 knockdown cells.
Dopamine Acutely Stimulates Na+/H+Exchanger (NHE3) Endocytosis via Clathrin-coated Vesicles
TLDR
One mechanism by which DA induces natriuresis is via protein kinase A-mediated phosphorylation of proximal tubule N HE3 leading to endocytosis of NHE3 via clathrin-coated vesicles.
Functional coupling of the downregulated in adenoma Cl-/base exchanger DRA and the apical Na+/H+ exchangers NHE2 and NHE3.
TLDR
The role of DRA is supported in electroneutral NaCl absorption involving functional coupling of Cl(-)/base exchange and apical NHE in intestinal epithelial cells.

Endosomal Recycling of the Na+/H+Exchanger NHE3 Isoform Is Regulated by the Phosphatidylinositol 3-Kinase Pathway*
TLDR
Observations demonstrate that NHE3 activity is controlled by dynamic endocytic and recycling events that are modulated by PI3-K.
Differential localization of Na+/H+ exchanger isoforms (NHE1 and NHE3) in polarized epithelial cell lines.
TLDR
The specific steady state expression of the two NHE isoforms in polarized epithelial cells in culture is investigated and it is concluded that the epithelial and more specialized NHE3 isoform is exclusively restricted to the apical side of epithelial Cells.
Polarized distribution of Na+/H+ exchanger isoforms in rabbit collecting duct cells.
TLDR
The results suggest that RC.SV3 cells have two Na+/H+ exchange activities separated spatially to the two cellular membranes, with the N HE-1 and the NHE-2 isoforms located on the basolateral and the apical membranes, respectively.
Na+/H+ exchangers, NHE-1 and NHE-3, of rat intestine. Expression and localization.
TLDR
Analysis of NHE-3 mRNA distribution in the gut by in situ hybridization demonstrated epithelial cell specificity, as well as expression preferential to villus cells, suggesting that unlike NHE/NHE-1, which probably serves a "housekeeping" function, N HE-3 may be involved in vectorial Na transport by the intestine.
Expression of NHE-3 in the apical membrane of rat renal proximal tubule and thick ascending limb.
TLDR
NHE-3 is the isoform responsible for NaCl and NaHCO3 absorption in the proximal convoluted tubule, and NahCO absorption in a thick ascending limb and apical membrane Na/H exchange activity is likely mediated by other isoform of the NHE family.
Rab17 Regulates Membrane Trafficking through Apical Recycling Endosomes in Polarized Epithelial Cells
TLDR
A role for rab17 in regulating traffic through the apical recycling endosome is supported, suggesting a function in polarized sorting in epithelial cells.
The Activity of the Epithelial Sodium Channel Is Regulated by Clathrin-mediated Endocytosis*
TLDR
The normal mechanism of turnover of ENaC from the cell surface is demonstrated and a mechanism that can account for the increased number of channels in the plasma membrane seen in Liddle’s syndrome is demonstrated.
Clathrin and HA2 adaptors: effects of potassium depletion, hypertonic medium, and cytosol acidification
TLDR
Quantitative ultrastructural observations reveal that acidification of the cytosol results in formation of heterogeneously sized and in average smaller clathrin-coated pits at the plasma membrane and buds on the TGN, and raise the possibility that adaptor aggregates can form nucleation sites forclathrin lattices.
