Abstract We report in this Letter the synthesis of prodrugs of 2-fluoro-2-deoxyarabinose-1-phosphate and 2,2-difluoro-2-deoxyribose-1-phosphate. We demonstrate the difficulty of realising a phosphorylation step on the anomeric position of 2-deoxyribose, and we discover that introduction of fluorine atoms on the 2 position of 2-deoxyribose enables the phosphorylation step: in fact, the stability of the prodrugs increases with the degree of 2-fluorination. Stability studies of produgs of 2…
Synthesis and biological evaluation of phosphoramidate prodrugs of two analogues of 2-deoxy-d-ribose-1-phosphate directed to the discovery of two carbasugars as new potential anti-HIV leads

Chemo-enzymatic synthesis of α-d-pentofuranose-1-phosphates using thermostable pyrimidine nucleoside phosphorylases
Abstract α- d -pentofuranose-1-phosphates (Pentose-1Ps) are key intermediates in nucleoside metabolism and important precursors for the enzymatic synthesis of modified nucleosides. To date,

The synthesis and crystal structure of a new mononuclear zinc(II) complex derived from N,N'-bis(5-fluorosalicylidene)-1,2-diaminoethane (H2L) are described. The infrared spectrum and thermal analysis
Targeting GNE myopathy: A dual prodrug approach for the delivery of N-acetyl mannosamine 6-phosphate.
Prodrug 13b showed rapid activation in a carboxylesterase enzymatic assay and favourable ADME properties, while also being more effective at increasing sialic acid levels in GNE-deficient cell lines than ManNAc, providing a potential platform to address substrate deficiencies in G NE myopathy and other CDGs.
Phosphoramidates and phosphonamidates (ProTides) with antiviral activity
In this review, the seminal contribution of Chris McGuigan’s research to this field is presented and his technology proved to be extremely successful in drug discovery and has led to two Food and Drug Administration-approved antiviral agents.
New biocatalysts for one pot multistep enzymatic synthesis of pyrimidine nucleoside diphosphates from readily available reagents
Since the preparation of nucleoside 5′-diphosphates by classical methodologies is complex, multistep enzymatic systems were explored to synthesize pyrimidine nucleoside 5′-diphosphates starting from
Independent Generation and Reactivity of Thymidine Radical Cations.
Independent generation of the N3-methyl analogue of 1 (NMe-1) produces considerably higher yields of products derived from water trapping, and these products are formed in much higher yields than those attributable to the C5-methyl group deprotonation in N me-1.
An overview of ProTide technology and its implications to drug discovery
The ProTide approach has been extremely successful in delivering blockbuster antiviral medicines and it seems promising in oncology, but its application to non-nucleoside-based small molecules is recently emerging and proving effective in other therapeutic areas.
Nitrogenous Derivatives of Phosphorus and the Origins of Life: Plausible Prebiotic Phosphorylating Agents in Water
This review covers the reactions of alternative forms of phosphate, specifically the nitrogenous versions of phosphate (and other forms of reduced phosphorus species) from a prebiotic, synthetic organic and biochemistry perspective.

Stereoselective Synthesis of 2-Deoxy-2-Fluoroarabinofuranosyl-α-1-Phosphate and Its Application to the Synthesis of 2′-Deoxy-2′-Fluoroarabinofuranosyl Purine Nucleosides by a Chemo-Enzymatic Method
This chemo-enzymatic method will be applicable to the synthesis of some 2′F-araNs, and three important 2′-deoxy-2′-fluoroarabinofuranosyl purine nucleosides were actually obtained in 30–40% yields from 1,3,5-tri-O-benzoyl-2- deoxy- 2-luoro-α-D-arabinose with high purity.
Inhibition of pyrimidine and purine nucleoside phosphorylases by a 3,5-dichlorobenzoyl-substituted 2-deoxy-D-ribose-1-phosphate derivative.
The 3,5-dichlorobenzoyl-substituted 2-deoxy-D-ribose-1-phosphate derivative, designated Cf2891, was found to inhibit a variety of pyrimidine and purine nucleoside phosphorylases (NPs) with preference
Synthesis of 2-deoxy-2-fluoro analogs of polyprenyl beta-D-arabinofuranosyl phosphates.
Although the coupling yields between the phosphate and polyprenyl trichloroacetimidates were comparable with either type of protecting group, access to the benzoyl-protected derivative was more efficient and therefore gave the products in higher overall yield.
