Most of the natural schweinfurthins are potent and selective inhibitors of cell growth as measured by the National Cancer Institute's 60-cell line screen. Due to the limited supply of these natural products, we have initiated a program aimed at their synthesis. To date, this effort has led to the preparation of three natural schweinfurthins and more than 40 analogues, and assays on these compounds have afforded some understanding of structure-activity relationships in this family. Fluorescent schweinfurthin B and F analogs with anti-proliferative activity.
TLDR
The synthesis of fluorescent analogs of schweinfurthins B and F are described and it is demonstrated that these compounds retain the potent and differentially toxic activities against select human cancer cells that are characteristic of the natural schwe Infurthin.
Relevance of the C-5 position to schweinfurthin induced cytotoxicity.
TLDR
The synthesis of several new schweinfurthin analogues varied at the C-5 position are reported, along with data on their biological activity in the NCI 60 cell-line assay.
Schweinfurthins: Lipid Modulators with Promising Anticancer Activity.
TLDR
A in-depth look at the history of schweinfurthins, their synthesis, where the research presently stands, and the questions that remain is taken.
Synthesis of indole analogues of the natural schweinfurthins.
TLDR
New indole derivatives bearing a phosphonomethyl substituent in the B-ring were required and found to undergo the desired condensation with nonracemic aldehydes representing the schweinfurthin left half.
Functional Evaluation of a Fluorescent Schweinfurthin: Mechanism of Cytotoxicity and Intracellular Quantification
TLDR
It is demonstrated that incubation with 3d SB or 3dSB-PNBS results in cleavage of poly-ADP-ribose polymerase (PARP) and caspase-9, both markers of apoptosis, and Glucose-regulated protein 78 and protein disulfide isomerase, both endoplasmic reticulum chaperones, are up-regulated with schweinfurthin exposure.
Biologically active biotin derivatives of schweinfurthin F.
TLDR
These compounds maintain activity in SF-295 cells comparable to the parent system, and display the lower potency in A549 cells that is a characteristic of the schweinfurthin pattern of activity.
Pleiotropic Effects of a Schweinfurthin on Isoprenoid Homeostasis
TLDR
These studies are the first to demonstrate that a schweinfurthin exerts pleiotropic effects on isoprenoid homeostasis.
3-Deoxyschweinfurthin B Lowers Cholesterol Levels by Decreasing Synthesis and Increasing Export in Cultured Cancer Cell Lines
TLDR
The data suggest that cholesterol homeostasis plays a significant role in the growth inhibitory activity of the schweinfurthins and may elucidate a mechanism that can be targeted in human cancers such as GBM.
First total synthesis of (+)-Vedelianin, a potent antiproliferative agent.
TLDR
Preparation of a key intermediate in nonracemic form through a Shi epoxidation has allowed determination of the absolute stereochemistry of the natural product as the (2S, 3R, 4aR, 9aR)-isomer.
Stilbenes as κ-Selective, Non-nitrogenous Opioid Receptor Antagonists
TLDR
Assays on two compounds determined that one shows potent opioid receptor binding activity and significantly improved selectivity for the κ receptor, and these studies begin to illuminate the structural features of these non-nitrogenous opioid receptor antagonists that are required for activity.

Synthesis and structure-activity studies of schweinfurthin B analogs: Evidence for the importance of a D-ring hydrogen bond donor in expression of differential cytotoxicity.
TLDR
Together, these assay results indicate the importance of at least one free phenol group on the aromatic D-ring of this system for differential cytotoxicity.
Cytotoxic geranyl stilbenes from Macaranga schweinfurthii.
TLDR
The cytot toxicity profile of the schweinfurthins tested in the NCI 60-cell screen was similar to that of the stelletins and cephalostatins, suggesting that these structurally diverse natural products may share similar mechanisms of cytotoxicity.
Plants as a source of anti-cancer agents.
TLDR
A number of promising new agents are in clinical development based on selective activity against cancer-related molecular targets, including flavopiridol and combretastin A4 phosphate, while some agents which failed in earlier clinical studies are stimulating renewed interest.
Synthesis of nonracemic 3-deoxyschweinfurthin B.
TLDR
This represents the first synthesis of the tetracyclic schweinfurthin skeleton, validating the overall synthetic strategy and providing the first schwe infurthin analogue with activity slightly greater than those of the natural products.
Polyproline-rod approach to isolating protein targets of bioactive small molecules: isolation of a new target of indomethacin.
TLDR
The results indicate that insertion of a long, rigid polyproline helix between a small-molecule bait and a biotin tag boosts the capacity of affinity purification and thereby permits isolation of low-abundance or low-affinity proteins.
Antiproliferative prenylated stilbenes and flavonoids from Macaranga alnifolia from the Madagascar rainforest.
Bioassay-guided fractionation of an extract of the fruit of Macaranga alnifolia from Madagascar led to the isolation of four new prenylated stilbenes, schweinfurthins E-H (1-4), and one new
Schweinfurthin D, A Cytotoxic Stilbene from Macaranga schweinfurthii
Abstract A novel cytotoxic stilbene, schweinfurthin D, was isolated from Macaranga schweinfurthii (Euphorbiaceae) and its structure was determined by spectroscopic methods. Schweinfurthin D showed
BF3 x Et2O-mediated cascade cyclizations: synthesis of schweinfurthins F and G.
TLDR
The total synthesis of the natural stilbene (+)-schweinfurthin G has been accomplished through a sequence based on an efficient cationic cascade cyclization through a novel reaction with a phenolic oxygen "protected" as its MOM ether.

Roscovitine Targets, Protein Kinases and Pyridoxal Kinase*[boxs]
TLDR
The results show that (R)-roscovitine is rather selective for CDKs, in fact most kinases are not affected, but it binds an unexpected, non-protein kinase target, pyridoxal kinase, the enzyme responsible for phosphorylation and activation of vitamin B6.
