Dog albumin contains a specific drug-binding site that binds most of the site II ligands of human albumin. This study was undertaken to elucidate the structural configuration of this binding site using a photoaffinity labeling technique. Dog albumin and albumins of other animal species were photolabeled with [14C]ketoprofen. The photolabeled albumins were cleaved with cyanogen bromide (CNBr) and analyzed autoradiographically after electrophoretic separation. A 11.6-kDa CNBr fragment of the…

Histidine146 of human serum albumin plays a prominent role at the interface of subdomains IA and IIA in allosteric ligand binding
TLDR
A similar experiment using H146I showed that a benzene ring‐like structure at position 146 is required for the allosteric ligand binding to occur, indicating that, when myristate binds to HSA, a new binding site in formed in that region.
Photoaffinity Labeling of Plasma Proteins
TLDR
The photolabeling of AGP with flunitrazepam has been utilized to shed light on the topology of the protein ligand binding site and a review of photoaffinity labeling performed on other major plasma proteins will also be discussed.

TLDR
It is shown that the electron-density distribution in the aromatic ring exerts crucial influence on the albumin affinity.
Updates on contemporary protein binding techniques.
TLDR
A new high-throughput assay based on the distribution of a drug among plasma water, plasma proteins, and solid-supported lipid membranes has been reported to produce valid results, even for drugs that are strongly bound to plasma proteins.

TLDR
The structure, function, and pharmaceutical properties of human serum albumin and alpha(1)-acid glycoprotein play important roles in protein binding for many drugs, which is of key importance to drug distribution in the body.
A comparative analysis on the binding characteristics of various mammalian albumins towards a multitherapeutic agent, pinostrobin
TLDR
Albumins from goat and bovine were found to be closely similar to human albumin on the basis of PS binding characteristics, and were grouped into two classes based on the PS–albumin binding characteristics.
Multimethodological study of molecular recognition phenomena
TLDR
High-performance liquid affinity chromatography, circular dichroism and optical biosensor represent three techniques that can be able to elucidate the interaction of a new drug with its target and with others proteins that could interfere with ADMET parameters.

TLDR
It is proposed herein that mammalian serum albumins used in this study contain specific drug binding sites: Rabbit and rat albumins contain a drug binding site, corresponding to site I on human albumin, and dog albumin contains a specific drugbinding site corresponding tosite II on the human album in molecule.

TLDR
The amino-acid sequence and three-dimensional structure of equine serum albumin have been determined and two binding sites for 2,3,5-triiodobenzoic acid identical with those previously reported for human serumalbumin which are located within subdomains in IIA and IIIA are discussed.
Crystal structure of human serum albumin complexed with fatty acid reveals an asymmetric distribution of binding sites
TLDR
The crystal structure of HSA complexed with five molecules of myristate at 2.5 Å resolution is determined and it is shown that fatty acid molecules bind in long, hydrophobic pockets capped by polar side chains, many of which are basic.
