Arteriviruses are enveloped, positive-stranded RNA viruses and include pathogens of major economic concern to the swine- and horse-breeding industries. The arterivirus replicase gene encodes two large precursor polyproteins that are processed by the viral main proteinase nonstructural protein 4 (nsp4). The three-dimensional structure of the 21-kDa nsp4 from the arterivirus prototype equine arteritis virus has been determined to 2.0 Å resolution. Nsp4 adopts the smallest known chymotrypsin-like… Expand
The structural biology of PRRSV
TLDR
Cryo-electron tomographic studies have shown the three-dimensional organization of the PRRSV virion and suggest that the viral nucleocapsid has an asymmetric, linear arrangement, rather than the isometric core previously described, which reveals a closer structural relationship between arteri- and coronaviruses than previously anticipated. Expand
Astrovirus Replication : An Overview
Human astroviruses are important pathogens that cause gastroenteritis worldwide. Significant progress has recently been made regarding the characterization of the RNA replication process, theExpand
Astrovirus Replication: An Overview
TLDR
It has been demonstrated that apoptotic host cell death seems necessary for maturation of astrovirus particles, and it has been predicted that the VP34 capsid protein contains the RNA binding domain at its N-terminus, responsible for packaging the viral genome. Expand
Characterization of a Torovirus Main Proteinase
TLDR
The characterization of the Mpro of toroviruses, the fourth main Nidovirus branch, is reported, which has a predicted three-domain structure, with two catalyticβ -barrel domains and an additional C-terminal domain of unknown function. Expand
Structure and Cleavage Specificity of the Chymotrypsin-Like Serine Protease (3CLSP/nsp4) of Porcine Reproductive and Respiratory Syndrome Virus (PRRSV)
TLDR
Structural evidence as well as enzymatic proof of the nsp4 protein being a functional 3CLSP are provided, which shows that the enzyme has a strong preference for glutamic acid at the P1 position of the substrate. Expand
Proteolytic maturation of replicase polyprotein pp1a by the nsp4 main proteinase is essential for equine arteritis virus replication and includes internal cleavage of nsp7.
The positive-stranded RNA genome of the arterivirus Equine arteritis virus (order Nidovirales) encodes the partially overlapping replicase polyproteins pp1a (1727 aa) and pp1ab (3175 aa). Previously,Expand
Expression, purification, and in vitro activity of an arterivirus main proteinase
TLDR
Protocols for the large-scale production of recombinant nsp4 in Escherichia coli were developed and both forms of the EAV proteinase were shown to be proteolytically active in two different trans-cleavage assays, including studies aiming to identify inhibitors of proteolytic activity. Expand
Variability of non-structural proteins of equine arteritis virus during persistent infection of the stallion.
The genetic stability of ORF1a encoding non-structural proteins nsp1, nsp2, nsp3 and nsp4 of equine arteritis virus (EAV) has been analysed for nearly seven years in a persistently infected stallionExpand
The PRRSV replicase: Exploring the multifunctionality of an intriguing set of nonstructural proteins
TLDR
The continued characterization of the nsps of the arterivirus prototype equine arteritis virus has not only corroborated several important functional predictions, but also revealed various novel features of arteriviral replication. Expand
The 3C-Like Proteinase of an Invertebrate Nidovirus Links Coronavirus and Potyvirus Homologs
TLDR
The study revealed that the GAV and potyvirus 3CLpros possess similar substrate specificities which correlate with structural similarities in their respective substrate-binding sites, identified in sequence comparisons. Expand

The Arterivirus Nsp4 Protease Is the Prototype of a Novel Group of Chymotrypsin-like Enzymes, the 3C-like Serine Proteases (*)
TLDR
The results support a replicase processing model in which the SP cleaves multiple Glu(Gly/Ser/Ala) sites, and characterize the arterivirus SP as a representative of a novel group of chymotrypsin-like enzymes, the 3C-like serine proteases. Expand
Proteolytic processing of the replicase ORF1a protein of equine arteritis virus
To study the proteolytic processing of the equine arteritis virus (EAV) replicase open reading frame 1a (ORF1a) protein, specific antisera were raised in rabbits, with six synthetic peptides and aExpand
Alternative proteolytic processing of the arterivirus replicase ORF1a polyprotein: evidence that NSP2 acts as a cofactor for the NSP4 serine protease
TLDR
The studies revealed that the C-terminal half of the ORF1a protein (nsp3-8) can be processed by the SP following two alternative pathways, which appear to be mutually exclusive. Expand
Structure of coronavirus main proteinase reveals combination of a chymotrypsin fold with an extra α-helical domain
TLDR
The crystal structure of the 33.1 kDa transmissible gastroenteritis (corona)virus Mpro is reported and illustrates that RNA viruses have evolved unprecedented variations of the classical chymotrypsin fold. Expand
Structure of Semliki Forest virus core protein
TLDR
The conformation of Tyr184, found in SFCP, is consistent with its availability for iodination, as observed in the homologous SINV cores, which suggests, by comparison with SCP, that E2 binding to cores causes major conformational changes, including the burial of Tyr 184, which would stabilize the intact virus on budding from an infected cell. Picornaviral 3C cysteine proteinases have a fold similar to chymotrypsin-like serine proteinases
TLDR
The overall architecture of HAV-3C reveals a fold resembling that of the chymotrypsin family of serine proteinases, which is consistent with earlier predictions7,8. Expand
Equine arteritis virus is not a togavirus but belongs to the coronaviruslike superfamily
TLDR
The data indicate that EAV, presently considered a togavirus, is evolutionarily related to viruses from the coronaviruslike superfamily. Expand
Proteolytic Processing of the Open Reading Frame 1b-Encoded Part of Arterivirus Replicase Is Mediated by nsp4 Serine Protease and Is Essential for Virus Replication
