When their growth is arrested in culture, susceptible 3T3 fibroblasts differentiate into adipose cells. Different clones form adipose cells with different frequency, depending upon the proportion of susceptible cells they contain. In cultures grown from small inocula, the fat cells appear in clusters formed by colonies of susceptible cells. Study of these clusters indicates the infrequent occurrence of cellular transitions from insusceptible to susceptible state. Beginning with a clone… Expand

A clonal analysis of the differentiation of 3T3‐L1 preadipose cells: Role of insulin
TLDR
In undisturbed cultures, the distribution of fat cells is not random and is consistent with a commitment event in single cells at any cell division during exponential growth followed by divisions of both committed and uncommitted cells. Expand
Growth and Differentiation of Preadipocyte Cell Lines in Serum-Free Medium
TLDR
Obesity arises from an abnormal growth and differentiation of adipose cells, and the identification of the replicating preadipocyte or differentiated adipocyte has yet to be accomplished. Expand
Differentiation of cultured pre‐adipose cells: A probability model
TLDR
Both the observed range of clone sizes and pattern of clones can be approximated by a simple stochastic model, suggesting that commitment to fat production in 3T3‐L1 cells is a random process occurring with a fixed probability in single cells in exponential growth, followed by division of both committed and uncommitted cells. Expand
Establishment of a clonal cell line that differentiates into adipose cells in vitro
TLDR
A clonal cell line that could be induced to differentiate into fat-accumulating cells in vitro and can be used as a model for mammalian cell differentiation and also as a convenient material for the study of lipid metabolism in adipocytes. Expand
Adipose conversion of OB17 preadipocytes. Relationships between cell division and fat cell cluster formation.
TLDR
It is shown that the formation of fat cell clusters of large size is due to mitoses of susceptible cells during a limited period of time after confluence, and post-confluent mitoses are not necessary for the differentiation to occur, but rather they serve to amplify the proportion of adipose cells relative to non-adipose cells. Expand
Complete differentiation of adipocyte precursors
TLDR
There is firm evidence that fat tissue of adult humans and rats contains adipocyte precursors that differentiate into mature fat cells, and the culture system described will facilitate the elucidation of the factors involved in replication and differentiation of adipocytes in vitro. Expand
Cell and tissue culture models of adipocyte development
TLDR
Despite the aneuploid chromosome status, preneoplastic nature, and occasional deviation from the patterns seen with adipose tissue-derived adipocytes, 3T3 and ST 13 preadipose systems supply valuable information that help to investigate the biochemical machinery for lipid storage and mobilization and how these develop during adipose conversion. Expand
Hormonal responsiveness of a preadipose cell line derived from newborn mouse calvaria
TLDR
Results strongly suggest that the clonal preadispose cell line, designated MC3T3‐G2/PA6, was derived from bone marrow, and indicate that the hormone accelerates the initiation of the adipose conversion, but is not required for the ongoing conversion process. Expand
Establishment and characterization of fibroblast-like cell lines derived from adipocytes with the capacity to redifferentiate into adipocyte-like cells.
TLDR
This chapter describes the isolation of cell lines from the adipocyte fraction of epididymal fat pads of C57 BL/6J genetically obese and nonobese mice, which offers the possibility to study morphological and biochemical events occurring during adipose conversion at both cellular and molecular levels. Expand
Adipose tissue development: The role of precursor cells and adipogenic factors
TLDR
Cell culture systems have proven to be valuable models for the study of the processes involved in the formation of new fat cells, and the ability to undergo adipose conversion is not stable in these cells, but gets lost after repeated subcultures or seeding at low densities. Expand

An established pre-adipose cell line and its differentiation in culture.
TLDR
The conversion of 3T3-L1 from pre-adipose to adipose cells therefore involves a process of differentiation which can be studied under cell culture conditions. Expand
An established preadipose cell line and its differentiation in culture II. Factors affecting the adipose conversion
TLDR
Lipogenic and lipolytic hormones and drugs affect the rate of synthesis and accumulation of triglyceride by 3T3-L1 cells, but in contrast to bromodeoxyuridine, these modulating agents do not seem to affect the proportion of cells which undergoes the adipose conversion. Expand
QUANTITATIVE STUDIES OF THE GROWTH OF MOUSE EMBRYO CELLS IN CULTURE AND THEIR DEVELOPMENT INTO ESTABLISHED LINES
TLDR
Disaggregated mouse embryo cells, grown in monolayers, underwent a progressive decline in growth rate upon successive transfer, the rapidity of the decline depending on the inoculation density, but nearly all cultures developed into established lines within 3 months of culture. Expand
The initiation of cell division in a contact-inhibited mammalian cell line.
TLDR
The sequence of event suggests that regulation of RNA synthesis is the means by which contact inhibition controls cell division, and therefore DNA synthesis and cell division are controlled. Expand
The production of hyaluronate by spontaneously established cell lines and viral transformed lines of fibroblastic origin.
Human diploid fibroblast strains and established mouse fibroblast lines continue to synthesize hyaluronate over many cell generations in culture. However, this property may be lost in the course ofExpand
Shaking 3T3 cells: further studies on diffusion boundary effects.
TLDR
Growth in confluent 3T3 cell cultures is limited by a diffusion barrier in the microenvironment, probably affecting uptake of a serum component, and can be overcome by increased fluid velocity as an alternative to raising the serum concentration. Expand
Sublines of mouse 3T3 cells that accumulate lipid
TLDR
Two clonal sublines are isolated from the established mouse fibroblast line 3T3 that accumulate large amounts of triglyceride fat when the cells are in the resting state and are reduced by lipolytic agents. Expand

TLDR
Cells of the mouse line Balb/3T3 as well as three virus-induced transformants and two spontaneous transformants grown in vitro have been studied for their topography by scanning electron microscopy and the significance of the surface morphologies and the factors influencing cell form are discussed.
