A pool of stem cells that arise from the mesoderm during embryogenesis initiates hematopoiesis. However, factors that regulate the expansion of blood stem cells are poorly understood. We show here that cytokine-induced proliferation of primitive human hematopoietic cells could be inhibited with antibodies to hedgehog (Hh). Conversely, Sonic hedgehog (Shh) treatment induced the expansion of pluripotent human hematopoietic repopulating cells detected in immunodeficient mice. Noggin, a specific… Expand
Gli3-mediated hedgehog inhibition in human pluripotent stem cells initiates and augments developmental programming of adult hematopoiesis.
TLDR
An unprecedented role for Hh signaling is revealed in the regulation of adult hematopoietic specification, thereby demonstrating the ability to modulate the default embryonic programs of hPSCs. Expand
Role of sonic hedgehog in maintaining a pool of proliferating stem cells in the human fetal epidermis.
TLDR
Shh is an important regulator of HPESC proliferation in the basal layer of human fetal epidermis and modulates the cell responsiveness to EGF, which will assist to unravel the mechanisms that regulate stem cell proliferation and neoplasia in the humanEpidermis. Expand
Hedgehog modulates cell cycle regulators in stem cells to control hematopoietic regeneration
TLDR
This study demonstrates that downstream activation of the Hedgehog (Hh) signaling pathway induces cycling and expansion of primitive bone marrow hematopoietic cells under homeostatic conditions and during acute regeneration. Expand
Deterministic role of sonic hedgehog signalling pathway in specification of hemogenic versus endocardiogenic endothelium from differentiated human embryonic stem cells.
TLDR
The previously unreported deterministic role of SHH pathway is revealed in specification of endothelial cells differentiated from human ESCs into hemogenic vs. endocardiogenic lineage; this finding could have major implications for clinical applications. Expand
Cytokines and BMP-4 promote hematopoietic differentiation of human embryonic stem cells.
TLDR
This study establishes the first evidence for the role of cytokines and BMP-4 in promoting hematopoietic differentiation of hESC lines and provides an unprecedented system to study early developmental events that govern the initiation of hematoiesis in the human. Expand

TLDR
Understanding the individual and collective contributions of Notch, Wnt and HH signaling to the initial development of hematopoiesis is critical for achieving successful ex vivo expansion and differentiation of heMatopoietic stem cells (HSCs) from human PSCs that will retain bona fide function comparable to adult-derived HSCs. Expand
Reduced Thymocyte Development in Sonic Hedgehog Knockout Embryos 1
TLDR
Genetic evidence that Sonic Hedgehog (Shh) has a role in the control of murine T cell development is provided and it is revealed that Shh regulates fetal thymus cellularity and thymocyte differentiation. Expand
Fetal liver stromal cells promote hematopoietic cell expansion.
TLDR
These findings suggest that FL-derived StroCs support proliferation of HSPCs via Shh inducing an autocrine Wnt signaling loop and the use of FL- derived StroCs and regulation of the Shh pathway might further enhance HPSC expansion. Expand
Hedgehog and Bmp polarize hematopoietic stem cell emergence in the zebrafish dorsal aorta.
TLDR
It is shown that while continued Hedgehog signaling from the overlying midline structures maintains the arterial program characteristic of the DA roof, a ventral Bmp4 signal induces the blood stem cell program in the DA floor. Expand
Sonic Hedgehog Regulates Early Human Thymocyte Differentiation by Counteracting the IL-7-Induced Development of CD34+ Precursor Cells1
TLDR
Results show that Shh may function as a maintenance factor for intrathymic CD34+ precursor cells that are committing to the T cell lineage and down-regulated in the precursor cell population the expression of IL-7R as well as stromal-derived factor-1 chemokine receptor, CXCR4, and inhibited IL- 7-dependent STAT5 phosphorylation. Bone Morphogenetic Proteins Regulate the Developmental Program of Human Hematopoietic Stem Cells
TLDR
It is reported that BMPs regulate the proliferation and differentiation of highly purified primitive human hematopoietic cells from adult and neonatal sources and a powerful model system for studying the biological mechanism of BMP action using primary human cells. Expand
Sonic hedgehog promotes somitic chondrogenesis by altering the cellular response to BMP signaling.
TLDR
It is shown that presomitic mesoderm cultured in the presence of Shh will differentiate into cartilage, and that the later stages of this differentiation process specifically depend on bone morphogenetic protein (BMP) signaling. Expand
Actions of hedgehog proteins on skeletal cells.

Hedgehog proteins seem to be significantly involved in skeletal formation through multiple actions on chondrogenic mesenchymal cells, chondrocytes, and osteogenic cells. Expand

The role of BMP-4 and GATA-2 in the induction and differentiation of hematopoietic mesoderm in Xenopus laevis.

Hedgehog signaling regulates differentiation from double-negative to double-positive thymocyte.
TLDR
The data suggest that termination of Hh signaling is necessary for differentiation from CD4-CD8-double-negative (DN) to CD4+CD8+ double-positive (DP) thymocyte, and that one consequence of pre-TCR signaling is downregulation of Smo, allowing DN thymocytes to proliferate and differentiate. Expand
Sonic hedgehog regulates patterning in early embryos.

By misexpression of Sonic hedgehog, it is shown that sonic hedgehog induces expression of Hoxd genes, known to be involved in patterning of the anterior-posterior axis of the limb, and Bmp-2, which might act as a secondary signal. Expand
Sonic hedgehog is an endodermal signal inducing Bmp-4 and Hox genes during induction and regionalization of the chick hindgut.
TLDR
Using virally mediated misexpression, it is shown that Sonic hedgehog is sufficient to induce ectopic expression of Bmp-4 and specific Hoxd genes within the mesoderm, which appears to act as a signal in an epithelial-mesenchymal interaction in the earliest stages of chick hindgut formation. Expand
