Background: Silk gland factor-2 (SGF-2) is a key factor regulating tissue-specific expression of the fibroin gene. Results: SGF-2 is a 1.1-MDa heteromeric complex containing Awh, Ldb, Lcaf, and fibrohexamerin proteins. Conclusion: Awh, Ldb, and Lcaf interact functionally in SGF-2 to control fibroin gene expression. Significance: This study provides new insight into the functional role of single-stranded DNA-binding proteins in protein-protein interaction and transcriptional regulation. SGF-2…

Genome-wide open chromatin regions and their effects on the regulation of silk protein genes in Bombyx mori

To investigate the regulation of silk-protein genes, DNA molecules isolated from the silk glands of third-day fifth-instar silkworm larvae and embryo-derived cells were subjected to formal dehyde-assisted isolation of regulatory elements (FAIRE) and high-throughput sequencing.

New insights into the proteins interacting with the promoters of silkworm fibroin genes

This study provides the first in-depth profile of proteins interacting with fibroin gene promoters and contributes to a better understanding of silk protein synthesis regulation.

A Juvenile Hormone Transcription Factor Bmdimm-Fibroin H Chain Pathway Is Involved in the Synthesis of Silk Protein in Silkworm, Bombyx mori*

The data suggest that the JH regulatory pathway, the transcription factor Bmdimm, and the targeted fib-H gene contribute to the synthesis of fibroin H-chain protein in B. mori.

Multiprotein bridging factor 2 regulates the expression of the fibroin heavy chain gene by interacting with Bmdimmed in the silkworm Bombyx mori

Findings suggest that MBF2 is a transcriptional repressor that is involved in controlling the regulation of the fibH gene in the posterior silk gland by interacting with Bmdimm.

Basic Helix-Loop-Helix Transcription Factor Bmsage Is Involved in Regulation of fibroin H-chain Gene via Interaction with SGF1 in Bombyx mori

Results suggest Bmsage is involved in the regulation of the expression of fib-H by being together with SGF1 in B. mori PSG cells.

A Hox Gene, Antennapedia, Regulates Expression of Multiple Major Silk Protein Genes in the Silkworm Bombyx mori*

It is suggested that Hox genes have a role as a master control in terminally differentiated tissues, possibly acting as a primary regulator for a range of physiological processes.

Silk Gland Factor-1 Involved in the Regulation of Bombyx Sericin-1 Gene Contains Fork Head Motif (*)

It is concluded that the 40- and 41-kDa SGF-1 proteins affect transcription of the sericin-1 gene via binding to the SA site.

Transcriptional regulatory elements in the upstream and intron of the fibroin gene bind three specific factors POU-M1, Bm Fkh and FMBP-1.

The results suggest that expression of the fibroin gene is controlled by co-ordination of these factors with distinct specificities during silk-gland development.

Single-stranded DNA-binding proteins regulate the abundance of LIM domain and LIM domain-binding proteins.

It is established here that two mammalian SSBPs, SSBP2 and SSBP3, contribute to an erythroid DNA-binding complex that contains the transcription factors Tal1 and GATA-1, the LIM domain protein Lmo2, and Ldb1 and binds a bipartite E-box-GATA DNA sequence motif.

Fibroin-modulator-binding protein-1 (FMBP-1) contains a novel DNA-binding domain, repeats of the score and three amino acid peptide (STP), conserved from Caenorhabditis elegans to humans

Mutational analysis of FMBP-1 showed that the STP repeats form a novel DNA-binding domain, and Sequences flanking STP repeat modulated DNA- binding activity.

Ssdp proteins interact with the LIM-domain-binding protein Ldb1 to regulate development

Functional conservation of Ssdp as a cofactor of Ldb1 during invertebrate and vertebrate development is indicated.

Ssdp proteins bind to LIM-interacting co-factors and regulate the activity of LIM-homeodomain protein complexes in vivo

The identification and characterization of members of the Ssdp family of proteins are described, which are shown to interact with Ldb1 and Chip and show structural similarity to two Arabidopsis proteins that cooperate with one another to regulate gene expression during flower development.

A family of LIM domain-associated cofactors confer transcriptional synergism between LIM and Otx homeodomain proteins.

This work isolated murine cDNAs encoding two highly homologous proteins that specifically interact with the LIM domains of P-Lim/Lhx3 and several other LIM homeodomain factors and link them to members of the Otx class of transcription factors in gene activation events during embryogenesis via the actions of specific cofactors.

Interactions of the LIM-domain-binding factor Ldbl with LIM homeodomain proteins

A new LIM-domain-binding factor is isolated on the basis of its ability to interact with the LIM-HD protein Lhxl (Liml) and high-affinity binding by Ldbl requires paired LIM domains and is restricted to the related subgroup of LIM domains found in LIM- HD and LMO proteins.
