&NA; To explore possible concentration‐effect relationships, gabapentin (GBP) and vigabatrin (VGB) serum concentrations were obtained from patients participating in an add‐on dose‐titration trial comparing GBP and VGB in partial epilepsy. Patients randomized to GBP started on 1800 mg/d and could have their dosage increased stepwise to 2400 and 3600 mg/d if seizures persisted. Those randomised to VGB started on 1000 mg/d, and the dose could be increased to 2000 and 4000 mg/d. Blood samples were… Expand
An acute gabapentin fatality: a case report with postmortem concentrations
TLDR
A fatality that appears to be associated with an isolated and acute GBP ingestion was presented, and screening for volatiles, drugs of abuse, alkaline compounds, and acid/neutral compounds was negative. Expand
A microdialysis study of oral vigabatrin administration in head injury patients: preliminary evaluation of multimodality monitoring.
TLDR
Multimodality monitoring, including cerebral microdialysis, is feasible for studying surrogate endpoints following drug administration in severe head injury patients, and microdialysate glucose and glycerol sometimes decreased, and glutamate and tyrosine sometimes increased, following VBG administration. Expand
Gabapentin concentrations and postmortem distribution.
TLDR
Gabapentin analyses were performed on 30 postmortem cases that had peripheral blood, central blood and liver tissue and suggest that gabapentin is unlikely to show postmortem redistribution. Expand
Cardiopulmonary bypass does not affect plasma concentration of preoperatively administered gabapentin
TLDR
Drug accumulation following third and fourth postoperative doses suggests the need for therapeutic drug monitoring in future trials, and Gabapentin is well established as an effective adjunct analgesic in a number of surgical settings. Expand
Performance Characteristics of the ARK Diagnostics Gabapentin Immunoassay
TLDR
The ARK immunoassay is suitable for the clinical use of monitoring gabapentin in serum or plasma on the Beckman AU400e. Expand
Therapeutic Drug Monitoring of Antiepileptic Medications
TLDR
This chapter focuses on therapeutic drug monitoring of AEMs in treatment of epilepsy, emphasizing whether the pharmacokinetics and clinical profile of the drug make TDM useful. Expand
Pharmacokinetic Variability of Newer Antiepileptic Drugs
TLDR
Therapeutic drug monitoring (TDM) may be useful despite the shortcomings of existing therapeutic ranges, by utilisation of the concept of ‘individual reference concentrations’ based on intra-individual comparisons of drug serum concentrations. Expand
An Updated Overview on Therapeutic Drug Monitoring of Recent Antiepileptic Drugs
Given the distinctive characteristics of both epilepsy and antiepileptic drugs (AEDs), therapeutic drug monitoring (TDM) can make a significant contribution to the field of epilepsy. The measurementExpand
Vigabatrin Lacks Proarrhythmic Potential: Results from a Thorough QT/QTc Study in Healthy Volunteers.
TLDR
The findings from this thorough QT study are consistent with existing clinical data and confirm a lack of proarrhythmic potential of vigabatrin. Expand
Monitoring vigabatrin in head injury patients by cerebral microdialysis: obtaining pharmacokinetic measurements in a neurocritical care setting
TLDR
Vigabatrin, given enterally to severe TBI patients, crosses the blood–brain barrier into the brain extracellular fluid, where it accumulates with multiple dosing, and Pharmacokinetics suggest delayed uptake from the blood. Expand

Changes in plasma GABA concentration during vigabatrin treatment of epilepsy: a prospective study
TLDR
Although plasma GABA increased significantly in the responder but not in the nonresponder group during vigabatrin treatment of patients with epilepsy, it does not seem to be a reliable marker of individual clinical response to vigab atrin treatment. Expand
Double‐Blind Study of Gabapentin in the Treatment of Partial Seizures
TLDR
GBP appears to be effective in the treatment of partial epileptic seizures in a dosage‐related manner in a double‐blind, placebo‐controlled study. Expand
Cerebrospinal fluid GABA and seizure control with vigabatrin.
TLDR
GVG is an effective anticonvulsant drug in complex partial seizures and in nonresponders the poor anticonVulsant response may be related to the lower elevation of the CSF markers of GABAergic neuronal activity in this group compared with the responders. Expand
γ‐Vinyl GABA (Vigabatrin): Relationship Between Dosage, Plasma Concentrations, Platelet GABA‐Transaminase Inhibition, and Seizure Reduction in Epileptic Children
Summary: The relationship between vigabatrin γ‐vinyl GABA (GVG, vigabatrin) daily dosage or steady‐state plasma concentrations (CSS), platelet GABA‐transaminase (GABA‐T) inhibition, and seizureExpand
Vigabatrin serum concentration to dosage ratio: influence of age and associated antiepileptic drugs.
TLDR
It was concluded that trough steady state VGB serum concentration may be more predictable in children based on the milligrams per kilogram dosage than on themilligram dosage, and that the influence of patient age should be considered if the VGB C/D ratio is used to estimate patient compliance. Expand
High dose gabapentin in refractory partial epilepsy: clinical observations in 50 patients
TLDR
The results do not support the suggestion that GBP absorption is saturable, but high dose GBP may be effective in controlling seizures in patients with refractory partial epilepsy. Expand
Population pharmacokinetics of gabapentin in infants and children
TLDR
Gabapentin oral clearance was directly related to creatinine clearance and the slope of the relationship was 36% greater in blacks than in subjects of other races, and volume of distribution appeared to differ between subjects and patients. Expand
Inter- and intra-subject variability in gabapentin absorption and absolute bioavailability
TLDR
The within subject variability of gabapentin is small enough that plasma drug monitoring may be used to assess gABapentin absorption for a given subject and the benefit of dose individualization. Expand
Determination of GABA and vigabatrin in human plasma by a rapid and simple HPLC method: correlation between clinical response to vigabatrin and increase in plasma GABA
TLDR
A rapid, selective and sensitive amino acid analysis HPLC method has been developed for plasma GABA determination with fluorescence detection, using o-phthaldialdehyde as a precolumn derivatizing agent, and data may indicate that determination of plasma GABA is a valuable non-invasive method for therapeutic monitoring in patients on medication with vigabatrin. Expand
Therapeutic monitoring of the new antiepileptic drugs
TLDR
The pharmacological properties of some of the new antiepileptic drugs, in particular lamotrigine, zonisamide and, possibly, oxcarbazepine, topiramate and tiagabine, suggest that they may be suitable candidates for TDM. Expand
