Dioxin (2,3,7,8-tetrachlorodibenzo-p-dioxin) has been the subject of intensive investigations in laboratory animals during the last 2 decades. Toxicity studies have been conducted in several species of rodents and include several carcinogenicity studies as well as numerous mechanistic studies initiated to elucidate dioxin's mode of action, as both a carcinogen and a toxicant. Hepatotoxicity is a primary effect of dioxin. There has been an increase in hepatocellular tumors reported in both rats… Expand
2,3,7,8-Tetrachlorodibenzo-P-Dioxin (TCDD) or Diethylstilbestrol (DES) Cause Similar Hematopoietic Hypocellularity and Hepatocellular Changes in Murine Fetal Liver, but Differentially Affect Gene Expression
TLDR
Real time quantitative PCR using a preliminary panel of 4 genes suggested that the chemicals act through different gene targets, indicating possible pro-proliferative and antiapoptotic effects. Expand
Uterine Carcinogenesis by Chemicals/Hormones in Rodents.
TLDR
Sp spontaneous occurrence of uterine tumors, uterine carcinogenesis by chemicals and/or hormones and the influence of several factors on occurrence of uterusine tumors in rodents are reviewed and discussed. Expand
Tetrachlorodibenzo-p-Dioxin (TCDD) Inhibits Differentiation and Increases Apoptotic Cell Death of Precursor T-Cells in the Fetal Mouse Thymus
TLDR
Increased thymocyte apoptosis contributes to fetal thymic atrophy after TCDD exposure, according to histopathologic and cytometric data. Expand
The effect of TCDD dioxin on the rat liver in biochemical and histological assessment.
TLDR
The study confirmed that TCDD damages the rat liver in a dose-dependent manner and damaged CYP1A1 (based on higher resorufin levels in epiluminescence), which was confirmed by increased immunohistochemical reactivity of central areas of hepatic lobules. Expand
Expression of the arylhydrocarbon receptor in the peri-implantation period of the mouse uterus and the impact of dioxin on mouse implantation.
TLDR
The spatio-temporal expression of AhR during the peri-implantation phase of the mouse uterus may indicate functional roles of this orphan receptor in fetomaternal interactions as well as substantiate the risk of exposure to chemicals such as dioxins during the reproductive period. Expand
Thyroid Follicular Lesions Induced by Oral Treatment for 2 Years with 2,3,7,8-Tetrachlorodibenzo-p-dioxin and Dioxin-like Compounds in Female Harlan Sprague-Dawley Rats
TLDR
It is suggested that although dioxin-like compounds alter thyroid hormones and increase follicular cell hyperplasia, there is not an increase in thyroid adenoma or carcinoma in female Sprague-Dawley rats. Expand
Differences in acute toxicity syndromes of 2,3,7,8-tetrachlorodibenzo-p-dioxin and 1,2,3,4,7,8-hexachlorodibenzo-p-dioxin in rats.
TLDR
HxCDD was more potent than TCDD in inducing acute mortality in H/W and line A rats, contrary to what is predicted by toxic equivalency factor (TEF) values. Expand
Gestational exposure to 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) severely alters reproductive function of female hamster offspring.

TLDR
It is demonstrated that in utero administration of TCDD adversely affects growth, reproductive function, and anatomy in female hamster offspring given a dosage level nearly four orders of magnitude below the dosage level toxic to the adult animal. Expand
The Role of Retinyl Acetate in Chemoprevention
Retinoids have a role in the process of chemoprevention. They can inhibit promotion through inhibiting cell proliferation, enhancing apoptosis and anti-oxidative properties. In the present studyExpand
Embryonic exposure to 2,3,7,8-tetrachlorodibenzo-p-dioxin in chickens: effects of dose and embryonic stage on hatchability and growth.
TLDR
The delayed growth in TCDD treated chickens was accompanied by changes in T(3)/T(4) ratio that at some ages were significantly higher compared to control animals. Expand

Results of a two-year chronic toxicity and oncogenicity study of 2,3,7,8-tetrachlorodibenzo-p-dioxin in rats.
TLDR
It is indicated that continuous doses of TCDD sufficient to induce severe toxicity increased the incidence of some types of tumors, while reducing other types, and no increase in tumors occurred in those rats receiving sufficient TCDDs to induce slight or no manifestations of toxicity. Expand
Pathology reevaluation of the Kociba et al. (1978) bioassay of 2,3,7,8-TCDD: implications for risk assessment.
TLDR
The results indicate that the carcinogenic risk to humans from exposure to 2,3,7,8-TCDD is at least 16-fold lower than previous estimates derived from the Kociba et al. (1978) bioassay. Expand
In utero 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) alters reproductive morphology and function in female rat offspring.
TLDR
In the LE rat, vaginal and behavioral estrous cyclicity, estrous cycle-mediated running wheel activity, and female sexual behaviors at proestrus were not affected by gestational GD 15 TCDD treatment, but untreated stud males had difficulty attaining intromission and took longer to ejaculate. Expand
Promotion of endometriosis by 2,3,7,8-tetrachlorodibenzo-p-dioxin in rats and mice: time-dose dependence and species comparison.
TLDR
Differences observed between the rat and the mouse with respect to the magnitude of the effect on endometrial site diameter, measured effects on ovarian function, and evidence that mice and rats differ in their immune response to TCDD suggest that the mechanisms mediating T CDD's action to promote endometriosis are complex and may be different in rats and mice. TLDR
Viewing dioxin and related compounds as endocrine disruptors that may alter multiple pathways sheds some light on the complexities of this potent class of growth dysregulators. Expand
Induction of endometriosis in mice: a new model sensitive to estrogen.
TLDR
In the mouse model, slices of uterus were sutured to intestinal mesenteric vessels and by 3 weeks after surgery, these sites were cyst-like structures and the growth of the sites was hormone dependent. Expand
2,3,7,8-tetrachlorodibenzo-p-dioxin and related halogenated aromatic hydrocarbons: examination of the mechanism of toxicity.
TLDR
The toxicity of halogenated aromatic hydrocarbons appears to be due to the sustained expression of a normal cellular regulatory system, of which the author was previously unaware. Expand
Dose response for TCDD promotion of hepatocarcinogenesis in rats initiated with DEN: histologic, biochemical, and cell proliferation endpoints.
TLDR
It is demonstrated that dose-response relationships for TCDD's effects on cell proliferation growth of altered hepatic foci are different from previously reported effects on P450 gene expression, indicating that different biological or biochemical responses may exhibit different dose- response relationships. Expand
Increased oxidative DNA damage in livers of 2,3,7,8-tetrachlorodibenzo-p-dioxin treated intact but not ovariectomized rats.
TLDR
The results are consistent with the hypothesis that increased metabolism of endogenous estrogens to catechols by TCDD-induced enzymes may lead to increased oxidative DNA damage and hence contribute to T CDD-mediated hepatocarcinogenicity in female rats. Expand
2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) and related compounds as antioestrogens: characterization and mechanism of action.
TLDR
