Nattokinase producing bacterium, B. subtilis YF38, was isolated from douchi, using the fibrin plate method. The gene encoding this enzyme was cloned by polymerase chain reaction (PCR). Cytoplasmic expression of this enzyme in E. coli resulted in inactive inclusion bodies. But with the help of two different signal peptides, the native signal peptide of nattokinase and the signal peptide of PelB, active nattokinase was successfully expressed in E. coli with periplasmic secretion, and the…
Heterologous expression of nattokinase from B. subtilis natto using Pichia pastoris GS115 and assessment of its thrombolytic activity
TLDR
This study suggests that the P. pastoris expression system is an ideal platform for the large-scale, low-cost preparation of nattokinase.
High Cell Density Cultivation of A Recombinant Bacillus Subtilis for Nattokinase Production
TLDR
There is a balance between the cell growth rate and NK expression when recombinant Bacillus subtilis is cultured with a fed-batch process and the equilibrium state can be attained by optimizing the induction and feeding strategy, and thus a high cell density and enzyme activity can be achieved.
Stepwise modifications of genetic parts reinforce the secretory production of nattokinase in Bacillus subtilis
TLDR
The strategy established in this study can not only be used to overproduce NK in B. subtilis but also might be a promising pipeline to modify the genetic element for the synthetic secretory system.
Efficient expression of nattokinase in Bacillus licheniformis: host strain construction and signal peptide optimization
TLDR
The maximum NK fermentation activity in this study reaches the commercial production level of solid state fermentation, and this study provides a promising engineered strain for industrial production of nattokinase, as well as a potential platform host for expression of other target proteins.
Bacterial Fibrinolytic Enzyme Coding Sequences from Indonesian Traditional Fermented Foods Isolated Using Metagenomic Approach and Their Expression in Escherichia Coli
ABSTRACT Fibrinolytic proteins play an important role in treatment of cardiovascular disease. This study aimed to express coding sequences of new bacterial fibrinolytic protein from Indonesian
Fibrinolytic Serine Protease Isolation from Bacillus amyloliquefaciens An6 Grown on Mirabilis jalapa Tuber Powders
TLDR

Expression and secretion of an α‐amylase gene from a native strain of Bacillus licheniformis in Escherichia coli by T7 promoter and putative signal peptide of the gene
TLDR
The gene encoding a hyperthermostable α‐amylase from a Bacillus licheniformis native strain was cloned in pET24d transcription vector containing T7 promoter, and expressed in Escherichia coli BL21(DE3) cells, and this signal peptide is recommended for extracellular production of other foreign proteins in E. coli.
Efficient system of artificial oil bodies for functional expression and purification of recombinant nattokinase in Escherichia coli.
TLDR
The results have shown a great potential of this system for bacterial expression and purification of functional recombinant proteins.
Improved Secretory Production of Recombinant Proteins by Random Mutagenesis of hlyB, an Alpha-Hemolysin Transporter from Escherichia coli
TLDR
Two mutants were succeeded in isolating that had 27- and 15-fold-higher levels of subtilisin E secretion activity than the wild type did at 23°C, indicating that the improved secretion activity of AE104F is not always dependent on low temperature.
The Profibrinolytic Enzyme Subtilisin NAT Purified fromBacillus subtilis Cleaves and Inactivates Plasminogen Activator Inhibitor Type 1*
TLDR
Subtilisin NAT dose dependently enhanced tissue-type plasminogen activator-induced fibrin clot lysis both in the absence of rpPAI-1 and especially in the presence of rkaryotic PAI- 1, and seems to be induced through directfibrin dissolution by subtilisin NAT.
The Roles of the Prosequence of Thermolysin in Enzyme Inhibition and Folding in Vitro*

Purification and characterization of a fibrinolytic enzyme of Bacillus subtilis DC33, isolated from Chinese traditional Douchi
TLDR
The enzyme had a high degrading activity for the Bβ-chains and Aα-chain of fibrin(ogen), and also acted on thrombotic and fibrinolytic factors of blood, such as plasminogen, urokinase, thrombin, and kallikrein.
