Generation of high-producing clones is a perquisite for achieving recombinant protein yields suitable for biopharmaceutical production. However, in many industrially important cell lines used to produce recombinant proteins such as Chinese hamster ovary, mouse myeloma line (NS0), and hybridomas, only a minority of clones show significantly above-average productivity. Thus, in order to have a reasonable probability of finding rare high-producing clones, a large number of clones need to be…
Genetic analyses of Per.C6 cell clones producing a therapeutic monoclonal antibody regarding productivity and long-term stability
TLDR
This study shows for the first time the correlation between Qp and HC or LC expression levels and also mRNA stability in clones derived from human cell line Per.C6(®).
Deletion of a telomeric region on chromosome 8 correlates with higher productivity and stability of CHO cell lines
TLDR
Evidence is presented that increased production rates and cell line stability are correlated with the loss of the telomeric region of the chromosome 8, and this allows reduced efforts in single cell sorting, screening of fewer clones and raises the opportunity to circumvent time and labor‐intensive stability studies.
High-throughput screening and automation approaches for the development of recombinant therapeutic proteins
TLDR
This review outlines the high-throughput methodologies and technologies currently used for clone screening, selection and evaluation in bioprocess development.
A novel selection strategy for antibody producing hybridoma cells based on a new transgenic fusion cell line
TLDR
A novel selective technology based on an artificial cell surface construct by which secreted antibodies were connected to the corresponding hybridoma cell when they possess the desired antigen-specificity is developed and enables a fast and early stage selection and validation of monoclonal antibodies in one step.
Antibodies and Selection of Monoclonal Antibodies.
TLDR
In this chapter different selection strategies for antibody-producing hybridoma cells are presented and analysed regarding to their benefits compared to conventional limited dilution technology.
A new CHO (Chinese hamster ovary)-derived cell line expressing anti-TNFα monoclonal antibody with biosimilar potential
TLDR
High-quality adabut clones are generated which mAbs may be further developed as the first in-house made Brazilian biosimilar, demonstrating a success case for the incipient biotechnology industry, or also modified as biobetters, thus representing an innovative strategy for the patients’ welfare.
Integration of high‐throughput analytics and cell imaging enables direct early productivity and product quality assessment during Chinese Hamster ovary cell line development for a complex multi‐subunit vaccine antigen
TLDR
It is shown that comprehensive and quantitative assessment of cell growth, productivity, and product quality attributes are feasible at the 200–1,200 cell colony stage, within 14 days of the single cell cloning in static 96‐well plate culture.

High-throughput clonal selection of recombinant CHO cells using a dominant selectable and amplifiable metallothionein-GFP fusion protein.
TLDR
A high-throughput clonal selection protocol for the rapid isolation of high-producing clones from transfected CHO cells is demonstrated, indicating that MTGFP could be used as a selectable and amplifiable gene for the coexpression of foreign genes.
Rapid automated selection of mammalian cell line secreting high level of humanized monoclonal antibody using Clone Pix FL system and the correlation between exterior median intensity and antibody productivity
TLDR
An automated colony picker, Clone Pix FL system was used to replace limiting dilution method and was able to screen 1 x 10 5 clones secreting humanized monoclonal antibodies and high producer clones were selected in just 7 days.
Fluorescent labeling in semi-solid medium for selection of mammalian cells secreting high-levels of recombinant proteins
TLDR
FLSSM allows rapid visualization of the high secretors from transfected pools prior to picking, thus eliminating the tedious task of screening a high number of cell isolates, and is a versatile method for the screening of high producers for research and industry.
Key Determinants in the Occurrence of Clonal Variation in Humanized Antibody Expression of CHO Cells during Dihydrofolate Reductase Mediated Gene Amplification
TLDR
The data obtained here show that clonal variations in regard to antibody expression are found to be significant because clones can acquire MTX resistance by mechanisms other than DHFR‐mediated gene amplification despite the stepwise selection.
Rapid isolation of antigen-specific clones from hybridoma fusions
The Genetix ClonePix FL is a powerful way to find and collect antigen-specific clones from hybridoma fusions in a rapid one-step process—shortening the process timeline and allowing parallel
Murphy's law of limiting dilution cloning.
TLDR
Estimates of the probability that interesting subcultures are monotypic or monoclonal are derived consistent with this principle, and any cell line established via these estimates is suspect and may be polyclonally metastable.
