Objectives:Atomoxetine has been considered as an agonist replacement therapy for cocaine. We investigated the safety of the interaction of atomoxetine with cocaine and also whether cognitive function was affected by atomoxetine during short-term administration. Methods:In a double-blind placebo-controlled inpatient study of 20 cocaine-dependent volunteers, participants received atomoxetine 80 mg daily followed by 100 mg daily for 5 days each. On the fourth and fifth day at each dose, cocaine…
Atomoxetine in abstinent cocaine users: Cognitive, subjective and cardiovascular effects
TLDR
Atomoxetine has been proposed as a pharmacotherapy for cocaine use disorder (CUD) and shows potential as a treatment for CUD based on its efficacy as a cognitive enhancer in other clinical populations and impact on addictive processes in preclinical and human laboratory studies.
Atomoxetine in abstinent cocaine users: Sex differences
TLDR
A sex-differences secondary analysis of a human laboratory investigation of single doses of atomoxetine versus placebo in abstinent individuals with cocaine use disorders (CUD) found no difference in subjective drug effects, cognitive performance and cardiovascular measures.

Clinically Significant Drug–Drug Interactions with Agents for Attention-Deficit/Hyperactivity Disorder
TLDR
Clinicians should not expect that ADHD drugs modify each other’s serum concentrations, and clinicians need to be aware of the potential for cytochrome P450 (CYP) 2D6 inhibitors to increase amphetamine levels, which is equivalent to increasing dosages.
Machine learning analysis of cocaine addiction informed by DAT, SERT, and NET-based interactome networks
TLDR
The approach sets up a systematic protocol for artificial intelligence (AI)-based anti-cocaine addiction lead discovery by building ML/DL models for DAT, SERT, and NET interactome networkinformed machine learning/deep learning studies of cocaine addiction.
Recent developments in the behavioural and pharmacological enhancement of extinction of drug seeking
TLDR
Recent advances in the behavioural and pharmacological manipulation of extinction learning are examined, based on research from pre‐clinical models to highlight areas where promising pre-clinical models have not yet been integrated into current clinical practice but, if applied, could improve upon existing behavioural and Pharmacological methods.
Assessing the Value of the Zebrafish Conditioned Place Preference Model for Predicting Human Abuse Potential
TLDR
The findings reveal that using the current methodology, the zebrafish CPP model does not add value to the preclinical assessment of abuse potential, and rat and NHP models generally possessed significantly improved concordance, sensitivity, and positive predictive values for both clinical measures.
Single-trial lie detection using a combined fNIRS-polygraph system
TLDR
This study demonstrates a plausible solution toward single-trial lie-detection by combining fNIRS and the polygraph, and indicates that the combined fN IRS-polygraph system delivers much higher classification accuracy than that of a singular system.
Effectiveness Of Atomoxetine For Cognitive Function In Methamphetamine Dependent Patients; A Randomized Controlled Trial
Background: A substantial body of evidences indicates that methamphetamine abuse can lead to persistent and serious cognitive dysfunction. Preclinical studies and early pilot clinical investigations

Safety, tolerability and subject-rated effects of acute intranasal cocaine administration during atomoxetine maintenance.
TLDR
Atomoxetine attenuated the systolic pressure increasing effects and enhanced the heart rate increasing effects of cocaine, but was otherwise devoid of effects, indicating that cocaine is well tolerated during atomxetine maintenance.
Abuse liability assessment of atomoxetine in a drug-abusing population.
TLDR
Atomoxetine has significantly less abuse liability than methylphenidate or phentermine and no greater abuse liability more than desipramine.
Comparison of the subjective, physiological, and psychomotor effects of atomoxetine and methylphenidate in light drug users.
TLDR
The results suggest that atomoxetine does not induce subjective effects similar to methylphenidate and suggest that it is unlikely that atomxetine will have abuse liability.
Cocaine effects during D-amphetamine maintenance: a human laboratory analysis of safety, tolerability and efficacy.
TLDR
The physiological and behavioral effects of cocaine during D-amphetamine maintenance in seven cocaine-dependent participants are determined, further suggesting that agonist replacement therapy may be a viable strategy for managing cocaine abuse.
Atomoxetine Improved Response Inhibition in Adults with Attention Deficit/Hyperactivity Disorder
TLDR
At atomoxetine treatment in the ADHD patients was associated with shorter stop-signal reaction times and lower numbers of commission errors on the sustained attention task, and improved inhibitory control, most likely via noradrenergically mediated augmentation of prefrontal cortex function.
Acute Cocaine Responses Following Cocaethylene Infusion
TLDR
Findings indicate that continuous 8-hour exposure to cocaethylene is safe, produces acute tolerance to itself, and reduces some behavioral effects of coadministered cocaine.
Peripheral biomarkers of cognitive response to dopamine receptor agonist treatment
TLDR
Peripheral expression of dopamine-related genes may be useful as a biomarker of cognitive response to dopamine agonist drugs and of severity of addiction to dopamine-releasing stimulant drugs.
Atomoxetine Versus Stimulants for Treatment of Attention Deficit/Hyperactivity Disorder
TLDR
Psychostimulants are regarded as first-line pharmacologic treatment for children and adolescents with ADHD, as the efficacy and safety of these agents have been well established based on clinical trials and extensive naturalistic use.
Atomoxetine increases histamine release and improves learning deficits in an animal model of attention-deficit hyperactivity disorder: the spontaneously hypertensive rat.
TLDR
The results indicated that atomoxetine significantly ameliorated performance in the Morris water maze, consistent with its histamine-enhancing profile, which provides further support for the notion that the therapeutic effect of atomxetine could involve activation of histamine neurotransmission within the prefrontal cortex.
Atomoxetine Increases Extracellular Levels of Norepinephrine and Dopamine in Prefrontal Cortex of Rat: A Potential Mechanism for Efficacy in Attention Deficit/Hyperactivity Disorder
TLDR
It is hypothesized that the atomoxetine-induced increase of catecholamines in PFC, a region involved in attention and memory, mediates the therapeutic effects of atomxetine in ADHD.
