Acute lung injury (ALI) is a lung disease characterized by pulmonary edema and severe hypoxia. The past decade hosted a search for endogenous mechanisms controlling lung inflammation and pulmonary edema during ALI. As such, recent evidence indicates extracellular adenosine in orchestrating the resolution of pulmonary edema and inflammation during ALI.
HIF1A Reduces Acute Lung Injury by Optimizing Carbohydrate Metabolism in the Alveolar Epithelium
A study of acute lung injury reveals the involvement of transcription factor HIF1A in lung protection, where normoxic HIF1A stabilization functions to control alveolar epithelial glucose metabolism.
Role of Apoptosis in Amplifying Inflammatory Responses in Lung Diseases
TLDR
The role of apoptosis is explored in ALI and COPD, with particular attention to the contribution of DAMP release in augmenting the inflammatory response in these disease states.
Adenosine at the Interphase of Hypoxia and Inflammation in Lung Injury
TLDR
The role ofadenosine at the interphase of hypoxia and inflammation in ARDS and chronic lung diseases, as well as the current strategy for therapeutic targeting of the adenosine signaling pathway are discussed.
Distinct Roles for the A2B Adenosine Receptor in Acute and Chronic Stages of Bleomycin-Induced Lung Injury
TLDR
In this model, the effects of A2BR removal on acute lung injury were negligible; however, there were substantial reductions in pulmonary fibrosis, supporting a profibrotic role for this receptor.
Alveolar Epithelial A2B Adenosine Receptors in Pulmonary Protection during Acute Lung Injury
TLDR
It is suggested that alveolar epithelial A2B adenosine receptor signaling contributes to lung protection, and they implicate inhaled A1B adenoine receptor agonists in ALI treatment.
Protective effect of adenosine receptors against lipopolysaccharide-induced acute lung injury.
TLDR
Results demonstrate that activation of ARs protects and restores vascular barrier functions and reduces inflammation in LPS-induced ALI.
CD73+ regulatory T cells contribute to adenosine‐mediated resolution of acute lung injury
TLDR
CD73+ regulatory T cells contribute to adenosine‐mediated resolution of acute lung injury, and adoptive transfer of cd73‐deficient Tregs into Rag–/– mice emulated the observed phenotype in cd73–/- mice.
Alcohol Worsens Acute Lung Injury by Inhibiting Alveolar Sodium Transport through the Adenosine A1 Receptor
TLDR
Exposure to alcohol reduced alveolar fluid clearance, downregulated Na,K-ATPase in the lung tissue and worsened hyperoxia-induced lung injury, and alcohol induced increases in lung adenosine levels may be responsible for reduction in alveolars fluid clearance and associated worsening of lung injury.
Enhancing Extracellular Adenosine Levels Restores Barrier Function in Acute Lung Injury Through Expression of Focal Adhesion Proteins
TLDR
A role of dipyridamole and adenosine signaling in preventing or treating ALI is highlighted and Ent2 and Adora2b are identified as key mediators in important for the resolution of ALI.

The role of platelets in acute lung injury (ALI).
TLDR
Key molecules involved in this interaction include P-selectin and the eicosanoid thromboxane A2 (TXA2), suggesting potential new targets for pharmacological intervention are suggested.
Role of pulmonary adenosine during hypoxia: extracellular generation, signaling and metabolism by surface adenosine deaminase/CD26
Numerous parallels exist between limited oxygen availability (hypoxia) and acute inflammation. The lungs in particular are prone to acute inflammation during hypoxia, resulting in pulmonary edema,
A2B adenosine receptor signaling attenuates acute lung injury by enhancing alveolar fluid clearance in mice.
TLDR
A role for A2BAR signaling in attenuating VILI is revealed and this receptor is implicate as a potential therapeutic target during acute lung injury.
Role of A2B adenosine receptor signaling in adenosine-dependent pulmonary inflammation and injury.
TLDR
Findings suggest that antagonism of A2BAR-mediated responses may prove to be a beneficial therapy in chronic lung diseases associated with increased adenosine, and influences pathways critical for pulmonary inflammation and injury in vivo.

TLDR
A landmark controlled randomized clinical trial in critically ill patients with ALI showed that mechanical ventilation with a lung protective strategy improves outcome as compared with conventional mechanical ventilation.
Complete reversal of acid-induced acute lung injury by blocking of platelet-neutrophil aggregation.
TLDR
Inhibition of platelet-neutrophil aggregation improved gas exchange, reduced neutrophil recruitment and permeability, and prolonged survival, and these findings may translate into improved clinical treatments for ALI.
Adenosine-Dependent Pulmonary Fibrosis in Adenosine Deaminase-Deficient Mice1
TLDR
It is demonstrated that chronic adenosine elevations are associated with pulmonary fibrosis in ADA-deficient mice and suggested that the adenosines functions as a profibrotic signal in the lung.
Identification of Ectonucleotidases CD39 and CD73 in Innate Protection during Acute Lung Injury1
TLDR
Treatment with their soluble compounds as a therapeutic strategy for noninfectious ALI is suggested and a previously unrecognized role for CD39 and CD73 in lung protection is revealed.
A protective role for the A1 adenosine receptor in adenosine-dependent pulmonary injury.
TLDR
It is demonstrated that the A(1)AR plays an anti-inflammatory and/or protective role in the pulmonary phenotype seen in ADA-deficient mice, which suggests that A( 1)AR signaling may serve to regulate the severity of pulmonary inflammation and remodeling seen in chronic lung diseases by controlling the levels of important mediators of pulmonaryinflammation and damage.
