}Exposure to air pollutants such as ozone (O(3)) induces airway hyperresponsiveness (AHR) and airway inflammation. Toll-like receptors (TLR) are first-line effector molecules in innate immunity to infections and signal via adapter proteins, including myeloid differentiation factor-88 (MyD88). We investigated the sensing of ozone by TLR2, TLR4, and MyD88. Ozone induced AHR in wild-type (WT) C57BL/6 mice, but AHR was absent in TLR2(-/-), TLR4(-/-), and MyD88(-/-) mice. Hyaluronan Signaling during Ozone-Induced Lung Injury Requires TLR4, MyD88, and TIRAP
- BiologyPloS one
- 2011
The findings support that ozone-induced airway hyperresponsiveness is dependent on the HA-TLR4-MyD88-TIRAP signaling pathway.
Resistin deficiency in mice has no effect on pulmonary responses induced by acute ozone exposure.
- BiologyAmerican journal of physiology. Lung cellular and molecular physiology
- 2015
It is demonstrated that murine resistin is increased in the lungs of wild-type mice following acute O3 exposure but does not promote O3-induced lung pathology.
TLR4 is necessary for hyaluronan-mediated airway hyperresponsiveness after ozone inhalation.
- Biology, Environmental ScienceAmerican journal of respiratory and critical care medicine
- 2010
The observations support the observation that extracellular matrix HA contributes to ozone-induced airways disease and support that TLR4 contributes to the biological response to HA by mediating both the production of proinflammatory cytokines and the development of ozone- induced AHR.
Non-Essential Role for TLR2 and Its Signaling Adaptor Mal/TIRAP in Preserving Normal Lung Architecture in Mice
- BiologyPloS one
- 2013
In vivo evidence is provided for the non-essential role for TLR2, unlike the related TLR4, in maintaining the normal architecture of the lung and it is revealed that Mal differentially facilitates the anti-apoptotic, but not oxidant suppressive, activities of TLR 4 in the lung, both of which appear to be essential forTLR4 to prevent the onset of emphysema.

Interleukin-1α Mediates Ozone-Induced Myeloid Differentiation Factor-88-Dependent Epithelial Tissue Injury and Inflammation
- Biology, MedicineFront. Immunol.
- 2018
It is demonstrated that the release of the alarmin IL-1α after ozone exposure is mediated by MyD88 signaling in epithelial cells and that IL- 1α may represent a therapeutic target to attenuate ozone-induced lung inflammation and hyperreactivity.
IL-17A Modulates Oxidant Stress-Induced Airway Hyperresponsiveness but Not Emphysema
- BiologyPloS one
- 2013
IL-17R signalling underlies the increase in airway hyperresponsiveness seen after ozone exposure, mediated by the increased contractility of airway smooth muscle.
Cigarette smoke regulates the expression of TLR4 and IL-8 production by human macrophages

TLR4 may be involved in the pathogenesis of lung emphysema and oxidative stress and seems to be a crucial contributor in lung inflammation.
Involvements of p38 MAPK and oxidative stress in the ozone-induced enhancement of AHR and pulmonary inflammation in an allergic asthma model
- Biology, MedicineRespiratory Research
- 2017
Both p38 MAPK and oxidative stress were found to play a critical role in this process and simultaneous inhibition of these two pathways significantly reduced the O3-elicited detrimental effects on the asthma exacerbation.

It is indicated that IL-6 deficiency reduces airway neutrophilia, as well as the levels of BALF s TNFR1 and sTNFR2 following acute high dose and/or subacute low-dose O(3) exposure, but has no effect on O( 3)-induced AHR.
