Distinct signaling pathways are reported to maintain pluripotency in embryo-derived stem cells. Mouse embryonic stem cells (ESCs) respond to leukemia inhibitory factor (LIF) and bone morphogenetic protein (BMP)-mediated activity, whereas human ESCs depend upon Fibroblast growth factor (FGF) and activin signaling. In the majority of mammals investigated, however, the signals that support stem cell pluripotency are not well defined, as is evident by the persistent difficulties in maintaining…

Porcine Induced Pluripotent Stem Cells Require LIF and Maintain Their Developmental Potential in Early Stage of Embryos
TLDR
The generation of piPS cells from fibroblasts using retrovirus carrying four mouse transcription factors showed thatpiPS cells retain the ability to integrate into inner and outer layers of the blastocysts, and support the NT embryos development to blastocyst.
Optimization of culture conditions for maintaining porcine induced pluripotent stem cells.
TLDR
This study investigated morphological and biological progression at the early stage of porcine somatic cell reprogramming, and explored suitable conditions to increase the induction efficiency of piPSCs.
Optimization of culture conditions for maintaining porcine induced pluripotent stem cells.
TLDR
Based on the potential for self-renewal and activation of pluripotent markers, it was found that the culture condition of 4F+T plus LIF and bFGF plus MEF feeder promoted PEF reprogramming more efficiently than the other conditions tested here.
DNA repair and replication links to pluripotency and differentiation capacity of pig iPS cells
TLDR
Improved induction conditions using various small chemicals generated pig iPSCs that exhibited high pluripotency and differentiation capacity that can contribute to chimeras, but their potency was reduced with increasing passages by teratoma formation test, and correlated with declined expression levels of Rex1, an important marker for naïve state.
Generation of Intermediate Porcine iPS Cells Under Culture Condition Favorable for Mesenchymal-to-Epithelial Transition
TLDR
This study found that combination of three growth factors (LIF, FGF2 and BMP4) and two inhibitors (2i: CHIR99021 and SB431542) could generate an intermediate pluripotent state of piPSCs, which were named as LFB2i-piPSCs.
Bovine trophectoderm cells induced from bovine fibroblasts with induced pluripotent stem cell reprogramming factors
TLDR
Bovine trophectoderm can be induced via reprogramming factor expression from bovine liver‐derived fibroblasts, although other fibroblast populations—e.g., derived from fetal thigh tissue—may produce similar results, albeit at lower frequencies.
Preserving self-renewal of porcine pluripotent stem cells in serum-free 3i culture condition and independent of LIF and b-FGF cytokines
TLDR
It was found that LIF and b-FGF were required for porcine cell reprogramming, but were not essential cytokines for maintaining the self-renewal and pluripotency of piPSCs.
Pluripotent and Metabolic Features of Two Types of Porcine iPSCs Derived from Defined Mouse and Human ES Cell Culture Conditions
TLDR
It is found that culture conditions affected pluripotent and metabolic features of piPSCs, and the mpiPSCs showed mitochondrial features of naive ESCs and lipid droplets accumulation, which may facilitate understanding of the gene regulation network and metabolism inPiPSCs and promote derivation of bona fide pESCs for translational medicine.
Barriers for Deriving Transgene‐Free Pig iPS Cells with Episomal Vectors
TLDR
It is found that incomplete reprogramming manifested by a reliance on sustained expression of exogenous‐reprogramming factors appears to be the main reason for the inability of porcine iPSCs to form iPSC‐derived piglets.

Derivation of induced pluripotent stem cells from pig somatic cells
TLDR
It is concluded that porcine somatic cells can be reprogrammed to form piPSC, and such cell lines derived from individual animals could provide a means for testing the safety and efficacy of stem cell-derived tissue grafts when returned to the same pigs at a later age.
Reprogramming of Sheep Fibroblasts into Pluripotency under a Drug-Inducible Expression of Mouse-Derived Defined Factors
TLDR
This study shows sheep fibroblasts can be reprogrammed to pluripotency by defined factors using a drug-inducible system and shows that the substitution of knockout serum replacement with fetal bovine serum in culture improves the reprogramming efficiency of sheep iPSCs.
Leukemia Inhibitory Factor (LIF)-dependent, Pluripotent Stem Cells Established from Inner Cell Mass of Porcine Embryos*
TLDR
These cells derived from the inner cell mass of pig blastocysts are clearly distinct from the FGF2-dependent “primed” induced pluripotent stem cells described recently from porcine mesenchymal cells, and are consistent with the hypothesis that the up-regulation of KLF4, as well as POU5F1, is required to create and stabilize the naive pluripotency.
Human embryonic stem cells with biological and epigenetic characteristics similar to those of mouse ESCs
TLDR
The generation of validated “naïve” human ESCs will allow the molecular dissection of a previously undefined pluripotent state in humans and may open up new opportunities for patient-specific, disease-relevant research.
Activin/Nodal and FGF pathways cooperate to maintain pluripotency of human embryonic stem cells
TLDR
It is reported that Activin/Nodal signalling through Smad2/3 activation is necessary to maintain the pluripotent status of hESCs, and this findings suggest that the Activin / Nodal pathway maintains pluripotency through mechanism(s) in which FGF acts as a competence factor.
Derivation of pluripotent epiblast stem cells from mammalian embryos
TLDR
It is shown that pluripotent stem cells can be derived from the late epiblast layer of post-implantation mouse and rat embryos using chemically defined, activin-containing culture medium that is sufficient for long-term maintenance of human embryonic stem cells.
Klf4 reverts developmentally programmed restriction of ground state pluripotency
TLDR
Mouse embryonic stem cells derived from pluripotent early epiblast contribute functionally differentiated progeny to all foetal lineages of chimaeras and were maintained after Cre-mediated deletion of the Klf4 transgene, formally demonstrating complete and stable reprogramming of developmental phenotype.
