SummaryThe relationship between tumour necrosis factor-α (TNF-α) gene polymorphism and inhibitory effects of triptolide on TNF-α production from perlpheral blood mononuclear cells (PBMC) of healthy humans was investigated. Genomic DNA from 41 healthy people was typed for TNF-α—308 polymorphism by allele-specific polymorphism chain reaction (AS-PCR). The TNF-α concentration in the supernatant was measured by ELISA. The results showed that the production of TNF-α from TNF-α—308 non-G/G genotype… Expand

Most studies of AKI genetics involve hypothesis‐driven (rather than hypothesis‐generating) candidate gene investigations that have failed to identify contributory variants consistently, and there remains a pressing need for additional GWA studies. Expand

Investigation of the Prognostic Value of TNF-α Gene Polymorphism among Patients Treated with Infliximab, and the Effects of Infliximab Therapy on TNF-α Production and Apoptosis
Infliximab, a chimeric anti-tumor necrosis factor (TNF) antibody, is highly effective for the treatment of Crohn’s disease and rheumatoid arthritis, and the determination of TNF polymorphism may help identify more suitable candidates for infliximab treatment. Expand
Investigation of the prognostic value of TNF-alpha gene polymorphism among patients treated with infliximab, and the effects of infliximab therapy on TNF-alpha production and apoptosis.
Although in vitro infliximab treatment for 48 h resulted in significant apoptosis in PBMCs, in ex vivo samples from RA patients who received inflIXimab, apoptosis was only 13.3 +/- 1.6% and it is suggested that the determination of TNF polymorphism may help identify more suitable candidates for inflixIMab treatment. Effects of a polymorphism in the human tumor necrosis factor alpha promoter on transcriptional activation.
It is demonstrated, with reporter genes under the control of the two allelic TNF promoters, that TNF2 is a much stronger transcriptional activator than the common allele (TNF1) in a human B cell line. Expand
Polymerase chain reaction analysis for the tumor necrosis factor alpha-308 (G>A) gene polymorphism in relation to ankylosing spondylitis
There is a strong association between the TNF α gene polymorphism and ankylosing spondylitis and TNF2 gene may play a role in AS susceptibility. Expand
The −308 tumor necrosis factor-α promoter polymorphism effects transcription
It is demonstrated that the region (−323 to −285) encompassing −308 in the TNF2 allele binds nuclear factors differently to the same region in the promoter of the more common TNF1 allele, suggesting that the −308 GA polymorphism may play a role in the altered TNF-α gene expression observed in individuals with the HLA Al, B8, DR3 haplotype.
