As a GSK-3β inhibitor reported by our group, K313 is a novel benzoxazole derivative and displays anti-inflammatory properties in RAW264.7 macrophages without cytotoxicity. The activity of GSK-3β affects the differentiation and maturation of bone marrow-derived dendritic cells (DCs). This study aims to investigate whether K313 can be used to induce regulatory/tolerogenic dendritic cells (DCregs), and the therapeutic effects of DCregs induced by K313 in the autoimmune model of experimental… Expand

Paving the way towards an effective treatment for multiple sclerosis: advances in cell therapy
TLDR
This review analyzes current MS treatments as well as different cell-based therapies that have been proposed to restore homeostasis in MS patients (tolerogenic dendritic cells, regulatory T cells, mesenchymal stem cells, and vaccination with T cells). Expand

Tolerogenic Dendritic Cells Induced by BD750 Ameliorate Proinflammatory T cell Responses and Experimental Autoimmune Encephalitis in Mice
TLDR
Treatment with BD750 induced tolDC that inhibited proinflammatory T cell immunity in vitro and in vivo and may be valuable for development of new therapies for EAE and other autoimmune diseases. Expand
Midkine Inhibits Inducible Regulatory T Cell Differentiation by Suppressing the Development of Tolerogenic Dendritic Cells
TLDR
It is demonstrated that MK was produced by inflammatory cells, in particular, CD4+ T cells under inflammatory conditions, and suggested that MK aggravates EAE by suppressing DCreg development, thereby impairing the Treg population. Expand
Tolerogenic Dendritic Cells Generated with Tofacitinib Ameliorate Experimental Autoimmune Encephalomyelitis through Modulation of Th17/Treg Balance
TLDR
The results revealed that tofacitinib-treated DCs maintained a steady semimature phenotype with a low level of proinflammatory cytokines and costimulatory molecules, and the antigen-specific tolDCs may represent a new avenue of research for the development of future clinical treatments for MS. Expand
Hepatocyte growth factor inhibits CNS autoimmunity by inducing tolerogenic dendritic cells and CD25+Foxp3+ regulatory T cells
TLDR
The results suggest that, by combining both potentially neuroprotective and immunomodulatory effects, HGF is a promising candidate for the development of new treatments for immune-mediated demyelinating diseases associated with neurodegeneration such as multiple sclerosis. Expand
IL-6 blockade inhibits the induction of myelin antigen-specific Th17 cells and Th1 cells in experimental autoimmune encephalomyelitis
TLDR
Findings indicate that anti-IL-6R mAb treatment might represent a novel therapy for human MS through the inhibition of the development of MOG-peptide-specific Th17 cells and Th1 cells, which in turn leads to reduced infiltration of T cells into the CNS. Expand
CDK inhibitors suppress Th17 and promote iTreg differentiation, and ameliorate experimental autoimmune encephalomyelitis in mice.
TLDR
Kenpaullone and Roscovitine suppressed experimental autoimmune encephalomyelitis (EAE), a typical Th17-mediated autoimmune disease model, and data suggest that CDK inhibitors modulate TGF-β-signaling pathways, which restore TGF -β-mediated suppression of IL-2 production, thereby modifying the Th17/iTreg balance. Expand
1,25‐dihydroxyvitamin D3‐induced dendritic cells suppress experimental autoimmune encephalomyelitis by increasing proportions of the regulatory lymphocytes and reducing T helper type 1 and type 17 cells
TLDR
It is concluded that transfer of VD3‐DCs suppressed EAE by increasing proportions of regulatory T cells, CD4+ IL‐10+ T cells and regulatory B cells in spleen and reducing infiltration of Th1 and Th17 cells into spinal cord, which suggests a possible immunotherapy method in MS. Expand
Dendritic cell maturation occurs through the inhibition of GSK-3β.
TLDR
The results show that GSK-3β signaling controls DC maturation and suggest that this kinase could be manipulated to modulate adaptive immunity. Expand
Estriol Generates Tolerogenic Dendritic Cells In Vivo That Protect against Autoimmunity
TLDR
E3 generates tolerogenic DCs, which protect against the inflammatory autoimmune disease EAE, and transfer of E3 DCs to mice prior to active induction of EAE protected them from developing EAE through immune deviation to a Th2 response. Expand
Allostimulatory Effects of Dendritic Cells with Characteristic Features of a Regulatory Phenotype
TLDR
LiClPAM3 DCs displayed a tolerogenic phenotype accompanied with a low expression of co-stimulatory molecules and a high IL-10:IL-12 production ratio, but appeared to be highly immunogenic, since allograft rejection was accelerated. Expand
