MicroRNAs (miRNAs) and trans-acting siRNAs (ta-siRNAs) are essential to the establishment of adaxial–abaxial (dorsoventral) leaf polarity. Tas3-derived ta-siRNAs define the adaxial side of the leaf by restricting the expression domain of miRNA miR166, which in turn demarcates the abaxial side of leaves by restricting the expression of adaxial determinants. To investigate the regulatory mechanisms that allow for the precise spatiotemporal accumulation of these polarizing small RNAs, we used… Expand
Auxin regulation of the microRNA390-dependent transacting small interfering RNA pathway in Arabidopsis lateral root development
TLDR
Evidence is provided that miR390 expression is sensitive to TIR1-dependent transcriptional regulation and auxin concentration, and also that mutual negative-regulation between the tasiRNA-ARF pathway and ARf4 modulates the spatiotemporal expression of ARF4. Expand
Trans-acting Small Interfering RNAs: Biogenesis, Mode of Action, and Role in Plant Development
TLDR
The phylogenetically conserved TAS3 pathway controls the abundance of several ARF genes, which encode transcription factors mediating the effects of the phytohormone auxin, three essential features of plant development. Expand
Endogenous TasiRNAs Mediate Non-Cell Autonomous Effects on Gene Regulation in Arabidopsis thaliana
TLDR
It is shown that the silencing activities of ARF-regulating tasiRNAs are indeed manifested non-cell autonomously to spatially control ARF activities and might provide effective gene silencing at a distance beyond the reach of most miRNAs. Expand
RBE controls microRNA164 expression to effect floral organogenesis
TLDR
It is shown that the C2H2 zinc finger transcriptional repressor encoded by RABBIT EARS (RBE) regulates the expression of all three miR164 genes, which indicates that one role of RBE is to fine-tune MiR164 expression to regulate the CUC1 and CUC2 effector genes, which, in turn, regulate developmental events required for sepal and petal organogenesis. Genome-Wide Analysis of leafbladeless1-Regulated and Phased Small RNAs Underscores the Importance of the TAS3 ta-siRNA Pathway to Maize Development
TLDR
Diversity in TAS pathways and their targets cannot fully account for the phenotypic differences conditioned by ta-siRNA biogenesis mutants across plant species, and it is proposed that divergence in the gene networks downstream of the ARF3 transcription factors or the spatiotemporal pattern during leaf development constitute key factors underlying the distinct contributions of the ta-SIRNA pathway to development in maize, Arabidopsis, and possibly other plant species. Expand
miRNAs in the biogenesis of trans-acting siRNAs in higher plants.
TLDR
The mechanisms for trans-acting (ta)-siRNA biogenesis and function are reviewed, including pathways that utilize components of the miRNA and transitive RNAi defense. Expand
Plant small RNAs: advancement in the understanding of biogenesis and role in plant development
TLDR
The advances made in understanding the biogenesis, functional conservation/divergence in miRNA-mediated gene regulation, and the developmental role of small RNAs in different plant species are discussed. Expand
Comparative analysis of non-autonomous effects of tasiRNAs and miRNAs in Arabidopsis thaliana
TLDR
Using a system based on the silencing of the CH42 gene, the mobility of silencing signals initiated in phloem companion cells by artificial microRNAs (miRNA) and trans-acting siRNA (tasiRNA) that have the same primary sequence are tracked, indicating that biogenesis can determine the non-autonomous effects of sRNAs. Expand
Role of miRNAs in root development of model plant Arabidopsis thaliana
TLDR
Recent advances made in understanding the miRNA-mediated regulation of root development in the model plant Arabidopsis thaliana are summarized and the molecular crosstalk between different miRNAs, ta-si RNAs, and concerned target genes that regulate root growth and branching have been addressed. Expand
Requirement of MIR165A primary transcript sequence for its activity pattern in Arabidopsis leaf primordia
TLDR
It is proposed that not only maturemiR165 sequence but also the MIR165A primary transcript sequence are required for the confinement of miR165 activity to the abaxial side of leaf primordia. Expand

microRNA-mediated repression of rolled leaf1 specifies maize leaf polarity
TLDR
It is shown that miRNA166 constitutes a highly conserved polarizing signal whose expression pattern spatially defines the expression domain of the maize hd-zipIII family member rolled leaf1 (rld1), suggesting that mi RNA166 may form a movable signal that emanates from a signalling centre below the incipient leaf. Expand
Two small regulatory RNAs establish opposing fates of a developmental axis.
TLDR
It is shown that leafbladeless1 encodes a key component in the trans-acting small interfering RNA (ta-siRNA) biogenesis pathway that acts on the adaxial side of developing leaves and demarcates the domains of hd-zipIII and miR166 accumulation. Expand
Spatio-temporal accumulation of microRNAs is highly coordinated in developing plant tissues.
TLDR
In situ analysis is used to demonstrate that miRNAs accumulate spatially and temporally in a highly restricted manner in Nicotiana benthamiana and Arabidopsis thaliana, implying a pivotal role in fine-tuning of target gene expression in plant development. Expand
microRNA-Directed Phasing during Trans-Acting siRNA Biogenesis in Plants
TLDR
Data support a model in which miRNA-guided formation of a 5' or 3' terminus within pre-ta-siRNA transcripts, followed by RDR6-dependent formation of dsRNA and Dicer-like processing, yields phased ta-siRNAs that negatively regulate other genes. Expand
microRNA-directed cleavage of ATHB15 mRNA regulates vascular development in Arabidopsis inflorescence stems.
TLDR
It is shown that miR166-mediated ATHB15 mRNA cleavage is a principal mechanism for the regulation of vascular development and is likely to be a conserved mechanism that regulates vascular development in all vascular plants. Expand
Redundancy and specialization among plant microRNAs: role of the MIR164 family in developmental robustness
TLDR
It was found that a loss of miR164 activity leads to a severe disruption of shoot development, in contrast to the effect of mutation in any single MIR164 gene, which indicates that these miRNAs are indeed functionally redundant. Expand
Specification of adaxial cell fate during maize leaf development
TLDR
It is proposed that a single genetic pathway involving lbl1, rld1 and the yabby genes integrates positional information within the SAM, and leads to adaxial/abaxial patterning and mediolateral outgrowth of the leaf. Expand
DRB4-Dependent TAS3 trans-Acting siRNAs Control Leaf Morphology through AGO7
TLDR
Results indicate that the dsRNA binding protein DRB4 is required for proper ta-siRNA production, presumably by interacting with DCL4, an interaction analogous to that of HYL1 with D CL1 during miRNA production , and that TAS3 ta-SIRNAs are required for Proper leaf development through the action of AGO7/ZIPPY. TLDR
It is shown that regulated expression of several Arabidopsis thaliana pre-miRNA genes induced a range of phenotypic alterations, the most extreme ones being a phenocopy of combined loss of their predicted target genes, indicating quantitative regulation by miRNA as a potential source for diversity in developmental outcomes.
