Abstract Regions of gliosis surround deposits of (β-amyloid peptide (βAP) in senile plaques of Alzheimer's disease (AD). The association between reactive astrocytes and βAP in senile plaques is most pronounced in cortex and hippocampus but not at other anatomical sites of βAP deposition. We hypothesized that this region-specific pathology in AD could be attributed to differences in glial reactivity in different parts of the central nervous system (CNS). To test this hypothesis, we assayed…

Understanding the spatial differences and roles of glia will allow us to assess how these interactions can influence the state and progression of the disease, and will be critical for identifying therapeutic strategies.

The lack of fibrillar amyloid deposits in cerebellum, despite the presence of well‐characterized markers thought to mediate the production of Aβ, suggests that this brain region may be lacking certain factors necessary for fibril formation or that the cere Bellum responds differently to stimuli that successfully mediate inflammation in affected cortex.

The role of inflammatory reactions in Alzheimer’s has been lent additional credence by indications of such reactions in mice transgenic for βAPP, and interactions between Aβ and α2-macroglobulin or components of the complement cascade are addressed.
