This paper describes the development of a reference measurement procedure to quantify human C-reactive protein (CRP) in serum using affinity techniques prior to tryptic digestion and liquid chromatography-tandem mass spectrometry (LC-MS/MS) for the certification of reference materials in clinically relevant ranges. The absence of a suitable internal standard for the CRP measurement, necessary to eliminate potential measurement bias in both the affinity purification and trypsin digestion steps…

Analysis of the physicochemical state of C-reactive protein in different preparations including 2 certified reference materials.
TLDR
Most of the 20% loss in measured CRP after lyophilization of the material can be accounted for by the dissociation of natively pentameric CRP into subunits.

Development of a primary reference material of natural C-reactive protein: verification of its natural pentameric structure and certification by two isotope dilution mass spectrometry.
TLDR
A CRP primary certified reference material (CRM) of well-characterised purity was determined that could be used to calibrate an IDMS-based reference method, that could then be use to assign target values to secondary CRMs, giving a good foundation for establishing a complete traceability chain for CRP.
Quantification of cardiac troponin I in human plasma by immunoaffinity enrichment and targeted mass spectrometry
TLDR
It is demonstrated that quantification of cTnI in native plasma from myocardial infarction patients can be achieved based on an ID-LC-MS/MS method, a first step for future certification of matrix-based reference materials, which may be used to help harmonize discordant cTNI clinical assays.
Understanding the role of immunoaffinity-based mass spectrometry methods for clinical applications.
TLDR
It is shown that the limited specificity of an anti–1α,25-dihydroxyvitamin D capture antibody could be used with LC-MS/MS to deliver a sensitive, highly specific assay for 5 vitamin D–related metabolites.
Chemical analysis of C-reactive protein synthesized by human aortic endothelial cells under oxidative stress.
TLDR
Test the hypothesis that oxidized low-density lipoprotein (ox-LDL) induces the release of CRP from human aortic endothelial cells (HAECs) and optimize several analytical methods to identify CRP released from cultured cells in a model of atherogenic stress.
Quantitative Analysis of Multiple Charged Large Molecules in Human or Rat Plasma Using Liquid Chromatography Tandem Mass Spectrometry
TLDR
This dissertation was focused on the development and validation of assays that can be used for quantitative analysis of peptides or proteins in plasma using liquid chromatography tandem mass spectrometry (LC-MS/MS).
Quantification of antibody coupled to magnetic particles by targeted mass spectrometry
TLDR
It is demonstrated that nanoparticles more efficiently bind antibody when compared to microparticles, which was expected due to the larger surface area per unit mass of the nanoparticles compared to the same mass ofmicroparticles.
