Objectives. Trichostatin A (TSA) is an antifungal antibiotic and a potent reversible inhibitor of histone deacetylase (HDAC). Pharmacologically, TSA is an antitumoral agent, but it is also a therapeutic agent in some non-tumoral diseases. Th e molecular mechanisms through which TSA inhibits the proliferation of cancerous cells are not well understood. Because the main target of HDAC inhibitors (HDI) is nuclear chromatin, we proposed to determine the eff ects of TSA morphologically, genetically… Expand

HDAC inhibition through valproic acid modulates the methylation profiles in human embryonic kidney cells
TLDR
This work establishes a dynamic correlation between histone methylation events and HDAC inhibition and may define newer epigenetic strategies for treating neurodevelopmental and oncological disorders. Expand

The histone deacetylase inhibitor trichostatin a has genotoxic effects in human lymphoblasts in vitro.
TLDR
TSA is both a clastogen and aneugen in the TK6 cell line and it is proposed that the observed cytostatic and apoptotic properties of TSA may partially be due to this genotoxicity. Expand
To the nucleolar density and size in apoptotic human leukemic myeloblasts produced in vitro by Trichostatin A.
TLDR
The presented results indicated that large nucleoli intensely stained for RNA need not be necessarily related to the high nucleolar biosynthetic or cell proliferation activity and may be also present in apoptotic cells responding to the cytostatic treatment. Expand
Inhibition of telomerase activity and human telomerase reverse transcriptase gene expression by histone deacetylase inhibitor in human brain cancer cells.
TLDR
It is suggested that telomersase and hTERT might be the primary targets of TSA which may have the potential to be used as a telomerase inhibitor in cancer therapy. Expand
Trichostatin A-induced histone acetylation causes decondensation of interphase chromatin
TLDR
The effect of trichostatin A (TSA)-induced histone acetylation on the interphase chromatin structure was visualized in vivo with a HeLa cell line stably expressing histone H2A, which was fused to enhanced yellow fluorescent protein to characterize the effect of TSA on cell cycle progression and develop a protocol that allowed the identification of G1 phase cells on microscope coverslips. Epigenetic drugs as pleiotropic agents in cancer treatment: Biomolecular aspects and clinical applications
TLDR
Data from in vitro experimentations and in vivo pre‐clinical and clinical studies clearly indicate epigenetic drugs as effective modifiers of cancer phenotype and as positive regulators of tumor cell biology with a relevant therapeutic potential in cancer patients are focused on. Expand
Nucleolus and Apoptosis
TLDR
It can be concluded that the plurifunctionality of nucleoli regarding cell proliferation and apoptosis could open new vistas toward understanding dysregulation in malignant cells. Expand
Rescue of heterochromatin organization in Hutchinson-Gilford progeria by drug treatment
TLDR
The results show that the combined treatment with mevinolin and the histone deacetylase inhibitor trichostatin A dramatically lowers progerin levels, leading to rescue of heterochromatin organization and reorganization of transcripts in HGPS fibroblasts. Expand
Effect of GnRH analogues on apoptosis and release of interleukin-1beta and vascular endothelial growth factor in endometrial cell cultures from patients with endometriosis.
TLDR
GnRH agonists appear to have a direct effect in endometrial cells cultures, by enhancing the percentage of apoptotic cells and decreasing the release of pro-mitogenic cytokines such as IL-1beta and VEGF. Expand
From Silencing to Gene Expression Real-Time Analysis in Single Cells
TLDR
An inducible system to visualize gene expression at the levels of DNA, RNA and protein in living cells is developed, able to correlate changes in chromatin structure with the progression of transcriptional activation allowing for a real-time integrative view of gene expression. Expand
Molecular mechanism for silencing virally transduced genes involves histone deacetylation and chromatin condensation.

TLDR
The results suggest that deacetylation of histone H4 lysines and coincident chromatin condensation are critically involved in the silencing of virally transduced genes. Expand
