The combination of next generation sequencing (NGS) and automated liquid handling platforms has led to a revolution in single-cell genomic studies. However, many molecules that are critical to understanding the functional roles of cells in a complex tissue or organs, are not directly encoded in the genome, and therefore cannot be profiled with NGS. Lipids, for example, play a critical role in many metabolic processes but cannot be detected by sequencing. Recent developments in quantitative…

Applications of Raman spectroscopy in the development of cell therapies: state of the art and future perspectives.
TLDR
An overview of the current state of emerging cell therapies and the related Raman spectroscopic state of the art analysis of human cells is provided, and a number of salient challenges as cell therapy products are translated from the laboratory to the clinic are outlined and proposed.
Yellow-Emitting Carbon Dots for Selective Fluorescence Imaging of Lipid Droplets in Living Cells.
TLDR
The carbon dots showed distinct preference to be present in the hydrophobic environment which was evident from their efficient transfer from aqueous phase to organic phase and this inherent lipophilic character of these carbon dots has been successfully utilized for the selective imaging of lipid droplets inside the living cells.
Analysis of Membrane Lipids
TLDR
This chapter provides a background of lipids that are present in biological systems and concludes that each type of phosphoglycerides or sphingolipids is enriched with a host of molecular species imparting additional dynamism to lipid composition.
Assessing fatty acid-induced lipotoxicity and its therapeutic potential in glioblastoma using stimulated Raman microscopy
TLDR
The results suggest that EPA treatment combined with depletion of LDs significantly reduces the survival rate of glioma cells by more than 50%, indicating the therapeutic potential of this approach.
Label-free Raman mapping of saturated and unsaturated fatty acid uptake, storage, and return toward baseline levels in macrophages.
TLDR
This work uses Raman imaging and analysis to evaluate the effects of different fatty acids following their uptake in macrophages and finds that the label-free nature of the methods means that they can evaluate the fatty acid dynamics without modifying endogenous cellular behavior and metabolism.
Toward rapid infectious disease diagnosis with advances in surface-enhanced Raman spectroscopy.
TLDR
Recent, complementary advances in SERS substrates, machine learning, and microfluidics and bioprinting can be readily translated from laboratory bench to patient bedside, accelerating point-of-care diagnosis, personalized medicine, and precision health.

TLDR
This work presents a meta-analysis of lung function research at the cellular and Integrative Physiology level that shows clear patterns of decline in patients with high LDL cholesterol and high triglyceride levels over a long period of time.
Observation of the changes in the chemical composition of lipid droplets using Raman microscopy.
TLDR
The dynamics of lipid droplet formation induced by introducing cis- and/or trans-fatty acids into cells is reported, showing that exogenous fatty acids initially enter original endogenous droplets, then induce additional droplets containing endogenous lipids, and finally form their droplets.
