Protonation equilibria of the neurotransmitter serotonin and its precursor 5-hydroxytryptophan (5-HTP) are characterized at the macroscopic and microscopic levels. 1H-NMR-pH and UV-pH titrations were carried out to determine the macroconstants. Although the non‐charged microspecies of 5‐HTP is 151 times more lipophilic than its zwitterionic protonation isomer, the overwhelming dominance of the zwingerionic form ensures that its contribution to the overall lipophilicity exceeds 1320 times that of the non-charged one.
