}Gamete membrane interaction is critical to initiate the development of a new organism. The signaling pathways governing this event, however, are poorly understood. In this report, we provide the first evidence that protein kinase C activity in mouse eggs plays a crucial role in the regulation of this process. Stimulating PKC activity in mouse eggs by phorbol 12‐myristate 13‐acetate (PMA) drastically inhibited the egg's membrane ability to bind and fuse with sperm. Surprisingly, this significant…

The Dynamics of PKC-Induced Phosphorylation Triggered by Ca2+ Oscillations in Mouse Eggs
- BiologyJournal of cellular physiology
- 2013
Monitoring the dynamics of PKC‐induced phosphorylation in mouse eggs suggests that Ca2+‐induced DAG formation on intracellular membranes may stimulate PKC activity oscillations at fertilization, and shows that signal integration only occurs at a non‐physiological, highCa2+ oscillation frequency.
Paternal Exposure to Cyclophosphamide Affects the Progression of Sperm Chromatin Decondensation and Activates a DNA Damage Response in the Prepronuclear Rat Zygote1
- BiologyBiology of reproduction
- 2010
Exposure of male rats to chronic low doses of cyclophosphamide accelerates spermatozoal decondensation and leads to the activation of gammaH2AX recognition of DNA damage in the male genome of the prepronuclear zygote.
RNAi High Throughput Screenings Facilitate the Identification of Proteins Necessary for TCDD-induced CYP1A1 Enzymatic Activity and Aid in the Discovery of a Novel Role for Sin3A in AHR-Mediated Gene Expression

An essential role for Sin3A is established in the AHR-mediated induction of gene expression and dexamethasone-mediated regulation of CYP2S1 is explored, which demonstrated that these effects are mediated through AHR.

The aryl hydrocarbon receptor (AHR) has a plethora of physiological roles, and upon dysregulation, carcinogenesis can occur, so methods applied to Cyp1a1 could be modified for identifying proteins that regulate other inducible genes.

It is suggested that the cytoskeletal cortex is not a mere physical barrier that blocks CGE, but rather a dynamic network that can be maneuvered towards allowing CGE by activated actin-associated proteins and/or by activated PKC.

Protein kinase C activation is an early event of egg activation and both Ca2+-dependent and Ca2-independent pathways contribute to increased PKC activity at fertilization, and PKCdelta is one of the isoforms participating in this signalling process.
Mechanistic studies of the plasma membrane block to polyspermy in mouse eggs
- BiologyMolecular reproduction and development

Data support the following conclusions: an alteration in the ability of the egg plasma membrane to allow sperm adherence accounts for the block to polyspermy, since artificial egg activation does not result in a block response; and the contents of the eggs' cortical granules do not play a role in the establishment of the plasmalemma block response.

Involvement of Calcium Signaling and the Actin Cytoskeleton in the Membrane Block to Polyspermy in Mouse Eggs1

A postfertilization decrease in egg membrane receptivity to sperm requires functions of the egg actin cytoskeleton that are disrupted by cytochalasin D, and egg activation-associated increased intracellular calcium levels are necessary but not sufficient to affect postfERTilization membrane dynamics that contribute to a membrane block to polyspermy.
