The production of prostacyclin (PGI2) and thromboxane (TXA2) by irradiated rat lung and the concentration of these prostanoids in the bronchoalveolar lavage (BAL) fluid were measured in rats sacrificed at 2 or 6 months after a single dose of 10-30 Gy of cobalt-60 gamma rays to the right hemithorax. Lung PGI2 and TXA2 production increased linearly with increasing radiation dose at rates of 3.1% and 4.3% per Gy, respectively, at 2 months and 21.7% and 6.1% per Gy, respectively, at 6 months. Functional responses of the pulmonary endothelium to thoracic irradiation in rats: differential modification by D-penicillamine.
TLDR
Data indicate that the four "markers" of radiation-induced pulmonary endothelial dysfunction do not respond identically to penicillamine dose-modification, and TXA2 production exhibits the most significant and widespread peniillamine sparing. Expand
Annexin I concentration and prostacyclin production in rat lung and alveolar macrophages following irradiation.
TLDR
Data demonstrate that reduced levels of annexin I, as well as increased protein concentration, LDH activity and macrophage numbers in irradiated rat lung are reflected in BAL fluid, which reflects lung status, and may serve as a minimally invasive index of radiation pneumonitis in this model. Expand
Captopril reduces collagen and mast cell accumulation in irradiated rat lung.
TLDR
These data are the first to demonstrate that the ACE inhibitor captopril might provide a novel intervention in the pathogenesis of radiation fibrosis. Expand
Annexin I concentration, phospholipase activity and thromboxane synthesis in irradiated rat lung.
TLDR
Phospholipase activity is decreased in the irradiated lung, despite a decline in the concentration of annexin I, its putative inhibitor, which is not consistent with the hypothesis that increased synthesis of thromboxane A2 in irradiated rat lung is a direct result of elevated phospholipases activity. Expand
Radiation-induced endothelial dysfunction and fibrosis in rat lung: modification by the angiotensin converting enzyme inhibitor CL242817.
TLDR
The data suggest that the ability of ACE inhibitors to ameliorate radiation-induced pulmonary endothelial dysfunction is not unique to captopril, rather it is a therapeutic action shared by other members of this class of compounds. Expand
Serum copper concentration as an index of lung injury in rats exposed to hemithorax irradiation.
TLDR
Serum copper concentration is an accurate and minimally invasive index of lung injury in rats irradiated to the hemithorax and can predict lung hydroxyproline (collagen) content in individual irradiated rats. Expand
The effect of Captopril on benign and malignant reactions in irradiated rat skin.
TLDR
Captopril, known to ameliorate acute lung damage in irradiated rats, also reduces chronic benign and malignant skin reactions in the radiation treatment field. Expand
Structure-function analysis of angiotensin-converting enzyme inhibitors as modifiers of radiation-induced pulmonary endothelial dysfunction in rats.
TLDR
The ACE inhibitor CGS13945 modifies radiation-induced pulmonary endothelial dysfunction in rats, indicating that the presence of a thiol group is not essential for therapeutic efficacy in this class of compounds and suggesting that there are at least two components to their mechanism of therapeutic action in this model. Expand
Thromboxane increase in irradiated animals is caused by stimulation of cyclooxygenase activity.
TLDR
The results indicate that the increase in thromboxane synthesis is not associated with the activation of phospholipase but is caused by stimulation of cyclooxygenase activity. Expand
On radiation damage to normal tissues and its treatment. II. Anti-inflammatory drugs.
TLDR
Observations after irradiation and results of empirical treatment of radiation injuries with anti-inflammatory drugs are highly suggestive of a contribution of humoral factors to the adverse responses of normal tissues and organs to radiation. Expand

Radiation injury in rat lung. IV. Modification by D-penicillamine.
TLDR
The antifibrotic agent D-penicillamine delays the onset of radiation-induced enzyme dysfunction in the pulmonary endothelium and is most effective in ameliorating endothelial damage during the first 2 months after irradiation, preceding the development of interstitial fibrosis. Expand

TLDR
The data suggest that significant alterations in prostaglandin and thromboxane concentrations in parenchymal lung tissues occur following irradiation, in a dose-dependent manner, and that altered responsiveness to H-1 receptor stimulation and divalent cation transport also occur. Expand
6-keto-prostaglandin F1α/thromboxane B2 ratio in vascular and lung tissue
Abstract In vitro production of 6-keto-PGF 1α and thromboxane B 2 (TxB 2 ) was determined by radioimmunoassay. The 6-keto-PGF 1α /TxB 2 ratio varied between 5 and 41 in vascular tissue and betweenExpand
Plasminogen activator activity in lung and alveolar macrophages of rats exposed to graded single doses of gamma rays to the right hemithorax.
TLDR
Data quantitate the dose response and time course of radiation-induced fibrinolytic defects in rat lung and suggest that information obtained from a minimally invasive procedure such as bronchoalveolar lavage may serve as an index of the degree of pulmonary fibrin clot lysis dysfunction after irradiation. TLDR
Monocrotaline-induced pulmonary injury is accompanied, and in some cases preceded, by structural and functional abnormalities in the pulmonary endothelium, as visualized by 99mTc lung scans. Expand

The relationship between endothelial dysfunction and collagen accumulation in irradiated rat lung.
TLDR
In rat lung, arterial perfusion, endothelial dysfunction and interstitial fibrosis, the dose-effect curves for these three responses of the lung in situ change less than 5% per Gy over the dose range of 10-30 Gy, a smaller variation than would be predicted from endothelial cell survival data based on clonogenic assays in vitro or in vivo. The ultrastructure of radiation injury in rat lung: modification by D-penicillamine.
TLDR
Penicillamine treatment produces a generalized inhibition of pathologic events such as collagen accumulation and arterial wall thickening, and acceleration of restorative processes such as revascularization andmore » collagen degradation. Expand
Monocrotaline-Induced Pulmonary Fibrosis in Rats: Amelioration by Captopril and Penicillamine
TLDR
Hydxyproline data demonstrate that the ACE inhibitor Captropril exhibits antifibrotic activity in monocrotaline-treated rat lung, and penicillamine, known to inhibit radiation-induced lung injury, thus is shown to be effective in a second model of pulmonary fibrosis. Expand
