Cell culture is a significant tool for studying endocrinology and diseases in shrimp.A primary male germ cell culture method in Macrobrachium nipponense is introduced in this paper. We found that spermatogonia and spermatocytes had active division capability.According to the data about cell adherence and cell growth,we determined the optimal pH and osmolarity for in vitro culture of these cells. The optimal pH was 7.2-7.4 and the proper osmolarity was 400-500 mmol/L.
