Brain-derived neurotrophic factor (BDNF) has been implicated in several forms of long-term potentiation (LTP) at different hippocampal synapses. Using two-photon imaging of FM 1-43, a fluorescent marker of synaptic vesicle cycling, we find that BDNF is selectively required for those forms of LTP at Schaffer collateral synapses that recruit a presynaptic component of expression. BDNF-dependent forms of LTP also require activation of L-type voltage-gated calcium channels. One form of LTP with… Expand
Genetic Dissection of Presynaptic and Postsynaptic BDNF-TrkB Signaling in Synaptic Efficacy of CA3-CA1 Synapses
TLDR
A viral-mediated approach to delete BDNF or TrkB specifically in CA1 and CA3 regions of the Schaffer collateral pathway reveals insight into BDNF release and TrkB activation sites in hippocampal LTP. Expand
Essential Role of Presynaptic NMDA Receptors in Activity-Dependent BDNF Secretion and Corticostriatal LTP
TLDR
It is reported that axonal NMDARs play an essential role in LTP induction at mouse corticostriatal synapses by triggering activity-induced presynaptic secretion of brain-derived neurotrophic factor (BDNF). Expand
Slow Presynaptic and Fast Postsynaptic Components of Compound Long-Term Potentiation
TLDR
The first direct evidence that presynaptic and postsynaptic components of LTP are temporally and mechanistically distinct is found. Expand
Presynaptic and postsynaptic NMDA receptors mediate distinct effects of BDNF on synaptic transmission
In addition to its effects on neuronal survival and differentiation, brain-derived neurotrophic factor (BDNF) plays an important role in modulating synaptic transmission and plasticity in many brainExpand
Early Presynaptic and Late Postsynaptic Components Contribute Independently to Brain-Derived Neurotrophic Factor-Induced Synaptic Plasticity
TLDR
These results are the first to demonstrate that presynaptic and postsynaptic components of BDNF-enhanced synaptic activity are independent and temporally distinct. Expand
Postsynaptic protein synthesis is required for presynaptic enhancement in persistent forms of long-term potentiation
TLDR
It is concluded that postsynaptic translation is required for the presynaptic component of LTP2 and LTP3 expression, which considerably strengthen the mechanistic separation ofLTP1, 2, and 3 and provide evidence for an expanded repertoire of communication between synaptic elements. Expand
BDNF function in adult synaptic plasticity: The synaptic consolidation hypothesis
TLDR
A model is proposed in which BDNF signaling at glutamate synapses drives the translation of newly transported and locally stored mRNA in dendrites, which defines a critical window for synaptic consolidation. Expand
Regulation of hippocampal synaptic plasticity by BDNF
TLDR
This review summarizes the evidence pointing to a role for BDNF in generating functional and structural changes at synapses required for both early- and late phases of LTP in the hippocampus and discusses the effects of BDNF on the synaptic proteome. Expand
Presynaptic and postsynaptic NMDA receptors mediate distinct effects of brain-derived neurotrophic factor on synaptic transmission.
TLDR
The results indicate that BDNF acutely modulates presynaptic release and postsynaptic responsiveness through simultaneous effects on pre- and post Synaptic NMDA receptors. Expand
BDNF and Hippocampal Synaptic Plasticity.
TLDR
The evidence pointing to a fundamental role of this neurotrophin in LTP is reviewed, especially within the hippocampus, and the synergy between TrkB and glucocorticoid receptor signaling to determine cellular responses to stress is discussed. Expand

Postsynaptic Induction of BDNF-Mediated Long-Term Potentiation
TLDR
The results suggest thatBDNF-mediated LTP is induced postsynaptically, and the finding that dendritic spines are the exclusive synaptic sites for rapid BDNF-evoked Ca2+ signaling supports this conclusion. Expand
Presynaptic Modulation of Synaptic Transmission and Plasticity by Brain-Derived Neurotrophic Factor in the Developing Hippocampus
TLDR
Results suggest that brain-derived neurotrophic factor acts presynaptically, and the preferential potentiation of highly active synapses by BDNF may have implications in the Hebbian mechanism of synaptic plasticity. Expand
Visualization of changes in presynaptic function during long-term synaptic plasticity
TLDR
Enhanced presynaptic function during long-term potentiation (LTP) induced either chemically (with tetraethylammonium), or by high-frequency (200-Hz) electrical stimulation implied that a long-lasting increase in the efficacy of synaptic transmission is likely to depend, at least in part, on enhanced transmitter release from the Presynaptic neuron. TLDR
It is reported that BDNF, within 5 min of exposure, elicits a dose-dependent increase in phosphorylation of the N-methyl-D-aspartate receptor subunit 1, suggesting a potential mechanism for trophin-induced potentiation of synaptic transmission. Expand
Regulation of synaptic responses to high-frequency stimulation and LTP by neurotrophins in the hippocampus
TLDR
The results suggest that BDNF may regulate LTP in developing and adult hippocampus by enhancing synaptic responses to tetanic stimulation, and a TrkB–IgG fusion protein, which scavenges endogenous BDNF11, reduced the synaptic responses in adult hippocampus as well as the magnitude of LTP. Expand
The Role of Brain-Derived Neurotrophic Factor Receptors in the Mature Hippocampus: Modulation of Long-Term Potentiation through a Presynaptic Mechanism involving TrkB
TLDR
BDNF appears to act through TrkB presynaptic, but not postsynaptically, to modulate LTP, indicating that BDNF does not modulate plasticity through postsynaptic TrkB. Expand
A role for BDNF in the late-phase of hippocampal long-term potentiation
TLDR
Slices from BDNF knock-out animals or slices treated with TrkB antibodies failed to exhibit L-LTP, indicating that TrkB ligands participate in extending synaptic enhancement from a short-lasting to a long-lasting form. Expand
Postsynaptic Target Specificity of Neurotrophin-Induced Presynaptic Potentiation
TLDR
The role of the target cell in neurotrophin-induced modifications of glutamatergic synaptic transmission was examined in cultured hippocampal neurons and the selective effect at E-->E suggests that retrograde signaling by the postsynaptic target cell endows a localized presynaptic action of neurotrophins. Expand
Synaptic reliability correlates with reduced susceptibility to synaptic potentiation by brain-derived neurotrophic factor.
TLDR
It is found that the degree of synaptic potentiation by BDNF depends on the initial reliability and strength of the synapse, and the dependence on initial synaptic strength was found for divergent outputs of the same presynaptic neuron, suggesting that synaptic terminals with different degrees of responsiveness to BDNF can coexist within in the same neuron. Expand
Some Forms of cAMP-Mediated Long-Lasting Potentiation Are Associated with Release of BDNF and Nuclear Translocation of Phospho-MAP Kinase
TLDR
Surprisingly, in these BDNF-dependent forms of potentiation, which are also MAPK dependent, TrkB activation is not critical for the activation of MAPK but instead appears to modulate the subcellular distribution and nuclear translocation of the activated MAPK. Expand
