The “RNA world” hypothesis is consistent with RNA’s being a prebiotic product, or the biotic invention of a precursor genetic system. A prebiotically plausible synthesis of ribonucleotides would help to differentiate between these two possibilities by providing strong support for the first, and, as such, is a major goal. The efficient conversion of d-ribose aminooxazoline 1 to ad-cytidine 2, by treatment with cyanoacetylene was demonstrated over 30 years ago (Scheme 1), 3] but has not gained
A Prebiotic Synthesis of Canonical Pyrimidine and Purine Ribonucleotides
TLDR
It is reported that the glycosidic bond in canonical pyrimidine and purine ribonucleotides can be formed by direct coupling of cyclic carbohydrate phosphates with free nucleobases, all reported to be available by experimentally supported pathways that might have operated on early Earth. Expand
Selective aqueous acetylation controls the photoanomerization of α-cytidine-5'-phosphate.
TLDR
It is reported that prebiotic acetylation of the non-natural, but prebiotically plausible, ribonucleotide α-cytidine-5'-phosphate, selectively protects the vicinal diol moiety. Expand
Studies towards the prebiotic synthesis and phosphorylation of ribonucleotides
TLDR
The discovery of a generational node in the network of prebiotic chemistry that links the syntheses of amino acids with nucleotides 5'-phosphates suggests that these different groups of metabolites need not have arisen from separate chemistries, helping to get a better understanding of the processes governing the chemical evolution of life. Expand
Chemoselective Multicomponent One-Pot Assembly of Purine Precursors in Water
TLDR
A high-yielding, aqueous, one-pot, multicomponent reaction that tethers masked-sugar moieties to prebiotically plausible purine precursors is developed, suggesting the first prebioticically plausible mechanism for regiospecific N9 purination. Expand
Photostability of oxazoline RNA-precursors in UV-rich prebiotic environments.
TLDR
UV-irradiation experiments and quantum-chemical calculations demonstrate that these compounds are remarkably photostable and could accumulate over long periods of time in UV-rich prebiotic environments to undergo stereoisomeric purification. Expand
Phosphoribosyl Pyrophosphate: A Molecular Vestige of the Origin of Life on Minerals.
TLDR
The formation under prebiotic conditions of phosphoribosyl pyrophosphate as a molecular precursor in the one-pot synthesis of a canonical nucleotide, namely adenosine monophosphate from its building blocks, on a fumed silica surface is reported. Expand
Mechanism and diastereoselectivity of the prebiotic synthesis of deoxyribonucleotide precursors C5-thiazoline: A DFT study
Abstract The prebiotic synthesis of deoxyribonucleotide precursor C 5 -thiazoline has been studied by performing density functional theory calculations. Two possible reaction pathways are exploredExpand
A Novel General Methodology for Ribozyme-Mimetic Synthesis of Methyl Esters of Various Natural Amino Acids, Simulating the Prebiotic Biomolecule Creation
TLDR
A new originally developed procedure for biomimetic synthesis of methyl esters of the natural amino acids was realized at conditions, imitating the action of the primitive ribozymes, and this strategic methodology presumes their crucial role in the prebiotic syntheses of biomolecules during the primordial RNA World. Expand

Understanding how life on Earth might have started is the major goal of prebiotic chemistry. The universal nature of extant life, with integrated informational, catalytic and compartment-formingExpand

On the Prebiotic Synthesis of Ribonucleotides: Photoanomerisation of Cytosine Nucleosides and Nucleotides Revisited
TLDR
The present study determined whether this low yield was the result of a low conversion or competing reaction pathways, and if an efficient means of anomerising this nucleoside or a derivative thereof were to be found, then the synthesis of one of the key β‐D‐nucleosides required to make RNA would be realised. Two-step potentially prebiotic synthesis of alpha-d-cytidine-5'-phosphate from d-glyceraldehyde-3-phosphate.
TLDR
A process in which ribose-5-phosphate is bypassed, and a cytidine 5‘-ribonucleotide is efficiently assembled under mild conditions in two steps from glyceraldehyde-3-ph phosphate, 2-aminooxazole, cyanoacetylene, and water is demonstrated. Expand
Reaction of Cytidine Nucleotides with Cyanoacetylene: Support for the Intermediacy of Nucleoside‐2′,3′‐cyclic Phosphates in the Prebiotic Synthesis of RNA
TLDR
It is found that cyanoacetylene, the reagent used for the second stage of nucleobase assembly in the synthesis of C >p, also reverses the effect of the hydrolysis by driving efficient cyclisation of C2′p and C3′p back to C>p. Expand

TLDR
The evidence that is currently available does not support the availability of ribose on the prebiotic earth, except perhaps for brief periods of time, in low concentration as part of a complex mixture, and under conditions unsuitable for nucleoside synthesis. Expand
