Excitotoxicity is a major cause of retinal ganglion cell (RGC) death during ischemic diseases such as vessel occlusion and diabetic retinopathy. However, the underlying mechanisms are not well understood. Statins, inhibitors of the HMG‐CoA reductase, have neuroprotective effects in addition to their original role in lowering cholesterol. We hypothesize that pitavastatin, a recently introduced potent statin, is protective against N‐methyl‐d‐aspartic acid (NMDA)‐induced RGC death. Pitavastatin… Expand
ERK1 plays a critical protective role against N‐methyl‐D‐aspartate‐induced retinal injury
TLDR
A previously unknown role for retinal glial cells in NMDA‐induced retinal injury that involves the activation of ERK1/2 is described, and it is suggested that, during NMDA injury, Müller cells are activated and play a protective role againstNMDA‐ induced retinal cell death. Expand
Pitavastatin Attenuates Leukocyte-Endothelial Interactions Induced by Ischemia-Reperfusion Injury in the Rat Retina
TLDR
Pitavastatin effectively attenuated ischemia-induced leukocyte-endothelial interactions in the rat retina and significantly reduced ICAM-1 mRNA expression in the retina during ischemIA-reperfusion injury. Expand
Metformin Protects against NMDA-Induced Retinal Injury through the MEK/ERK Signaling Pathway in Rats
TLDR
It is suggested that metformin protects against NMDA-induced retinal neurotoxicity through activation of the AMPK and MEK/extracellular signal-regulated kinase (ERK) signaling pathways. Expand
Preventive effects of rapamycin on inflammation and capillary degeneration in a rat model of NMDA-induced retinal injury.
TLDR
The results suggest that rapamycin has preventive effects on inflammation and capillary degeneration during retinal injury. Expand
Involvement of matrix metalloproteinases in capillary degeneration following NMDA‐induced neurotoxicity in the neonatal rat retina
TLDR
Results indicate that MMP‐9 plays an important role in neurovascular degeneration in the injured retina and inhibits MMP inhibitors protect neuronal and vascular cells in NMDA‐treated retinas. Expand
Protective effects of PF‐4708671 against N‐methyl‐d‐aspartic acid‐induced retinal damage in rats
TLDR
Results suggest that PF‐4708671 and rapamycin likely protect against NMDA‐induced retinal damage via distinct pathways, and suggest that the neuroprotective effect of PF‐ 4708671 is unlikely to be associated with inhibition of the S6 kinase, even though it is reported to be a S 6 kinase inhibitor. Expand
Rapamycin prevents N‐methyl‐D‐aspartate‐induced retinal damage through an ERK‐dependent mechanism in rats
TLDR
Results suggest that rapamycin has a neuroprotective effect against NMDA‐induced retinal neurotoxicity and that this effect could be patially mediated by activation of the ERK pathway in retinal Müller cells. Expand
Inhibition of vascular adhesion protein‐1 suppresses endotoxin‐induced uveitis
TLDR
VAP‐1 inhibition in EIU animals significantly suppressed leukocyte recruitment to the anterior chamber, vitreous, and retina, as well as retinal endothelial P‐selectin expression, suggesting an important role for VAP‐ 1 in the recruitment of leukocytes to the immune‐privileged ocular tissues during acute inflammation. Expand
Neuroprotective role of sphingolipid rheostat in excitotoxic retinal ganglion cell death.
TLDR
The findings in the present study provide the first evidence demonstrating the involvement of sphingolipid rheostat in the neuroprotection against excitotoxic RGC death and might serve as a potential therapy for retinal degenerative disease. Expand
The neuroprotective effect of hesperidin in NMDA-induced retinal injury acts by suppressing oxidative stress and excessive calpain activation
TLDR
It is found that hesperidin suppressed a number of cytotoxic factors associated with NMDA-induced cell death signaling, such as oxidative stress, over-activation of calpain, and inflammation, thereby protecting the RGCs in mice. Expand

Statin inhibits leukocyte-endothelial interaction and prevents neuronal death induced by ischemia-reperfusion injury in the rat retina.
TLDR
Investigating the effects of statin on leukocyte accumulation during ischemia-reperfusion injury and on subsequent retinal damage found that statin may exert neuroprotective effects by inhibiting leukocytes-endothelial interaction through the release of nitric oxide from the endothelium. Expand
Activation of matrix metalloproteinase-9 via neuronal nitric oxide synthase contributes to NMDA-induced retinal ganglion cell death.
TLDR
Investigating the mechanism of excitotoxic retinal ganglion cell (RGC) death in vivo and its relationship to MMP activation suggests that an extracellular proteolytic pathway in the retina contributes to RGC death via NO-activated MMP-9. Expand
Neuroprotective effects of atorvastatin against glutamate‐induced excitotoxicity in primary cortical neurones
TLDR
Atorvastatin exerts specific anti‐excitotoxic effects independent of 3‐hydroxy‐3‐methylglutaryl‐CoA reductase inhibition, which has potential therapeutic implications. Expand
p53 regulates apoptotic retinal ganglion cell death induced by N-methyl-D-aspartate.
TLDR
Investigation of the role of p53 in RGC death activated by overstimulation of the N-methyl-D-aspartate (NMDA) receptor, an ionotropic glutamate dependent calcium channel normally involved in glutamate neurotransduction, results suggest that p53 is not required for developmental programmed cell death of RGCs. Expand
Diltiazem-induced Neuroprotection in Glutamate Excitotoxicity and Ischemic Insult of Retinal Neurons
TLDR
The results are consistent with previous studies suggesting that Ca2+ channel activation contributes to retinal cell death following either glutamate excitotoxicity or retinal ischemia and under both conditions, the L-type Ca 2+ channel blocker, diltiazem, can limit cell death. Expand
NMDA‐induced retinal injury is mediated by an endoplasmic reticulum stress‐related protein, CHOP/GADD153
TLDR
CHOP is induced in mouse retina by NMDA treatment, and CHOP–/– mice are more resistant toNMDA‐induced retinal damage, suggesting that CHOP plays an important role in NMDA‐ induced retinal cell death. Expand

TLDR
It is demonstrated that pharmacological modulation of apoptotic cell death may affect the final tissue responses in vivo and suggested that the NMDA receptors may have an important role in ischemia-reperfusion insult as well as in mediating ischemic-induced apoptosis of retinal neurons. Expand
