SYNOPSIS MnII complexes formed with cis- and trans-DO2A (DO2A=1,4,7,10-tetraazacyclododecane-1,4 (or 1,7) -diacetic acid) chelators were investigated by pH-potentiometry, 1H relaxometry, UV-vis spectrophotometry and cyclic voltammetry. The physico-chemical characteristics of MnII complexes of these structure isomers do not differ dramatically, however the cis-DO2A platform has better potential for further development. Manganese (MnII) is a promising alternative to gadolinium (GdIII) as a… Expand

Novel CDTA-based, Bifunctional Chelators for Stable and Inert MnII Complexation: Synthesis and Physicochemical Characterization.
TLDR
Given the exquisite compromise between thermodynamic stability, kinetic inertness, and relaxivity achieved by [Mn(4-HET-CDTA], appropriately designed CuAAC-conjugates of 6a/6b are promising precursors for the preparation of targeted, bioresponsive, or high relaxivity manganese-based PET/MR tracers (52g/55 MnII) and MR contrast agents (MnII). A detailed relaxometric characterisation of the complexes using nuclear magnetic dispersion relaxation (NMRD) and NMR studies indicated that the increase in molecular weight associated with the presence of benzyl groups results in a remarkable increase of proton relaxivities r1p, which take values of 3.8, 3.5 and 2.5 mM-1 s-1. TLDR
Three 1,4,7,10-tetraazacyclodododecane-based ligands disubstituted in 1, 4-positions with phosphonic acid, phosphonate monoethyl-ester, and H-phosphinic acid pendant arms were synthesized and their coordination to selected metal ions was investigated. Expand
Effect of the Nature of Donor Atoms on the Thermodynamic, Kinetic and Relaxation Properties of Mn(II) Complexes Formed With Some Trisubstituted 12-Membered Macrocyclic Ligands
TLDR
Equilibrium, kinetic parameters, and relaxivity of the Mn(II) complexes formed with 12-membered macrocyclic ligands were studied, finding that the phosphonate pedant should not be considered as a suitable building block for further ligand design while the tertiary amide moiety will likely have some implications in this respect in the future. Mn(II) compounds as an alternative to Gd-based MRI probes.
TLDR
This review aims to critically discuss the chemical and magnetic properties of Mn(II) compounds relevant as MRI probes, both small complexes and nanosystems containing a large number of metal centers, the possible approaches for optimizing their efficiency by understanding the role of various molecular parameters that control the relaxation processes, and the most important issues related to stability and kinetic inertness.
